  • Evanescent Light-Scattering Microscopy for Label-Free Interfacial Imaging: From Single Sub-100 nm Vesicles to Live Cells

    • Advancement in the understanding of biomolecular interactions has benefited greatly from the development of surface-sensitive bioanalytical sensors. To further increase their broad impact, significant efforts are presently being made to enable label-free and specific biomolecule detection with high sensitivity, allowing for quantitative interpretation and general applicability at low cost. In this work, we have addressed this challenge by developing a waveguide chip consisting of a flat silica core embedded in a symmetric organic cladding with a refractive index matching that of water. This is shown to reduce stray light (background) scattering and thereby allow for label-free detection of faint objects, such as individual sub-20 rim gold nanoparticles as well as sub-100 nm lipid vesicles. Measurements and theoretical analysis revealed that light-scattering signals originating from single surface-bound lipid vesicles enable characterization of their sizes without employing fluorescent lipids as labels. The concept is also demonstrated for label-free measurements of protein binding to and enzymatic (phospholipase A2) digestion of individual lipid vesicles, enabling an analysis of the influence on the measured kinetics of the dye-labeling of lipids required in previous assays. Further, diffraction-limited imaging of cells (platelets) binding to a silica surface showed that distinct subcellular features could be visualized and temporally resolved during attachment, activation, and spreading. Taken together, these results underscore the versatility and general applicability of the method, which due to its simplicity and compatibility with conventional microscopy setups may reach a widespread in life science and beyond.

    • A waveguide structure for evanescent wave microscopy and/or spectroscopy, comprising an optically transparent core layer, a lower dielectric cladding layer and an upper dielectric cladding layer arranged on opposite sides of the core layer. The core layer has a refractive index higher than the refractive indices of the cladding layers. The upper cladding layer is made of an organic material. A sample well is arranged on an upper surface of the core layer formed by a cavity in the upper cladding layer, the sample well being adapted to contain a sample medium with one or more sample objects. The core layer is made of a first dielectric inorganic material, and the upper cladding layer has a refractive index which closely matches the refractive index of the sample medium. A method for manufacturing such waveguide structure, and a measurement system comprising the waveguide structure are also disclosed.

  • Microscopic Simulation of Pedestrian Traffic

    • There has recently been a renewed interest in planning for pedestrian traffic, primarily in connection to public transport interchange stations, since these are important for public transport to constitute an attractive alternative to car usage. This thesis concerns microscopic simulation of pedestrian traffic, which is a promising tool for analyzing and predicting the traffic situation in a given pedestrian facility; particularly powerful when the traffic is congested. Important applications of microscopic simulation include comparison of possible infrastructure designs such as proposed interchange stations, and evaluations of various traffic management solutions, for example information systems.The purpose of this thesis is to advance the capabilities of pedestrian microsimulation toward a level at which it can be reliably applied for quantitative analysis by practitioners in the field. The work is based on an established microscopic model of pedestrian dynamics, the Social Force Model (sfm), and the advances are made in a number of different areas.To be able to evaluate and compare simulated traffic situations suitable performance measures are needed. A set of local performance measures are proposed that quantifies the local delay rate density and estimates the discomfort perceived by the pedestrians.The sfm is extended to include waiting pedestrians through the introduction of a waiting model, demonstrated to be stable and free from oscillations. The inclusion of waiting pedestrians in the model is critical for accurate modelling of public transport interchange stations, where large groups of waiting pedestrians may hinder passing pedestrians if the design of the station is poor.The relaxation time of the adaptation to the preferred velocity is an important parameter in force based models of pedestrian traffic since it affects several behaviors of the simulated pedestrians, two of which are linear acceleration and turning movements. A comparison of observations of accelerating pedestrians reported in the literature and new observations of turning pedestrians indicates that no value of the relaxation time can give model behavior consistent with both sets of observations. This indicates that modifications of the model is needed to accurately reproduce the observed behavior.An important input to simulations is the preferred speed of the simulated pedestrians. The common assumption that the preferred speed distribution at a location does not vary during the day is tested through observations of pedestrian traffic at Stockholm Central Station. The results demonstrate that the preferred speeds are lower in the afternoon than in the morning, implying that the preferred speed should be treated as a source of uncertainty when applying pedestrian microsimulation.Finally, a sensitivity analysis of a simulation of the lower hall of Stockholm Central Station is performed to find the most important sources of uncertainty in the model predictions, given the available data. The results indicate that the uncertainty related to calibration is the largest of the considered potential error sources.

  • Affinity Purification and Single-Molecule Analysis of Integral Membrane Proteins from Crude Cell-Membrane Preparations

    • The function of integral membrane proteins is critically dependent on their naturally surrounding lipid membrane. Detergent-solubilized and purified membrane proteins are therefore often reconstituted into cell-membrane mimics and analyzed for their function with single-molecule microscopy. Expansion of this approach toward a broad range of pharmaceutically interesting drug targets and biomarkers however remains hampered by the fact that these proteins have low expression levels, and that detergent solubilization and reconstitution often cause protein conformational changes and loss of membrane-specific cofactors, which may impair protein function. To overcome this limitation, we here demonstrate how antibody-modified nanoparticles can be used to achieve affinity purification and enrichment of selected integral membrane proteins directly from cell membrane preparations. Nanoparticles were first bound to the ectodomain of β-secretase 1 (BACE1) contained in cell-derived membrane vesicles. In a subsequent step, these were merged into a continuous supported membrane in a microfluidic channel. Through the extended nanoparticle tag, a weak (∼fN) hydrodynamic force could be applied, inducing directed in-membrane movement of targeted BACE1 exclusively. This enabled selective thousand-fold enrichment of the targeted membrane protein while preserving a natural lipid environment. In addition, nanoparticle-targeting also enabled simultaneous tracking analysis of each individual manipulated protein, revealing how their mobility changed when moved from one lipid environment to another. We therefore believe this approach will be particularly useful for separation in-line with single-molecule analysis, eventually opening up for membrane-protein sorting devices analogous to fluorescence-activated cell sorting.

  • Topology optimization of periodic 3D heat transfer problems with 2D design

    • We consider a model for density-based topology optimization (TO) of stationary heat transfer problems with design-dependent internal convection in 3D structures with periodic design obtained by extruding a 2D design in 3D. The internal convection takes place at the interface between a solid material and a cooling fluid in internal channels through the design domain. The objective of the TO is to minimize the maximum temperature, which is approximated by means of an L-p norm. The finite element method is used to discretize the state problem and the resulting optimization problem is solved using gradient-based methods. The internal convection is modeled to be dependent on the design density gradient in the continuous problem. In discrete form, it is approximated as proportional to the difference in design densities of adjacent elements in the finite element mesh. The theory is illustrated by numerical examples based on a simplified guide vane geometry.

    • Background: Lateral sinus membrane elevation with simultaneous implant placement without grafting material (graft‐less LSFE) is a widely investigated method for bone augmentation of the maxillary sinus floor. Long‐term follow‐up studies are rare.Purpose: This study aimed to investigate the long‐term effects of implants placed with graft‐less LSFE.Materials and methods: The study group was comprised of 111 patients previously treated with graft‐less LSFE. The first follow‐up visit, which occurred after a mean of 5 years after surgery, included a clinical examination, cone beam computerized tomography, and panorama or intraoral radiography. The second follow‐up included panorama or intraoral radiography, and it was conducted after a mean of 8 years.Results: Overall, 218 implants were placed in 127 sinuses. Nine of the 218 implants failed resulting in an overall implant survival of 95.9%. The average bone gain at the follow‐up was 4.0 ±2.0 mm.Conclusion: The implant‐supported rehabilitation achieved using graft‐less LSFE was stable over time, and there was no or little impact on sinus health. Furthermore, it was concluded that the new bone formation and the amount of bone gain is proportional to the length of the implant protruding into the sinus cavity.

    • We have investigated the gas composition-dependent faceting of a Rh(553) surface during catalytic CO oxidation under semirealistic reaction conditions using Surface X- ray Diffraction. We find that under stoichiometric CO and O2 conditions, the Rh(553) surface maintains its surface orientation without facet formation. In oxygen excess, the CO oxidation reaction becomes mass transfer limited by the CO diffusion, and the sur- face is observed to expose (331) or (11 ̄1) facets in coexistence with larger (111) terraces. The observed facet formation has previously been observed for pure O2 exposures of the Rh(553) surface, but at significantly lower O2 partial pressures. Surprisingly, in CO excess, which results in a CO poisoned surface with low activity, we instead find coexisting (110) and (111) facets. The reasons for and possible implications of the observed facetings are discussed.

    • The fourth generation of the Sloan Digital Sky Survey (SDSS-IV) has been in operation since 2014 July. This paper describes the second data release from this phase, and the 14th from SDSS overall (making this Data Release Fourteen or DR14). This release makes the data taken by SDSS-IV in its first two years of operation (2014-2016 July) public. Like all previous SDSS releases, DR14 is cumulative, including the most recent reductions and calibrations of all data taken by SDSS since the first phase began operations in 2000. New in DR14 is the first public release of data from the extended Baryon Oscillation Spectroscopic Survey; the first data from the second phase of the Apache Point Observatory (APO) Galactic Evolution Experiment (APOGEE-2), including stellar parameter estimates from an innovative data-driven machine-learning algorithm known as "The Cannon"; and almost twice as many data cubes from the Mapping Nearby Galaxies at APO (MaNGA) survey as were in the previous release (N = 2812 in total). This paper describes the location and format of the publicly available data from the SDSS-IV surveys. We provide references to the important technical papers describing how these data have been taken (both targeting and observation details) and processed for scientific use. The SDSS web site (www.sdss.org) has been updated for this release and provides links to data downloads, as well as tutorials and examples of data use. SDSS-IV is planning to continue to collect astronomical data until 2020 and will be followed by SDSS-V.

Därför stödjer vi deras krav, skriver 270 klimatforskare.

  • Seve terranes of the Kebnekaise Mts., Swedish Caledonides, and their amalgamation, accretion and affinity

    • A major allochthon of the Scandinavian Caledonides, the Seve belt has traditionally been considered to be derived from the rifted margin and continent-ocean transition (COT) of Baltica. However, geochronological results obtained from its inferred northern equivalent, the Kalak Nappe Complex (KNC), have been taken to indicate an exotic affinity of this complex and of also Seve terranes, an interpretation adopted in recent palaeogeographic models. In the Kebnekaise Mts., the COT is represented by the Kebnekaise terrane composed of amphibolitized dykes of gabbro and dolerite of depleted magma source and rare felsic and ultramafic rocks. Coronitic dolerite and gabbro with abundant rutile suggest high pressures before or during amalgamation with the underlying Mårma terrane, composed of quartzofeldspathic gneisses intruded by mafic and granitic rocks, the latter including a previously dated c. 845 Ma-old granite. The granite mingled with mildly alkaline dolerites chemically similar to transitional basalts of continental rifts. Following emplacement at shallow (andalusite stability) crustal levels, the igneous complex and host rocks underwent extensive deformation, metamorphism within the sillimanite-kyanite stability field and local migmatization. The Kebnekaise and Mårma terranes amalgamated in early Ordovician as indicated by the U-Pb age of 487±7 Ma obtained from titanite fabrics of deformed granite in thrust vicinity, and by 40Ar-39Ar results. The pressure increase in both terranes suggests that amalgamation occurred during initial subduction and imbrication. Correlation of the Mårma terrane with the KNC is discussed. Results obtained in this study give no reason to ascribe an exotic affinity to the Seve terranes of the Kebnekaise Mts.
