  • Six-quasiparticle isomer in Nd-140

    • A search for isomeric states was performed in the nucleus Nd-140(60)80 using in-beam gamma-ray spectroscopy and the Te-126(O-18,4n) reaction. Prompt and delayed gamma-ray coincidences were measured with the AFRODITE spectrometer using the pulsed beam delivered by the Separated Sector Cyclotron of iThemba LABS. One new isomer was identified, with spin-parity I-pi=20(+) and lifetime T-1/2 >= 400 ns, at an excitation energy E-x=7430 keV. The lifetime of the 10(+) isomer at E-x=3619 keV was determined to be 32.9(1.8) ns, confirming the previously reported value. The configuration of the I-pi=20(+) isomer is assigned based on configuration-dependent cranked Nilsson-Strutinsky (CNS) calculations as a state pi(d(5/2)g(7/2))(10)(+-4)circle times nu(h(11/2)(-2))(10)(+), with the spin vectors of the six holes in the Gd-146(64)82 core fully aligned.

    • The identification and annotation of protein-coding genes is one of the primary goals of whole-genome sequencing projects, and the accuracy of predicting the primary protein products of gene expression is vital to the interpretation of the available data and the design of downstream functional applications. Nevertheless, the comprehensive annotation of eukaryotic genomes remains a considerable challenge. Many genomes submitted to public databases, including those of major model organisms, contain significant numbers of wrong and incomplete gene predictions. We present a community-based reannotation of the Aspergillus nidulans genome with the primary goal of increasing the number and quality of protein functional assignments through the careful review of experts in the field of fungal biology.   • Possible chirality in the doubly-odd Tl-198 nucleus: Residual interaction at play

    • A candidate for chiral bands was found in Tl-198 for the first time in a mass region of oblate (or nonaxial with gamma >= 30 degrees) deformed nuclei. Two bands show very similar quasiparticle alignments, moments of inertia, and B(M1)/B(E2) ratios. They have a relative excitation energy of about 500 keV and different patterns of energy staggering. Calculations using the two-quasiparticle-plus-triaxial-rotor model with residual proton-neutron interaction included show that a triaxial deformation with gamma similar to 44 degrees agrees very well with all the experimental observations. Furthennore, considerable energy staggering for both partner bands was calculated for this pi h(9/2) circle times vi(13/2)(-1) configuration at gamma - 30 degrees, suggesting that chiral bands may have substantial energy staggering.

    • High-spin states in Bi-197 were studied with the AFRODITE gamma-ray array at iThemba LABS using the Ta-181(Ne-22, 6n) reaction at a beam energy of 125 MeV. A new shears band was found and linked to the low-lying states in Bi-197. Its dynamic moment of inertia, F (2), is considerably larger than the ((2)) of the shears bands in the neighbouring Pb isotopes. This is probably a result of the involvement of an additional high-K h(9/2) proton orbital.

    • Best vitelliform macular dystrophy (BMD) is an autosomal dominant inherited macular degenerative disease caused by mutations in the gene BEST1 (formerly VMD2). Prior reports indicate that BMD is characterized histopathologically by accumulation of lipofuscin in the retinal pigment epithelium (RPE). However, this accumulation has not been quantified and the chemical composition of lipofuscin in BMD has not been examined. In this study we characterize the histopathology of a donor eye from a rare individual homozygous for a mutation (W93C) in BEST1. We find that this individual's disease was not any more severe than has been described for heterozygotes. We then used this tissue to quantify lipofuscin accumulation by enriching intracellular granules from RPE cells on sucrose gradients and counting the granules in each density fraction. Granules from the homozygous donor eye as well as a donor eye from an individual heterozygous for the mutation T6R were compared with age-matched control eyes. Interestingly, the least dense fraction, representing classical lipofuscin granules was either not present or significantly diminished in the BMD donor eyes and the autoflourescence associated with lipofuscin had shifted to denser fractions. However, a substantial enrichment for granules in fractions of higher density was also noted in the BMD samples. Inspection of granules from the homozygous donor eye by electron microscopy revealed a complex abnormal multilobular structure. Analysis of granules by HPLC indicated a 1.6- and fourfold overall increase in A2E in the BMD eyes versus age-matched control eyes, with a shift of A2E to more dense granules in the BMD donor eyes. Despite the increase in A2E and total intracellular granules, the RPE in the homozygous donor eyes was relatively well preserved. Based on these data we conclude that the clinical and histopathologic consequences to the homozygous donor were not any more severe than has been reported previously for individuals who are established or presumptive heterozygotes. We find that A2E is a component of the lipofuscin accumulated in BMD and that it is more abundant than in control eyes suggesting that the etiology of BMD is similar to Stargardt's disease and Stargardt-like macular dystrophy. Finally, the changes we observe in the granules suggest that the histopathology and eventual vision loss associated with BMD may be due to defects in the ability of the RPE to fully degrade phagocytosed photoreceptor outer segments.

    • Age-related macular degeneration (AMD) is a leading cause of blindness in the elderly. In its severest form, choroidal neovessels breach the macular Bruch's membrane, an extracellular matrix compartment comprised of elastin and collagen laminae, and grow into the retina. We sought to determine whether structural properties of the elastic lamina (EL) correspond to the region of the macula that is predilected toward degeneration in AMD. Morphometric assessment of the macular and extramacular regions of 121 human donor eyes, with and without AMD, revealed a statistically significant difference in both the integrity (P < 0.0001) and thickness (P < 0.0001) of the EL between the macular and extramacular regions in donors of all ages. The EL was three to six times thinner and two to five times less abundant in the macula than in the periphery. The integrity of the macular EL was significantly lower in donors with early-stage AMD (P = 0.028), active choroidal neovascularization (P = 0.020), and disciform scars (P = 0.003), as compared to unaffected, age-matched controls. EL thickness was significantly lower only in individuals with disciform scars (P = 0.008). The largest gaps in macular EL integrity were significantly larger in all categories of AMD (each P < 0.0001), as compared to controls. EL integrity, thickness, and gap length in donors with geographic atrophy did not differ from those of controls. These structural properties of the macular EL correspond spatially to the distribution of macular lesions associated with AMD and may help to explain why the macula is more susceptible to degenerative events that occur in this disease.

    • Lifetimes of high-spin states have been measured for 142Gd and 134Nd with EU-ROBALL IV and AFRODITE, respectively, using DSAM. From calculations in the cranked Nilsson-Strutinsky model it was concluded that at the high-spin states of the (+,0)1 band, 142Gd represents a triaxial nucleus rotating around the longest principal axis.
