    • Type 2 diabetes mellitus is a serious global health problem that is hard to treat in the long term, prompting great efforts to find new drug targets. Little attention has been paid, however, to the metabolic significance of the microcirculation in insulin-sensitive tissues, despite the fact that microvascular insulin resistance and endothelial dysfunction are closely associated and have been shown to precede Type 2 diabetes [1]. Endothelial nitric oxide plays a major role in mediating the beneficial effects of insulin on capillary recruitment and muscle glucose uptake [2] and in suppression of inflammatory pathways, including those activated by a high-fat diet [3].   • A pathology atlas of the human cancer transcriptome

    • Cancer is one of the leading causes of death, and there is great interest in understanding the underlying molecular mechanisms involved in the pathogenesis and progression of individual tumors. We used systems-level approaches to analyze the genome-wide transcriptome of the protein-coding genes of 17 major cancer types with respect to clinical outcome. A general pattern emerged: Shorter patient survival was associated with up-regulation of genes involved in cell growth and with down-regulation of genes involved in cellular differentiation. Using genome-scale metabolic models, we show that cancer patients have widespread metabolic heterogeneity, highlighting the need for precise and personalized medicine for cancer treatment. All data are presented in an interactive open-access database (www.proteinatlas.org/pathology) to allow genome-wide exploration of the impact of individual proteins on clinical outcomes.

  • Resilience to Intentional Electromagnetic Interference Is Required for Connected Autonomous Vehicles

    • The connected autonomous vehicle (CAV) will never be completely autonomous; on the contrary, it will be heavily dependent on so-called vehicular ad hoc networks (VANETs) for its function. To deserve the trust of the general public, the vehicles as well as the intelligent transport system (ITS) infrastructure must be able to handle not only natural disturbances but also attacks of malicious nature. In this paper, we discuss the effects of antagonistic attacks using intentional electromagnetic interference (IEMI) and how the antagonistic nature of the threat renders probabilistic risk analysis inadequate for the defense of the vehicles and the infrastructure. Instead, we propose a shift toward resilience engineering and vulnerability analysis to manage antagonistic threats. Finally, we also give two examples of possible scenarios to illustrate the type of situations a CAV must be able to handle.

  • Conjugation factors controlling F-plasmid antibiotic resistance transmission

    • The rapid horizontal transmission of many antibiotic resistance genes between bacterial host cells on conjugative plasmids is a major cause of the accelerating antibiotic resistance crisis. Preventing understanding and targeting conjugation, there currently are no experimental platforms for fast and cost-efficient screening of genetic effects on antibiotic resistance transmission by conjugation. We introduce a novel experimental framework to screen for conjugation based horizontal transmission of antibiotic resistance between >60,000 pairs of cell populations in parallel. Plasmid-carrying donor strains are constructed in high throughput. We then mix the resistance plasmid carrying donors with recipients in a design where only transconjugants can reproduce, measure growth in dense intervals and extract transmission times as the growth lag. As proof-of-principle, we exhaustively explored chromosomal genes controlling F plasmid donation within E. coli populations, by screening the Keio deletion collection at high replication. We recover all six known chromosomal gene mutants affecting conjugation and identify >50 novel factors, all of which diminish antibiotic resistance transmission. We verify 10 of the novel genes' effects in a liquid mating assay. The new framework holds great potential for exhaustive disclosing of candidate targets for helper drugs that delay resistance development in patients and societies and improves the longevity of current and future antibiotics.

    • Myocardial triglycerides stored in lipid droplets are important in regulating the intracellular delivery of fatty acids for energy generation in mitochondria, for membrane biosynthesis, and as agonists for intracellular signaling. Previously, we showed that deficiency in the lipid droplet protein perilipin 5 (Plin5) markedly reduces triglyceride storage in cardiomyocytes and increases the flux of fatty acids into phospholipids. Here, we investigated whether Plin5 deficiency in cardiomyocytes alters mitochondrial function. We found that Plin5 deficiency reduced mitochondrial oxidative capacity. Furthermore, in mitochondria from Plin5((-/)(-)) hearts, the fatty acyl composition of phospholipids in mitochondrial membranes was altered and mitochondrial membrane depolarization was markedly compromised. These findings suggest that mitochondria isolated from hearts deficient in Plin5, have specific functional defects.

  • Service innovations enabled by the "internet of things"

    • The purpose of this paper is to develop a new conceptual framework that reflects network dynamics in Internet of Things (IoT)-enabled service innovation processes. Design/methodology/approach -- Based on literature on service innovations, business networks and IoT, dynamic concepts are selected. Aided by information about an evolving case "The connected vehicle", propositions about interaction between the variables in the framework are formulated. Findings -- A conceptual framework consisting of four interacting variables: overlapping, intermediating, objectification of actors and business modelling is developed, linking several streams of research. Propositions are motivated and issues for further research questions formulated. Research limitations/implications -- The framework may stimulate further research on IoT-enabled service innovations. Practical implications -- Understanding network dynamics for developing and implementing business models for service innovations.

    • This paper presents the design and preliminary testing of a full-body assistive exoskeleton AXO-SUIT for older adults. AXO-SUIT is a system of modular exoskeletons consisting of lower-body and upper-body modules, and their combination as full body as well to provide flexible physical assistance as needed. The full-body exoskeleton comprises 27 degrees of freedom, of which 17 are passive and 10 active, which is able to assist people in walking, standing, carrying and handling tasks. In the paper, design of the AXO-SUIT is described. End-user testing results are presented to show the effectiveness of the exoskeleton in providing flexible physical assistance.

    • Isolated hepatic perfusion (IHP) is a procedure where the liver is surgically isolated and perfused with a high concentration of the chemotherapeutic agent melphalan. Briefly, the procedure starts with the setup of a percutaneous veno-venous bypass from the femoral vein to the external jugular vein. Via a laparotomy, catheters are then inserted into the proper hepatic artery and the caval vein. The portal vein and the caval vein, both supra- and infrahepatically, are then clamped. The arterial and venous catheters are connected to a heart lung machine and the liver is perfused with melphalan (1 mg/kg body weight) for 60 min. This way it is possible to locally perfuse the liver with a high dose of a chemotherapeutic agent, without leakage to the systemic circulation. In previous studies including patients with isolated liver metastases of uveal melanoma, an overall response rate of 33-100% and a median survival between 9 and 13 months, have been reported. The aim of this protocol is to give a clear description of how to perform the procedure and to discuss IHP as a treatment option for liver metastases of uveal melanoma.
