Anti-infectious and anti-inflammatory effects of peptide fragments sequentially derived from the antimicrobial peptide centrocin 1 isolated from the green sea urchin, Strongylocentrotus droebachiensis

- Bacterial resistance against antibiotic treatment has become a major threat to public health. Antimicrobial peptides (AMPs) have emerged as promising alternative agents for treatment of infectious diseases. This study characterizes novel synthetic peptides sequentially derived from the AMP centrocin 1, isolated from the green sea urchin, for their applicability as anti-infective agents. The microbicidal effect of centrocin 1 heavy chain (CEN1 HC-Br), its debrominated analogue (CEN1 HC), the C-terminal truncated variants of both peptides, i.e. CEN1 HC-Br (1–20) and CEN1 HC (1–20), as well as the cysteine to serine substituted equivalent CEN1 HC (Ser) was evaluated using minimal microbicidal concentration assay. The anti-inflammatory properties were assessed by measuring the inhibition of secretion of pro-inflammatory cytokines. All the peptides tested exhibited marked microbicidal and anti-inflammatory properties. No difference in efficacy was seen comparing CEN1 HC-Br and CEN1 HC, while the brominated variant had higher cytotoxicity. C-terminal truncation of both peptides reduced salt-tolerability of the microbicidal effect as well as anti-inflammatory actions. Also, serine substitution of cysteine residue decreased the microbicidal effect. Thus, from the peptide variants tested, CEN1 HC showed the best efficacy and safety profile. Further, CEN1 HC significantly reduced bacterial counts in two different animal models of infected wounds, while Staphylococcus aureus and methicillin-resistant S. aureus (MRSA) failed to develop resistance against this peptide under continued selection pressure. In summary, CEN1 HC appears a promising new antimicrobial agent, and clinical studies are warranted to evaluate the applicability of this AMP for local treatment of infections in man.

- In this article we argue that school inspection is an important and potentially influential way of governing education that deserves additional scholarly attention. This introductory article aims to situate and describe the origin, theoretical foundations and methods and materials gathered in the three research projects included in this special issue. We also briefly describe some important characteristics of the Swedish school inspection and finish off with short introductions to the six articles.

Projektet ska studera frekvensomriktare ur ett antal aspekter såsom uppbyggnad, styrning och implementering i syfte att utveckla mer robusta frekvensomriktare och implementering av dessa för att säkerställa drift av svaga nät och ö-drift och minimera ytterligare driftstörningar vid svåra påfrestningar på elnätet.

- In recent decades, governing practices in education have become highly contradictory: deregulation and decentralisation are accompanied by re-regulation and increased centralisation, contributing to considerable governing tensions in and across different national systems and within the emergent European education policy space. On the one hand there is the persistence of performance monitoring through target-setting, indicators and benchmarks, and on the other, the promotion of self-evaluation and ‘light touch’ regulation that express a ‘softer’ governance turn, and promote self-regulation as the best basis for constant improvement.Drawing on research undertaken into three national systems, this edited volume explores the attempts to manage these tensions in Europe through the development of inspection as a governing practice. Inspectorates and inspectors offer key locations for the exploration of governing tensions, positioned as they are between the international, the national, and the local and institutional, and with responsibility for both regulation and development. All three national systems offer contrasting approaches to inspection, all of which have changed considerably in recent years.Governing by Inspection positions inspection in the framework of changing education policy and politics, and in a period of intensive policy development and exchange in Europe. It will be key reading for academics, researchers and postgraduate students in the fields of education, political science and social policy.

- It has been unclear whether ischemic stroke induces neurogenesis or neuronal DNA rearrangements in the human neocortex. Using immunohistochemistry; transcriptome, genome and ploidy analyses; and determination of nuclear bomb test-derived (14)C concentration in neuronal DNA, we found neither to be the case. A large proportion of cortical neurons displayed DNA fragmentation and DNA repair a short time after stroke, whereas neurons at chronic stages after stroke showed DNA integrity, demonstrating the relevance of an intact genome for survival.
