    • Ischemic heart disease is a leading cause of mortality due to irreversible damage to cardiac muscle. Inspired by the post-ischemic microenvironment, we devised an extracellular matrix (ECM)-mimicking hydrogel using catalyst-free click chemistry covalent bonding between two elastin-like recombinamers (ELRs). The resulting customized hydrogel included functional domains for cell adhesion and protease cleavage sites, sensitive to cleavage by matrix metalloproteases overexpressed after myocardial infarction (MI). The scaffold permitted stromal cell invasion and endothelial cell sprouting in vitro. The incidence of non-transmural infarcts has increased clinically over the past decade, and there is currently no treatment preventing further functional deterioration in the infarcted areas. Here, we have developed a clinically relevant ovine model of non-transmural infarcts induced by multiple suture ligations. Intramyocardial injections of the degradable ELRs-hydrogel led to complete functional recovery of ejection fraction 21 days after the intervention. We observed less fibrosis and more angiogenesis in the ELRs-hydrogel-treated ischemic core region compared to the untreated animals, as validated by the expression, proteomic, glycomic, and histological analyses. These findings were accompanied by enhanced preservation of GATA4(+) cardiomyocytes in the border zone of the infarct. We propose that our customized ECM favors cardiomyocyte preservation in the border zone by modulating the ischemic core and a marked functional recovery. The functional benefits obtained by the timely injection of the ELRs-hydrogel in a clinically relevant MI model support the potential utility of this treatment for further clinical translation.

  • Probing the uniformity of silver-doped epitaxial graphene by micro-Raman mapping

    • We present a Raman spectroscopy study on epitaxial graphene decorated with thin Ag films (2-15 nm), which are deposited using magnetron sputtering. We find that the presence of Ag on the graphene surface induces doping, the uniformity and efficiency of which is determined by Ag nominal thickness. Deposition of Ag films with thicknesses up to 5 nm favors the effective electron transfer from Ag to epitaxial graphene. A significant redshift and broadening of the 2D peak are observed with increasing the Ag-layer thickness above 5 nm, which is indicative of large strain and doping fluctuations. We also observe a non-trivial linear growth of 2D/G peak intensity ratio with increasing D/G ratio for all Ag-decorated samples, which is explained by increase of peak amplitude due to surface enhanced Raman scattering and charged impurity-induced screening caused by the presence of Ag on the graphene surface.

  • Conducting Polymer Paper-Based Cathodes for High-Areal-Capacity Lithium–Organic Batteries

    • Conducting polymers have been considered for use as cathode materials in rechargeable lithium‐ion batteries (LIBs) since 1981 but problems with poor cycling stability, rapid self‐discharge, and low energy and power densities have so far limited their applicability. Herein it is shown that nanostructured freestanding conducting polymer composites [e.g., polypyrrole (PPy) and polyaniline (PANI)] can be used to circumvent these shortcomings. Freestanding and binder‐free PPy and cellulose‐based composites can straightforwardly be used as versatile organic cathode materials for LIBs. The composite, reinforced with chopped carbon filaments (CCFs), exhibited a large active mass loading of approximately 10 mg cm−2, an areal capacity of 1.0 mAh cm−2 (corresponding to 102 mAh g−1), and stable cycling. With an active mass loading of 4.4 mg cm−2, a capacity of 0.22 mAh cm−2 (corresponding to 58 mAh g−1) was found for current densities of 5 A g−1 yielding discharge times of approximately 40 seconds, and a capacity retention of 91 % over 100 cycles was obtained at 0.2 A g−1. The present method constitutes a straightforward approach for the manufacturing of high‐performance freestanding electroactive conducting‐polymer‐based paper‐like electrodes for use in inexpensive and sustainable, high‐performance organic LIBs.

    • During the drying of sawn timber, hydrostatic tension forces within the cell may exceed the compressive strength perpendicular to the grain of the thin cell wall and the cell then collapses. This phenomenon is common in hardwoods such as Sequoia sempervirens, Thuja plicata, Tsuga heterophylla, Juglans nigraand many species of eucalyptus and oak. Usually, this leads to severe surface deformation, and both surface and internal checking(honeycombing) may occur. The quality of the final product is lowered by these cracks and deformations. The aim of this study was to investigate, by CT-scanning samples throughout the drying process, whether it is possible to detect when and how cracking and deformation occurs and develops in specimens of Eucalyptus nitens. Based on this knowledge, better drying schedules can be developed to improve the yield and ensure a higher quality of the sawn timber. Three specimens, one specimen in each drying run, of Eucalyptus nitens were used for the tests. Their cross-sectional dimensions, prior to drying, were 105x23 mm2 and their length was 70 cm. A specially designed laboratory drying kiln that fits within the gantry of a Siemens Somatom Emotion medical CT-scanner was used (Fig. 1). With this equipment, it is possible to scan the inside of the kiln without interrupting the drying process.

    • Introduction: In early-life, the immature mucosal barrier allows contact between the gut microbiota and the developing immune system. Due to their strategic location and their ability to sample luminal antigen, dendritic cells (DC) play a central role in the interaction of microbes and immune cells in the gut. Here, we investigated how two bacteria associated with opposite immune profiles in children, that is, Lactobacillus (L.) reuteri and Staphylococcus (S.) aureus, influenced the differentiation of monocytes in vitro as well how the generated DC impacted T cell responses.Methods: We exposed monocyte cultures to cell-free supernatants (CFS) from these bacteria during their differentiation to DC.Results: The presence of L. reuteri-CFS during DC differentiation resulted in DC with a more mature phenotype, in terms of up-regulated surface markers (HLA-DR, CD86, CD83, CCR7) and enhanced cytokine production (IL6, IL10, and IL23), but had a reduced phagocytic capacity compared with non-treated monocyte-derived DC (Mo-DC). However, upon LPS activation, L. reuteri-CFS-generated DC displayed a more regulated phenotype than control Mo-DC with notable reduction of cytokine responses both at mRNA and protein levels. In contrast, S. aureus-CFS-generated DC were more similar to control Mo-DC both without and after LPS stimulation, but they were still able to induce responses in autologous T cells, in the absence of further T cell stimulation.Conclusions: We show that bacterial signals during DC differentiation have a profound impact on DC function and possibly also for shaping the T cell pool.

  • Hemp Seeds in Post-Arthroplasty Rehabilitation: A Pilot Clinical Study and an In Vitro Investigation

    • Osteoarthritis is a type of degenerative joint disease that results from the breakdown of joint cartilage and underlying bone. Due to their antioxidants and anti-inflammatory action, the phytochemical constituents of many vegetable varieties could represent a new frontier for the treatment of patients with Osteoarthritis and are still being explored. The aim of this pilot human study was to investigate the effects of pasta enriched with hemp seed flour on osteoarticular pain and bone formation markers in patients in post-arthroplasty rehabilitation. Another purpose was to evaluate the effect of hemp seed extract on bone metabolism, in vitro. A pilot, controlled, clinical study was conducted to verify the feasibility of pain symptom reduction in patients with Osteoarthritis undergoing arthroplasty surgery. We also investigated the effect of hemp seed extract on the Wnt/beta-catenin and ERK1/2 pathways, alkaline phosphatase, RANKL, RUNX-2, osteocalcin, and COL1A on Saos-2. After 6 weeks, the consumption of hemp seed pasta led to greater pain relief compared to the regular pasta control group (-2.9 +/- 1.3 cm vs. -1.3 +/- 1.3 cm; p = 0.02). A significant reduction in serum BALP was observed in the participants consuming the hemp seed pasta compared to control group (-2.8 +/- 3.2 mu g/L vs. 1.1 +/- 4.3 mu g/L; p = 0.04). In the Saos-2 cell line, hemp seed extract also upregulated Wnt/beta-catenin and Erk1/2 pathways (p = 0.02 and p = 0.03) and osteoblast differentiation markers (e.g., ALP, OC, RUNX2, and COL1A) and downregulated RANKL (p = 0.02), compared to the control. Our study demonstrated that hemp seed can improve pain symptoms in patients with osteoarthritis undergoing arthroplasty surgery and also improves bone metabolism both in humans and in vitro. However, more clinical studies are needed to confirm our preliminary findings.

    • This thesis presents an object-oriented technique for rewriting abstract syntax trees in order to simplify compilation. The technique, Rewritable Reference Attributed Grammars (ReRAGs), is completely declarative and supports both rewrites and computations by means of attributes. We have implemented ReRAGs in our aspect-oriented compiler compiler tool JastAdd II. We present the ReRAG formalism, its evaluation algorithm, and examples of its use. JastAdd II uses three synergistic mechanisms for supporting separation of concerns: inheritance for model modularization, aspects for cross-cutting concerns, and rewrites that allow computations to be expressed on the most suitable model. This allows compilers to be written in a high-level declarative and modular fashion, supporting language extensibility as well as reuse of modules for different compiler-related tools. Each technique is presented using a series of simplified examples from static semantic analysis for the Java programming language. A case study is presented where ReRAGs are used extensively to implement a compiler for the Control Module extension to the IEC61131-3 automation languages. That Control Modules concept is further extended, in a modular fashion, with object-oriented features to improve encapsulation, composition mechanisms, code re-use, and type safety.
