    • In this study, 1,833 systemic sclerosis (SSc) cases and 3,466 controls were genotyped with the Immunochip array. Classical alleles, amino acid residues, and SNPs across the human leukocyte antigen (HLA) region were imputed and tested. These analyses resulted in a model composed of six polymorphic amino acid positions and seven SNPs that explained the observed significant associations in the region. In addition, a replication step comprising 4,017 SSc cases and 5,935 controls was carried out for several selected non-HLA variants, reaching a total of 5,850 cases and 9,401 controls of European ancestry. Following this strategy, we identified and validated three SSc risk loci, including DNASE1L3 at 3p14, the SCHIP1-IL12A locus at 3q25, and ATG5 at 6q21, as well as a suggested association of the TREH-DDX6 locus at 11q23. The associations of several previously reported SSc risk loci were validated and further refined, and the observed peak of association in PXK was related to DNASE1L3. Our study has increased the number of known genetic associations with SSc, provided further insight into the pleiotropic effects of shared autoimmune risk factors, and highlighted the power of dense mapping for detecting previously overlooked susceptibility loci.

    • To discuss and share knowledge around advances in the care of patients with thrombotic disorders, the Third International Symposium of Thrombosis and Anticoagulation was held in So Paulo, Brazil, from October 14-16, 2010. This scientific program was developed by clinicians for clinicians, and was promoted by four major clinical research institutes: the Brazilian Clinical Research Institute, the Duke Clinical Research Institute of the Duke University School of Medicine, the Canadian VIGOUR Centre, and the Uppsala Clinical Research Center. Comprising 3 days of academic presentations and open discussion, the symposium had as its primary goal to educate, motivate, and inspire internists, cardiologists, hematologists, and other physicians by convening national and international visionaries, thought-leaders, and dedicated clinician-scientists. This paper summarizes the symposium proceedings.

    • An increasing need exists to quantify the impact of diagenesis on complex and heterogeneous geological reservoirs and to predict the related field-scale porosity-permeability distribution. Development of approaches to quantify diagenetic events is an important step towards the construction of integrated numerical reservoir models predicting the impact of successive diagenetic events on petrophysical properties at various scales. This article describes two distinct approaches, which allow capturing, quantifying and explaining reservoir heterogeneities of the Jurassic Arab Formation (offshore oilfield in Abu Dhabi, U.A.E.) at the field and plug scale. Based on a classical diagenesis study, whereby the major diagenetic phases were characterized, distribution patterns were highlighted to further explain reservoir heterogeneities through a set of 2D interpolation diagenesis maps across the field. The best reservoir properties are encountered in the central-northern (grainstones with syntaxial calcite overgrowth cement – typical of Arab D) and south-western (dolostones – typical of Arab C) parts of the oilfield. Following another independent approach at the scale of the plug, quantification of rock components and description of the pore space connectivity and geometry are obtained by 2D and 3D image analyses, μ-CT and pore network modelling. This latter approach resulted in a better understanding of the effect of anhydrite dissolution on the pore structure of the investigated dolostone. The overall, presented workflow provides valuable constraints for assessing reservoir heterogeneities in statistical field-scale models.

  • Moderation and mediation of the effect of attention training in social anxiety disorder

    • While attention modification programs (AMP) have shown promise as laboratory-based treatments for social anxiety disorder, trials of internet-delivered AMP have not yielded significant differences between active and control conditions. To address these inconsistencies, we examined the moderational and mediational role of attention bias in the efficacy of attention training. We compared data reported by Carlbring et al. (2012) to an identical AMP condition, with the exception that participants were instructed to activate social anxiety fears prior to each attention training session (AMP + FACT; n = 39). We also compared all attention training groups to an internet-delivered cognitive-behavioral therapy (iCBT) condition (n = 40). Participants in the AMP + FACT group experienced greater reductions in social anxiety symptoms than both active (n = 40) and control (n = 39) groups reported by Carlbring et al., and did not differ in symptom reductions from the iCBT group. Higher attention bias predicted greater symptom reductions for participants who completed AMP, but not for the control group. Moreover, change in attention bias mediated the relationship between AMP group (active condition reported by Carlbring et al. versus AMP + FACT) and change in social anxiety symptoms. These results suggest the importance of interpreting findings related to symptom change in attention training studies in the context of bias effects.
