    • The production of free radicals in human neutrophils was studied in both Pneumocystis carinii derived from cultures of L2 rat lung epithelial-like cells and Pneumocystis carinii purified from human lung. Using the cytochrome C technique, which selectively measured extracellular superoxide generation, hardly any free radical production was observed after stimulation with cultured rat-derived P. carinii. A chemiluminescence technique, which separately measured intra- and extracellular free radical production, was subsequently employed to differentiate the free radical generation. It was established that 1) P. carinii stimulated intra- but not extracellular free radical production in human neutrophils, 2) opsonized cultured rat-derived P. carinii stimulated human neutrophils to a strong intracellular response of superoxide production, and 3) opsonized P. carinii, purified from human lung also stimulated human neutrophils to produce intracellular free radicals.

    • The Yersinia pseudotuberculosis cell surface-located protein invasin was found to promote binding between the pathogen and resting peripheral B cells via beta 1 integrin receptors (CD29). B cells responded by expressing several activation markers and by growing, In contrast, T cells did not react, although these cells express CD29. An isogenic invA mutant failed to activate B cells. The mutation could be complemented by providing the invA+ gene in trans. Purified invasin alone did not activate B cells, although it was able to block the binding of bacteria to the cells.

  • Heparin-binding protein targeted to mitochondrial compartments protects endothelial cells from apoptosis

    • Neutrophil-borne heparin-binding protein (HBP) is a multifunctional protein involved in the progression of inflammation. HBP is stored in neutrophil granules and released upon stimulation of the cells in proximity to endothelial cells. HBP affects endothelial cells in multiple ways; however, the molecular and cellular mechanisms underlying the interaction of HBP with these cells are unknown. Affinity isolation and enzymatic degradation demonstrated that HBP released from human neutrophils binds to endothelial cell-surface proteoglycans, such as syndecans and glypican. Flow cytometry indicated that a significant fraction of proteoglycan-bound HBP is taken up by the endothelial cells, and we used radiolabeled HBP to determine the internalization rate of surface-bound HBP. Confocal and electron microscopy revealed that internalized HBP is targeted to perinuclear compartments of endothelial cells, where it colocalizes with mitochondria. Western blotting of isolated mitochondria from HBP-treated endothelial cells showed that HBP is present in 2 forms - 28 and 22 kDa. Internalized HBP markedly reduced growth factor deprivation-induced caspase-3 activation and protected endothelial cells from apoptosis, suggesting that uptake and intracellular routing of exogenous HBP to mitochondria contributes to the sustained viability of endothelial cells in the context of locally activated neutrophils.

  • Women's experience of the encounter with the midwife during childbirth

    • OBJECTIVE: to describe women's experience of the encounter with the midwife during childbirth.DESIGN: a qualitative study using a phenomenological approach. Data were collected via tape-recorded interviews.SETTING: the Alternative Birth Care Centre, Sahlgrenska University Hospital, Gothenburg, Sweden in 1994.PARTICIPANTS: 18 women, six primiparous and 12 multiparous who were two to four days post delivery.KEY FINDINGS: the essential structure of the studied phenomenon was described as 'presence' and included three themes: to be seen as an individual, to have a trusting relationship and to be supported and guided on one's own terms.IMPLICATIONS FOR PRACTICE: the need to be seen as an individual can be realised by affirmation and familiarity with the midwife and surroundings. A trusting relationship can be obtained by good communication and proficient behaviour. By providing a sense of control the women can be supported and guided on their own terms. Above all they must feel that the midwife is present.

  • Coverage- and temperature-dependent site occupancy of carbon monoxide on Rh(111) studied by high-resolution core-level photoemission

    • High-resolution core-level photoemission is used to study structural aspects for the molecular adsorption of CO on the Rh(111) single-crystal surface, and in particular to derive the adsorption sites. The site sensitivity of the core-level binding energy and the fact that the core level photoemission signal is proportional to the adsorbate coverage make it possible to study quantitatively how the occupation of different sites changes with temperature and/or CO coverage. For the CO/Rh(111) adsorption system we find two sites (on-top and three-fold hollow) to be occupied by the CO molecules. At coverages up to 0.33 ML only on-top sites are occupied, whereas at higher coverages a mixture of three-fold hollow and on-top sites are found. The distribution between these two sites is found to depend strongly on temperature. Quantitative studies of these reversible, temperature-dependent site changes have been carried out for a number of CO coverages. For coverages between 0.33 and ∼0.54 ML, increasing the temperature results in part of the molecules moving from on-top to three-fold hollow sites. This change is strongest for a (4 × 4) structure formed at 0.5 ML where an order-disorder transition is observed at a temperature of 120 K. For coverages above ∼0.54 ML, increasing the temperature leads instead to a decrease of the relative occupation of the three-fold hollow sites. For coverages below 0.33 ML, the molecules occupy on-top sites at all temperatures.

  • On the adsorption sites for CO on the Rh(111) single crystal surface

    • High resolution photoemission applied to the C 1s and Rh 3d core levels has been used to investigate the adsorption sites at low temperature of CO on the Rh(111) single crystal surface as a function of CO coverage. Two different sites are found to be occupied by the CO molecules. At coverages up to 0.5 monolayers the majority of the molecules are found to adsorb in on-top sites whereas at higher coverages three-fold hollow sites become increasingly populated. The different C 1s binding energy of the CO molecules in these two sites makes it possible to measure diffraction induced intensity variations versus photon energy in a site specific manner. The saturation (2 x 2)-3CO structure formed at a coverage of 0.75 monolayers is argued to contain one on-top and two three-fold hollow molecules per unit cell.
