  • An anergic immune signature in the tumor microenvironment of classical Hodgkin lymphoma is associated with inferior outcome

    • Objective: The classical Hodgkin lymphoma (cHL) tumor microenvironment shows anongoing inflammatory response consisting of varying degrees of infiltrating eosinophils,mast cells, macrophages, regulatory T lymphocytes (Tregs), and activated lymphocytes surrounding the malignant cells. Herein, different immune signatures are characterized and correlated with treatment outcome.Methods: Tumor-infiltrating leukocytes were phenotyped in biopsies from 459 patients with cHL. Time to progression (TTP) (primary progression, relapse, or death from cHL) and overall survival were analyzed using Cox proportional hazards regression.Results: The leukocyte infiltration in the microenvironment was highly diverse between patients and was categorized in 4 immune signatures (active, anergic, innate, or mixed). A high proportion of Tregs (anergic) resulted in shorter TTP (median 12.9-year follow-up) in age-adjusted analyses (hazard ratio = 1.82; 95% confidence interval 1.05-3-15). Epstein-Barrvirus (EBV)-positive cases had higher proportions of macrophages and activated lymphocytes than EBV negative, but neither of those leukocytes predicted prognosis.Conclusions: Abundant Tregs (anergic signature) indicate a shorter TTP, particularly in younger patients. This is probably due to a reduced ability of the immune system to attack the tumor cells. Our data warrant further investigation if these suggested immune signatures could predict outcome of immunotherapy such as immune checkpoint inhibitors.

  • Towards automatic discovery and assessment of vulnerability severity in cyber-physical systems

    • Despite their wide proliferation, complex cyber–physical systems (CPSs) are subject to cybersecurity vulnerabilities and potential attacks. Vulnerability assessment for such complex systems are challenging, partly due to the discrepancy among mechanisms used to evaluate their cyber-security weakness levels. Several sources do report these weaknesses like the National Vulnerability Database (NVD), as well as manufacturer websites besides other security scanning advisories such as Cyber Emergency Response Team (CERT) and Shodan databases. However, these multiple sources are found to face inconsistency issues, especially in terms of vulnerability severity scores. We advocate an artificial intelligence based approach to streamline the computation of vulnerability severity magnitudes. This approach decreases the error rate induced by manual calculation processes, that are traditionally used in cybersecurity analysis. Popular repositories such as NVD and SecurityFocus are employed to validate the proposed approach, assisted with a query method to retrieve vulnerability instances. In doing so, we report discovered correlations among reported vulnerability scores to infer consistent magnitude values of vulnerability instances. The method is applied to a case study featuring a CPS application to illustrate the automation of the proposed vulnerability scoring mechanism, used to mitigate cybersecurity weaknesses.

  • On the resource efficiency of kraft lignin extraction

    • Lignin is regarded as a promising raw material for the production of biobased products, such as chemicals, materials and fuels, and will most probably be a key component in future lignocellulosic biorefineries.This thesis examines the lignin extraction process in a kraft pulp mill, the technologies that are available for this purpose, and the impact made on the mill. Several different kraft lignin extraction processes and technologies are currently available and are basically linear: chemicals are brought from outside the mill and introduced into the process and the mill balance. Depending on their origin, the addition of these chemicals will affect the mill to a lesser or greater degree, both economically and environmentally. A conceivable way of reducing the impact made on the mill´s balance would be the in-house production of the chemicals used, sulphuric acid and CO2, which takes a more sustainable circular approach.The results obtained show that utilisation of existing process streams in the mill as a source of chemicals could be a way of not only reducing these impacts but also making lignin extraction more sustainable. Internal production of sulphuric acid is possible and could generate a substantial amount for use as replacement of the fresh sulphuric acid needed for the lignin extraction process; CO2 is available in large quantities in the mill and could be captured and used for lignin extraction.

  • On Visual Area Coverage Using Micro Aerial Vehicles

    • The aim of this Licentiate is to advance the field of cooperative visual coverage path planners for multiple Micro Aerial Vehicles (MAVs), while aiming for their real life adoption towards the tasks of aerial infrastructure inspection. The fields that will be addressed are focusing in: a) the collaborative perception of the environment, b) the collaborative visual inspection, and c) the optimization of the aerial missions based on the remaining flying battery, camera constraints, coverage constraints and other real life mission induced constraints.Towards this envisioned aim, this Licentiate will present the following main theoretical contributions: a) centralized and distributed Model Predictive Control (MPC) schemes for the cooperative motion control of MAVs focusing in the establishing of a formation control architecture to enable a dynamic visual sensor from monocular cameras towards a reconfigurable environmental perception, b) revisiting the Cooperative Coverage Path Planning (C-CPP) problem for the inspection of complex infrastructures, c) developing a holistic approach to the problems of 2-D area coverage with MAVs for polygon areas, while considering the camera footprint, and d) designing of a scheme to estimate the Remaining Useful Life (RUL) of the battery during a flight mission, a fact that directly effects the flying capabilities of the MAVs. The theoretical contributions of this thesis have been extensively evaluated in simulation and real life large scale field trials, a direction that adds another contribution of the suggested framework towards the massive insertion of the aerial platforms as aerial tools in the close future.In the first part of this Licentiate, the vision, motivation, open challenges, contributions, and future works are discussed, while in the second part the full articles connected to the presented contributions in this Licentiate are presented in the annex.

    • In the Protestant tradition the trade and commerce that surrounded different aspects of Catholic spirituality was regarded as something relatively alien. In Protestant Sweden the critique towards these aspects of Catholicism became an essential component in the anti-Catholic rhetoric that was recurring during the latter nineteenth and the early twentieth century. From the perspective of Lutheran theologians in Sweden, this part of Catholic spirituality was regarded as superstitious and a way for the clergy to deceit ordinary people.

  • The Prognostic Role of LRIG Proteins in Endometrial Cancer

    • Endometrial cancer (EC) is the most common gynecologic malignancy in Sweden and it has various prognostic factors. The LRIG family is a group of three integral surface proteins with a similar domain organization. The study aimed to explore LRIG family as prognostic factor proteins in EC. The initial study cohort included 100 women with EC who were treated at the Department of Women’s and Children’s Health, Karolinska University Hospital Solna, between 2007 and 2012. We assessed the associations between LRIG protein expression and type, grade, and stage of EC, as well as progression-free and overall survival. Immunohistochemistry results revealed that most women in the analytical sample had >50% LRIG1-, LRIG2- and LRIG3-positive cells. A statistically significant association was observed between having a high number of LRIG3-positive cells and superior overall survival (incidence rate ratio = 0.977; 95% confidence interval: 0.958–0.996, p = 0.019). Moreover, positive LRIG3 staining of the cell membrane was associated with reducing in the risk of death (hazard ratio = 0.23; 95% confidence interval: 0.09–0.57). Our results show that LRIG3 expression might be a prognostic factor in EC. The role of LRIG1 and LRIG2 expression remains to be further investigated.
