  • Effect of unlabeled helper probes on detection of an RNA target by bead-based sandwich hybridization

    • Unlabeled helper oligonucleotides assisting a bead-based sandwich hybridization assay were tested for the optimal placement of the capture and detection probes. The target used was a full-length in vitro synthesized mRNA molecule. Helper probes complementary to regions adjacent to the binding site of the 5' end attached capture probe were found much more effective than helper probes targeting positions adjacent to the detection probe binding site. The difference is believed to be caused by a disruption of the RNA secondary structure in the area where the capture probe binds, thereby reducing structural interference from the bead. The use of additional helpers showed an additive effect. Using helpers, at both sides of the capture and detection probes showed a 15- to 40-fold increase in hybridization efficiency depending on the target, thereby increasing the sensitivity of the hybridization assays. Using an electrical chip linked to the detection probe for the detection of p-ominophenol, which is produced by alkaline phosphatase, a detection limit of 2 x 10(-13) M mRNA molecules was reached without the use of a nucleic acid amplification step.

    • In August 23-25, 2007, the Scandinavian Society for Prosthetic Dentistry in collaboration with the Danish Society of Oral Implantology arranged a consensus conference on the topic 'Implants and/or teeth'. It was preceded by a workshop in which eight focused questions were raised and answered in eight review articles using a systematic approach. Twenty-eight academicians and clinicians discussed the eight review papers with the purpose to reach consensus on questions relevant for the topic. At the conference the consensus statements were presented as well as lectures based on the review articles. In this article the methods used at the consensus workshop are briefly described followed by the statements with comments.

    • The Leendertz dual-beam symmetric illumination-normal observation arrangement is widely employed for real time evaluation of in-plane displacement components as well as surface shape. Instead of observing along the optical axis, we have examined the Leendertz arrangement by observing the scattered light along the direction of the illumination beams, and imaged it as two separate images onto the photo sensor of a CCD camera. The interferometer is a combination of two channels, each of which measures independently and simultaneously the information pertaining to either the in-plane displacement component of a deformation vector, or the surface relief variation of a three-dimensional object. In addition, a summary of possible measurements that can be carried out from the present arrangement is also highlighted. Experimental results using a four-frame phase shifting technique are illustrated

  • Five-year clinical prospective evaluation of zirconia-based Denzir 3-unit FPDs

    • Purpose: The aim of this prospective study was to evaluate the clinical performance of fully sintered hot isostatic pressed yttria-partially-stabilized zirconia (Denzir) 3-unit fixed partial dentures (FPDs).Materials and Methods: Nineteen 3-unit FPDs were placed in 18 patients. Ten FPDs were placed in the maxilla and 9 in the mandible. Two calibrated examiners evaluated the FPDs independently 1 week (baseline), 1 year, 3 years, and 5 years after placement using the California Dental Association quality evaluation system.Results: All FPDs were intact at the 5-year examination. One FPD lost retention after 12 months but remained intact; it was recemented and is still in function after 5 years. All FPDs were rated satisfactory over 5 years. No changes were seen in terms of color and anatomic form. The number of slightly rough or pitted occlusal surfaces increased approximately 30% over 5 years. Visible evidence of ditching along the margin increased over time, but only for those FPDs luted with zinc phosphate cement.Conclusion: The 5-year results indicate that yttria-partially-stabilized zirconia 3-unit FPDs with anatomically designed frameworks are promising prosthetic alternatives, even in the premolar and molar regions. However, for all-ceramic FPDs with more units in function, further studies are necessary.

  • Increased apoptosis and increased clonogenic survival of 12V-H-ras transformed rat fibroblasts in response to cisplatin

    • Mutationally activated Ras is involved in tumor progression and likely also in drug resistance. Using survival, viability and apoptosis assays, we have here compared the cisplatin sensitivities of FR3T3 rat fibroblasts and a 12V-H-ras transformed subline (Ras2:3). Around 24 h after cisplatin treatment Ras2:3 cells showed higher apoptosis levels and lower viability than FR3T3. This increased sensitivity correlated with weaker cisplatin-induced activation of Jun N-terminal kinase (JNK). In contrast to apoptosis assays, colony formation assays showed that Ras2:3 were more resistant to cisplatin than were FR3T3. This was partly due to the increased cisplatin sensitivity of FR3T3 seeded at low densities, as required in colony formation assays. In addition, Ras2:3 cisplatin survivors had a higher relative proliferative capacity. Cell cycle analyses showed that FR3T3 cells initially responded with a dose-dependent G2 arrest, while Ras2:3 accumulated in S-phase. Experiments with an anti-apoptotic mutant of MEKK1 suggested that the apoptotic response of Ras2:3 cells is not specific to the S-phase fraction. In summary, the cisplatin response of ras-transformed fibroblasts is distinct from that of parental cells, in that they show increased apoptosis, a different cell cycle response and increased post-treatment proliferative capacity. The results illustrate the need to carefully consider methods and protocols for in vitro studies on chemotherapy sensitivity.
