  • An absolute risk model to identify individuals at elevated risk for pancreatic cancer in the general population.

    • PURPOSE: We developed an absolute risk model to identify individuals in the general population at elevated risk of pancreatic cancer.PATIENTS AND METHODS: Using data on 3,349 cases and 3,654 controls from the PanScan Consortium, we developed a relative risk model for men and women of European ancestry based on non-genetic and genetic risk factors for pancreatic cancer. We estimated absolute risks based on these relative risks and population incidence rates.RESULTS: Our risk model included current smoking (multivariable adjusted odds ratio (OR) and 95% confidence interval: 2.20 [1.84-2.62]), heavy alcohol use (>3 drinks/day) (OR: 1.45 [1.19-1.76]), obesity (body mass index >30 kg/m(2)) (OR: 1.26 [1.09-1.45]), diabetes >3 years (nested case-control OR: 1.57 [1.13-2.18], case-control OR: 1.80 [1.40-2.32]), family history of pancreatic cancer (OR: 1.60 [1.20-2.12]), non-O ABO genotype (AO vs. OO genotype) (OR: 1.23 [1.10-1.37]) to (BB vs. OO genotype) (OR 1.58 [0.97-2.59]), rs3790844(chr1q32.1) (OR: 1.29 [1.19-1.40]), rs401681(5p15.33) (OR: 1.18 [1.10-1.26]) and rs9543325(13q22.1) (OR: 1.27 [1.18-1.36]). The areas under the ROC curve for risk models including only non-genetic factors, only genetic factors, and both non-genetic and genetic factors were 58%, 57% and 61%, respectively. We estimate that fewer than 3/1,000 U.S. non-Hispanic whites have more than a 5% predicted lifetime absolute risk.CONCLUSION: Although absolute risk modeling using established risk factors may help to identify a group of individuals at higher than average risk of pancreatic cancer, the immediate clinical utility of our model is limited. However, a risk model can increase awareness of the various risk factors for pancreatic cancer, including modifiable behaviors.

    • Background:The clinical course of World Health Organisation grade II gliomas remains variable and their time point of transformation into a more malignant phenotype is unpredictable. Identification of biological markers that can predict prognosis in individual patients is of great clinical value. PROX1 is a transcription factor that has a critical role in the development of various organs. PROX1 has been ascribed both oncogenic and tumour suppressive functions in human cancers. We have recently shown that PROX1 may act as a diagnostic marker for high-grade gliomas. The aim of this study was to address the prognostic value of PROX1 in grade II gliomas.Methods:A total of 116 samples were evaluated for the presence of PROX1 protein. The number of immunopositive cells was used as a variable in survival analysis, together with established prognostic factors for this patient group.Results:Higher PROX1 protein was associated with poor outcome. In the multivariate analysis, PROX1 was identified as an independent factor for survival (P=0.024), together with the presence of mutated isocitrate dehydrogenase 1 R132H protein, and with combined losses of chromosomal arms 1p/19q in oligodendrocytic tumours.Conclusion:PROX1 is a novel predictor of survival for grade II gliomas.

    • The structure of paper influences its properties and simulations of it are necessary to understand the impact of fiber and papermaking conditions on the sheet properties. We show a method to develop a representative structure of paper by merging different simulation techniques for the forming section and the pressing operation. The simulation follows the bending and drape of fibers over one another in the final structure and allows estimation of sheet properties without recourse to arbitrary bending rules or experimental measurements of density and/or RBA. Fibers are first modeled as jointed beams following the fluid mechanics in the forming section. The sheet structure obtained from this is representative of the wet sheet from the couch. The pressing simulation discretizes fibers into a number of solid elements around the lumen. Bonding between fibers is simulated using spring elements. The resulting fiber network was analyzed to determine its elastic modulus and deformation under small strains. The influence of fiber dimensions, namely fiber lengths, widths and thicknesses as well as bond stiffnesses on the elasticity of the network are studied. A brief account of inclusion of fines, represented by individual cubical elements is also shown.

    • Since health promotion was launched in 1986 through the Ottawa Charter it has suffered from the lack of a good theoretical foundation. However, it has now been demonstrated that the original salutogenic theory (The Sense of Coherence Theory of Antonovsky) matches the principles of the Ottawa Charter (OC). The evidence of a global systematic review on SOC proves that people and systems that have developed a strong SOC have a positive health outcome regarding the OC action areas. In 2007 IUHPE established a Global Working Group on Salutogenesis (GWG-SAL). In December 2010 10 research centers formed a core group for the GWG-SAL. These centers represent different profiles related to salutogenesis such as pollicy development, reorientation of health services, settings approaches such as workplace health, community action, health behaviours and learning for health, NCD interventions, development of research methods and indicators, contemporary evidence base and systematic reviews, the life-course approach, mental health, quality of life, population health and specific target groups such as youth and elderly... The proposed subkeynote/workshop or symposium proposes to coherently address these aspects where representatives of the Core Group speak to the core aspects of the above issues and discuss further research and interventions regarding the salutogenic approach to health and health promotion.

    • The first worldwide interlaboratory assesment on persistent organic pollutants (POPs) under the Stockholm Convention was organized in the Asian/Pacific, Latin American and African regions during 2009-11.A relatively large number of laboratories reported data for the PCDDs/PCDFs and dioxin-like PCBs, especially in the Asian region. Within the Asian region, several participants used high-resolution GC/high-resolution MS systems optimized for dioxin analysis. The availibility of High-resolution mass spectrometer instrumentation is limited in the Latin America and African regions, although recently several new laboratories for dioxins have started in the Latin American region.
