  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Chemoprophylaxis, diagnosis, treatments, and discharge management of COVID-19 : An evidence-based clinical practice guideline (updated version)

    • The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of a rapidly spreading illness, coronavirus disease 2019 (COVID-19), affecting more than seventeen million people around the world. Diagnosis and treatment guidelines for clinicians caring for patients are needed. In the early stage, we have issued "A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus (2019-nCoV) infected pneumonia (standard version)"; now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline. We formed a working group of clinical experts and methodologists. The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas: chemoprophylaxis, diagnosis, treatments, and discharge management. We searched the literature for direct evidence on the management of COVID-19, and assessed its certainty generated recommendations using the Grading of Recommendations, Assessment, Development and Evaluation (GRADE) approach. Recommendations were either strong or weak, or in the form of ungraded consensus-based statement. Finally, we issued 34 statements. Among them, 6 were strong recommendations for, 14 were weak recommendations for, 3 were weak recommendations against and 11 were ungraded consensus-based statement. They covered topics of chemoprophylaxis (including agents and Traditional Chinese Medicine (TCM) agents), diagnosis (including clinical manifestations, reverse transcription-polymerase chain reaction (RT-PCR), respiratory tract specimens, IgM and IgG antibody tests, chest computed tomography, chest x-ray, and CT features of asymptomatic infections), treatments (including lopinavir-ritonavir, umifenovir, favipiravir, interferon, remdesivir, combination of antiviral drugs, hydroxychloroquine/chloroquine, interleukin-6 inhibitors, interleukin-1 inhibitors, glucocorticoid, qingfei paidu decoction, lianhua qingwen granules/capsules, convalescent plasma, lung transplantation, invasive or noninvasive ventilation, and extracorporeal membrane oxygenation (ECMO)), and discharge management (including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge). We also created two figures of these recommendations for the implementation purpose. We hope these recommendations can help support healthcare workers caring for COVID-19 patients.

  • Inhibition of copper transporter 1 prevents α-synuclein pathology and alleviates nigrostriatal degeneration in AAV-based mouse model of Parkinson's disease

    • The formation of α-synuclein aggregates is a major pathological hallmark of Parkinson's disease. Copper promotes α-synuclein aggregation and toxicity in vitro. The level of copper and copper transporter 1, which is the only known high-affinity copper importer in the brain, decreases in the substantia nigra of Parkinson's disease patients. However, the relationship between copper, copper transporter 1 and α-synuclein pathology remains elusive. Here, we aim to decipher the molecular mechanisms of copper and copper transporter 1 underlying Parkinson's disease pathology. We employed yeast and mammalian cell models expressing human α-synuclein, where exogenous copper accelerated intracellular α-synuclein inclusions and silencing copper transporter 1 reduced α-synuclein aggregates in vitro, suggesting that copper transporter 1 might inhibit α-synuclein pathology. To study our hypothesis in vivo, we generated a new transgenic mouse model with copper transporter 1 conditional knocked-out specifically in dopaminergic neuron. Meanwhile, we unilaterally injected adeno-associated viral human-α-synuclein into the substantia nigra of these mice. Importantly, we found that copper transporter 1 deficiency significantly reduced S129-phosphorylation of α-synuclein, prevented dopaminergic neuronal loss, and alleviated motor dysfunction caused by α-synuclein overexpression in vivo. Overall, our data indicated that inhibition of copper transporter 1 alleviated α-synuclein mediated pathologies and provided a novel therapeutic strategy for Parkinson's disease and other synucleinopathies.

  • Trans-ancestry genome-wide association meta-analysis of prostate cancer identifies new susceptibility loci and informs genetic risk prediction

    • Prostate cancer is a highly heritable disease with large disparities in incidence rates across ancestry populations. We conducted a multiancestry meta-analysis of prostate cancer genome-wide association studies (107,247 cases and 127,006 controls) and identified 86 new genetic risk variants independently associated with prostate cancer risk, bringing the total to 269 known risk variants. The top genetic risk score (GRS) decile was associated with odds ratios that ranged from 5.06 (95% confidence interval (CI), 4.84-5.29) for men of European ancestry to 3.74 (95% CI, 3.36-4.17) for men of African ancestry. Men of African ancestry were estimated to have a mean GRS that was 2.18-times higher (95% CI, 2.14-2.22), and men of East Asian ancestry 0.73-times lower (95% CI, 0.71-0.76), than men of European ancestry. These findings support the role of germline variation contributing to population differences in prostate cancer risk, with the GRS offering an approach for personalized risk prediction. A meta-analysis of genome-wide association studies across different populations highlights new risk loci and provides a genetic risk score that can stratify prostate cancer risk across ancestries.

    • We investigated the influence of chloride on the secondary passive film (SPF) on laser directed energy deposition Alloy 718 during electrochemical machining. The results show that SPF formed in chloride-containing electrolyte is more defective than that formed in chloride-free solution, due to the stepped-up cation ejection by chloride. Chloride accelerates the SPF failure via enhanced cation vacancy condensation, SPF dissolution and possibly via surface vacancy pairs' coalescence, restraining the formation of CrO3 and inducing a better surface quality than does the chloride-free electrolyte. Based on the Point Defect Model, a mechanism describing the influence of chloride on SPF was developed.

  • Fabrication and bonding of In bumps on Micro-LED with 8 μ m pixel pitch

    • Indium (In) is currently used to fabricate metal bumps on micro-light-emitting diode (Micro-LED) chips due to its excellent physical properties. However, as Micro-LED pixel size and pitch decrease, achieving high-quality In bumps on densely packed Micro-LED chips often presents more challenges. This paper describes the process of fabricating In bumps on micro-LEDs using thermal evaporation, highlighting an issue where In tends to grow laterally within the photoresist pattern, ultimately blocking the pattern and resulting in undersized and poorly dense In bumps on the Micro-LED chip. To address this issue, we conducted numerous experiments to study the height variation of In bumps within a range of photoresist aperture sizes (3 mu m -7 mu m) under two different resist thickness conditions (3.8 mu m and 4.8 mu m). The results showed that the resist thickness had a certain effect on the height of In bumps on the Micro-LED chip electrodes. Moreover, we found that, with the photoresist pattern size increasing under constant resist thickness conditions, the height and quality of the bumps significantly improved. Based on this finding, we rationalized the adjustment of the photoresist pattern size within a limited emission platform range to compensate for the height difference of In bumps caused by different resist thicknesses between the cathode and anode regions. Consequently, well-shaped and dense In bumps with a maximum height of up to 4.4 mu m were fabricated on 8 mu m pitch Micro-LED chips. Afterwards, we bonded the Micro-LED chip with indium bumps to the CMOS chip, and we found that we could successfully control the CMOS chip to drive the Micro-LED chip to display specific characters through the Flexible Printed Circuit (FPC). This work is of significant importance for the fabrication of In bumps on Micro-LED chips with pitches below 10 mu m and subsequent bonding processes.

  • The selection strategy of ammonium-group organic salts in vapor deposited perovskites: From dimension regulation to passivation

    • Dimension regulation and defect passivation are two key strategies for highly efficient and stable perovskite solar cell. Vapor deposition of perovskite is a toxic-solvent-free method for large-scale fabrication. However, without the assistance of solvent for crystal optimization, effective structural regulation and defect passivation become challenging. Here, detailed investigations on the structural evolution of perovskite thin film are carried out in sequential vapor deposition using mixed-vapor (R-NH3I/MAI). Correlation between electron donating ability of R-NH3I (BAI, PEAI, PMAI and ALI) molecule and the way of structural transition is established. It is found that RNH3I with stronger electron-donating ability promoted the phase transition from three-dimensional (3D) to twodimensional (2D) perovskite. Typically, the n value from 1 to 5 can be tuned by reaction time or component control using BAI with the strongest electron donating ability. R-NH3I with weak electron-donating ability suppresses the 3D to 2D transition, but enhances the defect passivation effect. The ALI with the weakest electron donating ability shows the best passivation effect, leading to the best device performance than that of the control 3D device, with PCE of 18.23% (0.045 cm2) and 15.48% (1 cm2) and the significantly improved stability. This study provides the evidence that the concept of Lewis acid-base reaction is applicable in vapor deposition, which provides us with the selection guide of R-NH3I molecules for structural design in vapor fabrication of perovskite thin film.

    • Colloidal quantum dots (QDs) are promising building blocks towards the development of cost-effective and high-efficiency photoelectrochemical (PEC) cells. Unfortunately, the frequent use of QDs possessing heavy metals (e.g. Cd and Pb) in state-of-the-art QD-based PEC technologies is a major obstacle regarding their future commercial perspective. In this work, we synthesized heavy metal-free quaternary CuZnInS3 (CZIS) with variable Cu : Zn ratios and fabricated corresponding QDs-PEC devices via a facile chemical bath deposition (CBD) technique. It is revealed that the tuned CZIS (1Zn) QDs (i.e. Cu : Zn ratio of 1 : 1) can result in optimized optical properties including enhanced quantum yield, suppressed nonradiative recombination and extended excitonic lifetime. Accordingly, as-fabricated CZIS (1Zn) QD-based photoanodes demonstrated increased charge transfer rate and decreased electron transport resistance for improved PEC performance. The results indicate that tuning the composition of heavy metal-free multinary QDs is one of the promising pathways to achieve eco-friendly and high-performance PEC systems for solar hydrogen production.

    • Colloidal semiconductor quantum dots (QDs) are promising building-blocks for the manufacture of cost-effective photoelectrochemical (PEC) cells towards efficient solar-to-hydrogen conversion. Nevertheless, the state-of-the-art QDs-based PEC systems still suffer from the frequent utilization of highly toxic elements in QDs (Cd and Pb), hindering their future practical applications and potential commercialization. Here, we report a PEC device fabricated using eco-friendly, near-infrared (NIR) ZnAgInSe (ZAISe) QDs and hybrid TiO2/graphene oxide (GO) film. Based on the synergistic effect of QD’s broad light absorption and excellent charge extraction/transport properties of TiO2/GO film, as-assembled QDs-photoanode exhibits an outstanding saturated photocurrent density of ∼6.7 mA/cm2 with good stability under standard 1 sun illumination. The introduction of functional GO can lead to the reduced charge transfer resistance, suppressed charge recombination, and enhanced electron transport within the QDs-TiO2 photoanodes. The results offer a facile and effective method to enhance the performance of environmentally friendly QDs-based PEC devices and shed light on the development of low-cost, “green” and high-efficiency solar-to-hydrogen conversion system.

    • The brain, with its diverse physiology and intricate cellular organization, is the most complex organ of the mammalian body. To expand our basic understanding of the neurobiology of the brain and its diseases, we performed a comprehensive molecular dissection of 10 major brain regions and multiple subregions using a variety of transcriptomics methods and antibody-based mapping. This analysis was carried out in the human, pig, and mouse brain to allow the identification of regional expression profiles, as well as to study similarities and differences in expression levels between the three species. The resulting data have been made available in an open-access Brain Atlas resource, part of the Human Protein Atlas, to allow exploration and comparison of the expression of individual protein-coding genes in various parts of the mammalian brain.
