    • Throughout the Northern Hemisphere, changes in local and regional climate conditions are coupled to the recurring and persistent large-scale patterns of pressure and circulation anomalies spanning vast geographical areas, the so-called teleconnection patterns. Indeed, the atmospheric fluctuations described by the North Atlantic Oscillation (NAO) are closely associated with the last four decades of inter-annual variability in local snow and ice conditions observed in the Arctic. Since the NAO has also been connected with changes in the global climate, the behaviour of species, communities and other ecosystem elements at Zackenberg in relation to the NAO enables us to view these in circumpolar and global contexts. Large-scale systems like the NAO constitute the link between the global change and local climate variability to which ecosystem components respond. Here, we place selected ecosystem elements from the monitoring programme Zackenberg Basic presented in previous chapters in a circumpolar context related to NAO-mediated climatic changes. We begin by linking the local variability in winter weather conditions at Zackenberg to fluctuations in the NAO. We then proceed by linking the observed intra- and inter-annual behaviour of selected ecosystem elements to changes in the NAO. The functional ecosystem characteristics in focus are landscape gas exchange dynamics phenological patterns at different trophic levels, consumer-resource dynamics and community stability. The influence of the NAO is presented and discussed in a broader perspective based on information obtained from other arctic localities. The relation between the NAO and the Zackenberg winter weather, is nonlinear, reflecting differential effects of the NAO as the index moves between high and low phases. The inverse hyperbolic relationship found between the NAO and the amount of winter snow was also evident as non-linear response in organisms and systems to inter-annual changes in the NAO. Responses investigated included growth and reproduction in plants and animals, population dynamics and synchrony, inter-trophic interactions and community stability together with system feedback dynamics.

    • Arabidopsis MAP kinase 4 (MPK4) functions as a regulator of pathogen defense responses, because it is required for both repression of salicylic acid (SA)-dependent resistance and for activation of jasmonate (JA)-dependent defense gene expression. To understand MPK4 signaling mechanisms, we used yeast two-hybrid screening to identify the MPK4 substrate MKS1. Analyses of transgenic plants and genome-wide transcript profiling indicated that MKS1 is required for full SA-dependent resistance in mpk4 mutants, and that overexpression of MKS1 in wild-type plants is sufficient to activate SA-dependent resistance, but does not interfere with induction of a defense gene by JA. Further yeast two-hybrid screening revealed that MKS1 interacts with the WRKY transcription factors WRKY25 and WRKY33. WRKY25 and WRKY33 were shown to be in vitro substrates of MPK4, and a wrky33 knockout mutant was found to exhibit increased expression of the SA-related defense gene PR1. MKS1 may therefore contribute to MPK4-regulated defense activation by coupling the kinase to specific WRKY transcription factors.

    • Decision making for incompetent patients is a much-discussed topic in bioethics. According to one influential decision making standard, the substituted judgment standard, the decision that ought to be made for the incompetent patient is the decision the patient would have made, had he or she been competent. Although the merits of this standard have been extensively debated, some important issues have not been sufficiently explored. One fundamental problem is that the substituted judgment standard, as commonly formulated, is indeterminate in content and thus offers the surrogate little or no guidance. What the standard does not specify is just how competent one should imagine the patient to be, and what else one ought to envision about the patient’s hypothetical outlook and the circumstances surrounding his or her decision making. The article discusses this problem of underdetermined decision conditions.

    • OBJECTIVES: Prostate cancer (PC) is a highly lethal neoplastic disease affecting the physical, mental and social well-being of patients, i.e. their quality of life (QOL). Patients suffering from metastatic PC are faced with serious decisions regarding treatment strategies. Therefore, QOL information has become a crucial element of decision making in this group of patients. The first objective of this study was to describe QOL in a group of patients diagnosed with metastatic PC and skeletal metastases. At the time of evaluation the patients had not received any treatment but were evaluated before entering a study of androgen-modulating therapy (the Scandinavian Prostate Cancer Group study 5). The second objective was to identify demographic and disease-related factors affecting QOL. MATERIAL AND METHODS: A total of 917 patients with metastatic PC were evaluated using a well-described and validated questionnaire [European Organization for Research and Treatment of Cancer Quality-of-Life Questionnaire-C30 (EORTC QLQ-C30)]. The characteristics of the PC were noted, and simultaneously patients were evaluated with respect to use of analgesics, pain and performance status using a scoring system. Biochemical tests were performed when patients entered the study. A multivariate regression analysis was performed to analyse the correlations between QOL scores, patient demographics and disease-related data. RESULTS: The patients reported QOL scores significantly lower than those in the background population. Pain and fatigue were pronounced, whereas dyspnoea, insomnia, loss of appetite, constipation and diarrhoea were less prominent. Patients with high tumour grades, high PSPA scores (the sum of the pain score, the performance status and the use of analgesics) and those using analgesics had significantly lower QOL scores than the other patients. CONCLUSIONS: Patients with metastatic PC have reduced QOL. Our findings are in line with those of other studies of QOL among patients with this disease as evaluated by means of the EORTC QLQ-C30 questionnaire. Baseline data from studies like this provide important information when treatment modalities for PC are evaluated.

    • Apolipoprotein A-V (apoA-V) is present in low amounts in plasma and has been found to modulate triacylglycerol levels in humans and in animal models. ApoA-V displays affinity for members of the low density lipoprotein receptor (LDL-R) gene family, known as the classical lipoprotein receptors, including LRP1 and SorLA/LR11. In addition to LDL-A binding repeats, the mosaic receptor SorLA/LR11 also possesses a Vps10p domain. Here we show that apoA-V also binds to sortilin, a receptor from the Vsp10p domain gene family that lacks LDL-A repeats. Binding of apoA-V to sortilin was competed by neurotensin, a ligand that binds specifically to the Vps10p domain. To investigate the biological fate of receptor-bound apoA-V, binding experiments were conducted with cultured human embryonic kidney cells transfected with either SorLA/LR11 or sortilin. Compared with nontransfected cells, apoA-V binding to SorLA/LR11- and sortilin-expressing cells was markedly enhanced. Internalization experiments, live imaging studies, and fluorescence resonance energy transfer analyses demonstrated that labeled apoA-V was rapidly internalized, co-localized with receptors in early endosomes, and followed the receptors through endosomes to the trans-Golgi network. The observed decrease of fluorescence signal intensity as a function of time during live imaging experiments suggested ligand uncoupling in endosomes with subsequent delivery to lysosomes for degradation. This interpretation was supported by experiments with (125)I-labeled apoA-V, demonstrating clear differences in degradation between transfected and nontransfected cells. We conclude that apoA-V binds to receptors possessing LDL-A repeats and Vsp10p domains and that apoA-V is internalized into cells via these receptors. This could be a mechanism by which apoA-V modulates lipoprotein metabolism in vivo.

  • Monitoring of power system events at transmission and distribution level

    • Monitoring of events in the power system provides a great deal of insight into the behavior of the system. Events with impact on the entire power system typically occur in or near the transmission network and are therefore best monitored at transmission system substations. This puts great demands on the equipment and in general restricts the use of the data since it becomes property of the transmission system operator. An alternative is to measure at lower voltage levels. This paper documents the close agreement between voltage and frequency at low and high voltage levels during a remote and a nearby fault in the transmission network. The very high quality of the data suggests that valuable information about the transmission system can be obtained at lower voltages.
