  • Cell–cell adherence as a selection method for the generation of anti-melanoma monoclonal antibodies

    • The aim of the present study was to obtain monoclonal antibodies (mAbs) recognising human melanoma-associated antigens after immunisation of BALB/c mice with a 70–150 kDa membrane fraction from melanoma tumour tissues. Screening of specific antibody- producing hybridomas was performed using a novel cell–cell adherence method with the melanoma cell line M-14. Three mAbs of IgG1 isotype were selected: Mel-1, Mel-2 and Mel-3 which recognised the immunogen by ELISA and stained several melanoma cell lines positive in immunofluorescence. The molecular weight of the antigen was studied by different methods; a 170-kDa band was identified following immunoblotting of tumour lysate and a 72-kDa band was observed following immunoaffinity purification. Cell–cell adherence appears to be a reliable procedure for the generation of mAbs against native cellular antigens.

  • Recent advances in electrochromics for smart windows applications

    • Electrochromic smart windows are able to vary their throughput of radiant energy by low-voltage electrical pulses. This function is caused by reversible shuttling of electrons and charge balancing ions between an electrochromic thin film and a transparent counter electrode. The ion transport takes place via a solid electrolyte. Charge transport is evoked by a voltage applied between transparent electrical conductors surrounding the electrochromic film/electrolyte/counter electrode stack. This review summarizes recent progress concerning: (i) calculated optical properties of crystalline WO3, (ii) electrochromic properties of heavily disordered W oxide and oxyfluoride films produced by reactive magnetron bias sputtering, (iii) novel transparent reactively sputter-deposited Zr-Ce oxide counter electrodes and (iv) a new proton-conducting antimonic-acid-based polymer electrolyte. Special in depth presentations are given on elastic light scattering from W-oxide-based films and of electronic band structure effects affecting opto-chronopotentiometry data in Zr-Ce oxide.   • Li intercalation in transparent Ti-Ce oxide films : Energetics and ion dynamics

    • Films of Ti dioxide, mixed Ti–Ce oxide, and Ce dioxide were produced by reactive dc magnetron sputtering.Electrochemical lithiation was probed by chronopotentiometry, cyclic voltammetry together with optical transmittance recording, and impedance spectroscopy. Evidence was found for inserted electrons being accommodated in Ce4f states; this contention was supported by preliminary results from x-rayabsorption fine-structure spectroscopy. These electrons do not produce luminous electrochromism. The variation of the chemical diffusion coefficient of Li, with film composition and Li content, was also studied.

  • Physical properties data at Hole 959D : comparison of core and log measurements and a proposed revision of lithologic units

    • A revised division of lithologic units is proposed for Hole 959D in the interval from 400 to 1000 meters below seafloor from Ocean Drilling Program Leg 159 based on combining the geophysical and sedimentological information. This new division is essential for further geological studies of the Cote d’Ivoire-Ghana Transform Margin area. A comparison is made between shipboard laboratory measurements made on core samples and in situ downhole logging data. Logging data, in conjunction with physical properties data, provide a more detailed record of the variation in the sediment column, which enables a more qualitative assessment of lithotype distribution. Emphasis is placed on distinguishing between the lithologic factors influencing the geophysical signals and artifacts resulting from imperfect data collection. Good overall agreement between core and log data for rotary core barrel cored sediments at Hole 959D is observed, and effects of core expansion caused by the removal of overburden pressure do not appear significant.

  • Towards the smart window : progress in electrochromics

    • Electrochromic devices have the ability to produce reversible and persistent changes of their optical properties. The phenomenon is associated with joint ion and electron transport into/out of an electrochromic thin film, in most cases being a transition metal oxide. This paper outlines the various applications of such devices in smart windows suitable for energy-conscious architecture, in variable-reflectance mirrors, and in display devices. Critical materials issues and design concepts are discussed. The paper also covers two specific research topics: computed electronic structure of crystalline WO3 incorporating ionic species, showing how reflectance modulation emerges from a first-principles calculation; and Li+ dynamics in heavily disordered Ti oxide, illustrating how diffusion constants derived from impedance spectroscopy can be reconciled with the Anderson—Stuart model.

    • The karyotypes of three cloned stocks, CL Brener (CL), CA I/72 (CA) and Sylvio X10/7 (X10), of Trypanosoma cruzi were studied by pulsed-field gel electrophoresis followed by ethidium bromide staining and hybridization with 35 different probes, 30 of which identified single chromosomes. The chromosome-specific probes identified between 26 and 31 chromosomal bands in the three cloned stocks, corresponding to 20 unique chromosomes in CL and 19 in CA and X10. Considering the DNA content of the parasite, it was predicted that the markers recognise at least half of all T. cruzi chromosomes. A majority of identified chromosomes showed large differences in size among different strains, in some cases by up to 50%. Interestingly, CL had in general larger chromosomes than the two other studied cloned stocks. Several of the markers showed linkage and nine different linkage groups were identified, each comprising 2-4 markers. The linkage between the markers was maintained in 8 of the 9 linkage groups when a panel comprising 26 different T. cruzi strains representing major T. cruzi populations was tested. One linkage group was found to be maintained in some strains but not in others. This result shows that chromosomal rearrangements occur in the T. cruzi genome, albeit with a low frequency. Repetitive DNA, both non-coding and in one case coding, was more abundant in the cloned stock CL Brener than in CA and X10. The information presented will make it possible to select chromosomes for the construction of physical chromosomal maps required for the T. cruzi genome project.
