  • A transcriptional timetable of autumn senescence

    • Background We have developed genomic tools to allow the genus Populus (aspens and cottonwoods) to be exploited as a full-featured model for investigating fundamental aspects of tree biology. We have undertaken large-scale expressed sequence tag (EST) sequencing programs and created Populus microarrays with significant gene coverage. One of the important aspects of plant biology that cannot be studied in annual plants is the gene activity involved in the induction of autumn leaf senescence. Results On the basis of 36,354 Populus ESTs, obtained from seven cDNA libraries, we have created a DNA microarray consisting of 13,490 clones, spotted in duplicate. Of these clones, 12,376 (92%) were confirmed by resequencing and all sequences were annotated and functionally classified. Here we have used the microarray to study transcript abundance in leaves of a free-growing aspen tree (Populus tremula) in northern Sweden during natural autumn senescence. Of the 13,490 spotted clones, 3,792 represented genes with significant expression in all leaf samples from the seven studied dates. Conclusions We observed a major shift in gene expression, coinciding with massive chlorophyll degradation, that reflected a shift from photosynthetic competence to energy generation by mitochondrial respiration, oxidation of fatty acids and nutrient mobilization. Autumn senescence had much in common with senescence in annual plants; for example many proteases were induced. We also found evidence for increased transcriptional activity before the appearance of visible signs of senescence, presumably preparing the leaf for degradation of its components.

  • A Populus EST resource for plant functional genomics

    • Trees present a life form of paramount importance for terrestrial ecosystems and human societies because of their ecological structure and physiological function and provision of energy and industrial materials. The genus Populus is the internationally accepted model for molecular tree biology. We have analyzed 102,019 Populus ESTs that clustered into 11,885 clusters and 12,759 singletons. We also provide >4,000 assembled full clone sequences to serve as a basis for the upcoming annotation of the Populus genome sequence. A public web-based EST database (POPULUSDB) provides digital expression profiles for 18 tissues that comprise the majority of differentiated organs. The coding content of Populus and Arabidopsis genomes shows very high similarity, indicating that differences between these annual and perennial angiosperm life forms result primarily from differences in gene regulation. The high similarity between Populus and Arabidopsis will allow studies of Populus to directly benefit from the detailed functional genomic information generated for Arabidopsis, enabling detailed insights into tree development and adaptation. These data will also valuable for functional genomic efforts in Arabidopsis.

  • Designing leisure applications for the mundane car-commute

    • Commuting by car from home to work can be very time consuming. We have conducted a study to explore what people are doing, and want to do, while commuting. People use their time in the car on a wide variety of activities with great innovation. There was no unanimous activity that everyone wanted, rather a wide variety of activities were requested. Three different categories of activity were identified which we refer to as mundane, vocational and traffic related. To demonstrate a possible IT service supporting commuters, a prototype based on speech output and a simple input mechanism from a wheel was developed. This service moves sampling of music from the conventional shop into the car. The prototype was informally tested with users, which resulted in a number of improvements. Preliminary user results indicate good functionality, a comprehensive interaction interface.

    • Objective: Platelets and granulocytes play important roles in coronary disorders. We therefore, investigated platelet and granulocyte alterations in myocardial infarctions (MIs).Patients and study design: A total of 36 individuals having MI with raised ST-segments who were receiving thrombolytic therapy were studied. Sampling was carried out after thrombolysis within 24 h after hospital admission. After 3 to 6 months of recovery, 25 patients were reinvestigated. At the infarction, peak platelet density was determined using a special designed computerised apparatus. In addition, we did counts on platelets, neutrophils and monocytes. Moreover, plasma levels of soluble P-selectin, myeloperoxidase and interleukin 6 were determined to estimate the degree of platelet, neutrophil and monocyte activation, respectively. Peak platelet density was analysed at the MI. All other parameters were determined at the acute event and at recovery.Results: At the MI, compared to the recovery, platelet counts were lower (P<.001). In addition, increased neutrophil counts (P<.001), elevated monocyte counts (P<.001), enhanced myeloperoxidase (P<.001) and interleukin 6 (P<.001) levels were demonstrated. We failed to show elevated soluble P-selectin. Compared to individuals with ST-segment elevations and low platelet density (≤1.058 kg/l), patients having peak platelet densities >1.058 kg/l displayed lower neutrophil counts (P<.01) and decreased interleukin 6 levels (P<.01). Furthermore, we demonstrate that individuals with higher inflammatory response at the MI had higher neutrophil (r=.6; P<.01) and higher monocyte counts (r=.6; P<.001) at recovery.Conclusion: We conclude that MI is associated with an inflammatory response. However, a subgroup of patients having MI with ST-elevations and low peak platelet density was identified. Compared to subjects with higher platelet density, they had more severe inflammatory characteristics. The differences persisted during recovery.

  • Betonguttorkning med byggcement

    • I denna artikel redogörs för uttorkning hos betong med Byggcement jämfört med andra cementsorter: Slite Std-cement samt anläggningscement. Inverkan av silikastoft på uttorkningen hos Slite Std-cement redovisas. För ändamålet studerades 14 betonger upp till ett halvt år. Inverkan av självuttorkning, lufthärdning och vattenlagring fanns med i programmet. Resultaten visar att uttorkningen i huvudsak följer vattencementtalet, dvs eftersom Byggcement innehåller cirka åtta procent mindre rent cement än vad Slite Std-cement gör, får man räkna med något sämre självuttorkning vid användning av Byggcement jämfört med självuttorkningen i betong med Slite Std-cement.

  • Structure for Data Management in Simulation Based Planning Activities

    • In today’s manufacturing, there is an increase in the need for fast and accurate operational production planning and scheduling. Usually, scheduling is performed several times each day, and new information,which is retrieved from the manufacturing facilities, has to be taken into consideration. Scheduling is normally performed with the aid of tools available on the market today. However, Discrete Event Simulation (DES) can address the facility’s dynamic behavior with the help of data from the facility, data including both historical and snapshot data with information about how the system currently is performing. This data needs to be structured in such a way that it can be used in the simulation model. This paper presents an approach forthis problem and examples of solutions. The main focus of the research and the paper is on the methodology and system architecture
