  • Adipose tissue transcriptomics and epigenomics in low birthweight men and controls : role of high-fat overfeeding

    • Aims/hypothesis Individuals who had a low birthweight (LBW) are at an increased risk of insulin resistance and type 2 diabetes when exposed to high-fat overfeeding (HFO). We studied genome-wide mRNA expression and DNA methylation in subcutaneous adipose tissue (SAT) after 5 days of HFO and after a control diet in 40 young men, of whom 16 had LBW. Methods mRNA expression was analysed using Affymetrix Human Gene 1.0 ST arrays and DNA methylation using Illumina 450K BeadChip arrays. Results We found differential DNA methylation at 53 sites in SAT from LBW vs normal birthweight (NBW) men (false discovery rate < 5%), including sites in the FADS2 and CPLX1 genes previously associated with type 2 diabetes. When we used reference-free cell mixture adjustments to potentially adjust for cell composition, 4,323 sites had differential methylation in LBW vs NBW men. However, no differences in SAT gene expression levels were identified between LBW and NBW men. In the combined group of all 40 participants, 3,276 genes (16.5%) were differentially expressed in SAT after HFO (false discovery rate < 5%) and there was no difference between LBW men and controls. The most strongly upregulated genes were ELOVL6, FADS2 and NNAT; in contrast, INSR, IRS2 and the SLC27A2 fatty acid transporter showed decreased expression after HFO. Interestingly, SLC27A2 expression correlated negatively with diabetes- and obesity-related traits in a replication cohort of 142 individuals. DNA methylation at 652 CpG sites (including in CDK5, IGFBP5 and SLC2A4) was altered in SAT after overfeeding in this and in another cohort. Conclusions/interpretation Young men who had a LBW exhibit epigenetic alterations in their adipose tissue that potentially influence insulin resistance and risk of type 2 diabetes. Short-term overfeeding influences gene transcription and, to some extent, DNA methylation in adipose tissue; there was no major difference in this response between LBW and control participants.

  • Fasting unmasks differential fat and muscle transcriptional regulation of metabolic gene sets in low versus normal birth weight men

    • Background: Individuals born with low birth weight (LBW) have an increased risk of metabolic diseases when exposed to diets rich in calories and fat but may respond to fasting in a metabolically preferential manner. We hypothesized that impaired foetal growth is associated with differential regulation of gene expression and epigenetics in metabolic tissues in response to fasting in young adulthood. Methods: Genome-wide expression and DNA methylation were analysed in subcutaneous adipose tissue (SAT) and skeletal muscle from LBW and normal birth weight (NBW) men after 36 h fasting and after an isocaloric control study using microarrays. Findings: Transcriptome analyses revealed that expression of genes involved in oxidative phosphorylation (OXPHOS) and other key metabolic pathways were lower in SAT from LBW vs NBW men after the control study, but paradoxically higher in LBW vs NBW men after 36 h fasting. Thus, fasting was associated with downregulated OXPHOS and metabolic gene sets in NBW men only. Likewise, in skeletal muscle only NBW men downregulated OXPHOS genes with fasting. Few epigenetic changes were observed in SAT and muscle between the groups. Interpretation: Our results provide insights into the molecular mechanisms in muscle and adipose tissue governing a differential metabolic response in subjects with impaired foetal growth when exposed to fasting in adulthood. The results support the concept of developmental programming of metabolic diseases including type 2 diabetes.   • Impact of age, BMI and HbA1c levels on the genome-wide DNA methylation and mRNA expression patterns in human adipose tissue and identification of epigenetic biomarkers in blood.

    • Increased age, BMI and HbA1c levels are risk factors for several non-communicable diseases. However, the impact of these factors on the genome-wide DNA methylation pattern in human adipose tissue remains unknown. We analyzed DNA methylation of ∼480,000 sites in human adipose tissue from 96 males and 94 females, and related methylation to age, BMI and HbA1c. We also compared epigenetic signatures in adipose tissue and blood. Age was significantly associated with both altered DNA methylation and expression of 1,050 genes (e.g. FHL2, NOX4 and PLG). Interestingly, many reported epigenetic biomarkers of ageing in blood, including ELOVL2, FHL2, KLF14 and GLRA1, also showed significant correlations between adipose tissue DNA methylation and age in our study. The most significant association between age and adipose tissue DNA methylation was found upstream of ELOVL2. We identified 2,825 genes (e.g. FTO, ITIH5, CCL18, MTCH2, IRS1 and SPP1) where both DNA methylation and expression correlated with BMI. Methylation at previously reported HIF3A sites correlated significantly with BMI in females only. HbA1c (range 28-46 mmol/mol) correlated significantly with methylation of 711 sites, annotated to e.g. RAB37, TICAM1 and HLA-DPB1. Pathway analyses demonstrated that methylation levels associated with age and BMI are overrepresented among genes involved in cancer, type 2 diabetes and cardiovascular disease. Our results highlight the impact of age, BMI and HbA1c on epigenetic variation of candidate genes for metabolic diseases and cancer in human adipose tissue. Importantly, we demonstrate that epigenetic biomarkers in blood can mirror age-related epigenetic signatures in target tissues for metabolic diseases such as adipose tissue.

    • Syftet med artikeln är att bidra till kunskapandet om pedagogisk dokumentation med specifikt fokus på dokumentationens betydelse ur ett demokratiperspektiv. Trots många studier kring pedagogisk dokumentation finns det få studier som explicit utgår ifrån barn och vårdnadshavares perspektiv. I artikeln analyseras barns och vårdnadshavares samtal utifrån minnesbilder, vilka framträder i fokusgruppssamtal, från förskoletiden i relation till pedagogiskt dokumentationsarbete. Resultatet visar att den pedagogiska dokumentationspraktiken bidrog till skapandet av en praktikgemenskap på förskolan vilket i sin tur möjliggjorde en demokratisk undervisning.

  • Between ideals and domestic practices: the micro-geographies and media technologies of home management : A Swedish case study

    • The home is constituted of materials, cultural meanings and practices (Blunt & Dowling 2006), which means, among other things, that we make home through our domestic practices (Hollows 2008). In an ongoing cultural historical project on home management we are studying practices of domestic paperwork during the 20th century in Sweden. Through interviews with people born before 1940, and the study of public documents, the projects focuses on an area that are often neglected in research about the home. In this paper we take the writing desk and paperwork practices as a starting point to discuss the micro geographies and media technologies of home management. In focusing on these concrete objects, we activate questions about everyday life, gender roles, relation between home and work, and memory practices, as well as ideas about modernity and industrialization in Sweden. During the middle of the 20th century in Sweden the home was an object of rationalization – something that manifested in the famous scientific studies and standardization of the kitchen. In parallel to this modern project many people still lived in informal economy under poor circumstances, in old homes furnished with old furniture. The home can here be seen as an interesting example of different time scales.

    • Type 1 diabetes mellitus (T1D) is a metabolic disease causing hyperglycemia due to β-cell destruction. Despite adequate treatment, complications such as diabetic retinopathy (DR) are common. The first aim was to investigate if acute onset of type 1 diabetes differed between those who had developed retinopathy and who had not after 15 years from diagnosis. The second aim was to investigate if mean glycosylated hemoglobin (HbA1c) levels affect the time to development of DR. The medical records of all children and adolescents diagnosed with type 1 diabetes during 1993-2001 in our area in Sweden were studied retrospectively and the mean HbA1c each year until the development of retinopathy was investigated. In total 72 patients were included and the follow-up time was between 15 and 23 years. Gender, p-glucose, age and HbA1c at diagnosis were analyzed for possible correlations to years to retinopathy. HbA1c was significantly higher among those who had developed DR after 15 years from diagnosis, 98±9.2 (n=25) vs. 86±9.2 (n=46; p=0.025). A negative correlation was found between age at diagnosis and years to DR (rs=-0.376; p=0.026). Mean HbA1c levels at years 6-10 after diabetes diagnosis correlated significantly (rs=-0.354, p=0.037) to years until retinopathy. Mean HbA1c levels at years 1-15 after diabetes diagnosis were significantly higher at years 2-3 and years 5-8 for those who had developed retinopathy after 15 years from diagnosis. Higher HbA1c levels shortened the time to development of retinopathy. It is therefore important to keep HbA1c as close to normal as possible.

    • Climate-change is predicted to drive a migration of European beech (Fagus sylvatica) into the boreal Norway spruce (Picea abies) zone. Still, the mechanisms underpinning beech success on novel soils is little understood. Further, projected increasing summer temperatures will increase risk of fire and subsequent charcoal deposition. Here we investigate how soil type and presence of charcoal affect growth and key plant traits of beech and spruce seedlings.Beech and spruce seedlings were grown in pots with forest soils of beech- or spruce origin to which beech- or spruce derived charcoal was added. Concentrations of phenolic compounds, carbon (C) and nitrogen (N) were analysed separately for root, stem, and leaf tissues.Our spruce forest soil contained more N than the beech forest soil, and both beech- and spruce seedlings grew bigger in spruce forest soil. Beech seedlings also had overall lower tissue concentrations of phenolic compounds when grown in spruce soil. For both species, shoots and roots displayed opposite phenolic responses. The addition of charcoal had no effect on growth, while effects on phenolic compounds were largely idiosyncratic.Our results indicate that beech expansion is not limited by soil factors in the study area, nor is it facilitated by increased levels of charcoal.

  • Hepatic DPP4 DNA methylation associates with fatty liver

    • Hepatic DPP4 expression is elevated in subjects with ectopic fat accumulation in the liver. However, whether increased dipeptidyl peptidase 4 (DPP4) is involved in the pathogenesis or is rather a consequence ofmetabolic disease is not known. We therefore studied the transcriptional regulation of hepatic Dpp4 in young mice prone to diet-induced obesity. Already at 6 weeks of age, expression of hepatic Dpp4 was increased in mice with high weight gain, independent of liver fat content. In the same animals, methylation of four intronic CpG sites was decreased, amplifying glucose-induced transcription of hepatic Dpp4. In older mice, hepatic triglyceride content was increased only in animals with elevated Dpp4 expression. Expression and release of DPP4 were markedly higher in the liver compared with adipose depots. Analysis of human liver biopsy specimens revealed a correlation of DPP4 expression and DNA methylation to stages of hepatosteatosis and nonalcoholic steatohepatitis. In summary, our results indicate a crucial role of the liver in participation to systemic DPP4 levels. Furthermore, the data show that glucoseinduced expression of Dpp4 in the liver is facilitated by demethylation of the Dpp4 gene early in life. This might contribute to early deteriorations in hepatic function, which in turn result in metabolic disease such as hepatosteatosis later in life.

    • Context. The first long-term in-situ observation of the plasma environment in the vicinity of a comet, as provided by the European Rosetta spacecraft.Aims. Here we offer characterisation of the solar wind flow near 67P/Churyumov-Gerasimenko (67P) and its long term evolution during low nucleus activity. We also aim to quantify and interpret the deflection and deceleration of the flow expected from ionization of neutral cometary particles within the undisturbed solar wind.Methods. We have analysed in situ ion and magnetic field data and combined this with hybrid modeling of the interaction between the solar wind and the comet atmosphere.Results. The solar wind deflection is increasing with decreasing heliocentric distances, and exhibits very little deceleration. This is seen both in observations and in modeled solar wind protons. According to our model, energy and momentum are transferred from the solar wind to the coma in a single region, centered on the nucleus, with a size in the order of 1000 km. This interaction affects, over larger scales, the downstream modeled solar wind flow. The energy gained by the cometary ions is a small fraction of the energy available in the solar wind.Conclusions. The deflection of the solar wind is the strongest and clearest signature of the mass-loading for a small, low-activity comet, whereas there is little deceleration of the solar wind.
