  • Gene Therapy with Etranacogene Dezaparvovec for Hemophilia B

    • Background: Moderate-to-severe hemophilia B is treated with lifelong, continuous coagulation factor IX replacement to prevent bleeding. Gene therapy for hemophilia B aims to establish sustained factor IX activity, thereby protecting against bleeding without burdensome factor IX replacement. Methods: In this open-label, phase 3 study, after a lead-in period (≥6 months) of factor IX prophylaxis, we administered one infusion of adeno-associated virus 5 (AAV5) vector expressing the Padua factor IX variant (etranacogene dezaparvovec; 2×1013 genome copies per kilogram of body weight) to 54 men with hemophilia B (factor IX activity ≤2% of the normal value) regardless of preexisting AAV5 neutralizing antibodies. The primary end point was the annualized bleeding rate, evaluated in a noninferiority analysis comparing the rate during months 7 through 18 after etranacogene dezaparvovec treatment with the rate during the lead-in period. Noninferiority of etranacogene dezaparvovec was defined as an upper limit of the two-sided 95% Wald confidence interval of the annualized bleeding rate ratio that was less than the noninferiority margin of 1.8. Superiority, additional efficacy measures, and safety were also assessed. Results: The annualized bleeding rate decreased from 4.19 (95% confidence interval [CI], 3.22 to 5.45) during the lead-in period to 1.51 (95% CI, 0.81 to 2.82) during months 7 through 18 after treatment, for a rate ratio of 0.36 (95% Wald CI, 0.20 to 0.64; P<0.001), demonstrating noninferiority and superiority of etranacogene dezaparvovec as compared with factor IX prophylaxis. Factor IX activity had increased from baseline by a least-squares mean of 36.2 percentage points (95% CI, 31.4 to 41.0) at 6 months and 34.3 percentage points (95% CI, 29.5 to 39.1) at 18 months after treatment, and usage of factor IX concentrate decreased by a mean of 248,825 IU per year per participant in the post-treatment period (P<0.001 for all three comparisons). Benefits and safety were observed in participants with predose AAV5 neutralizing antibody titers of less than 700. No treatment-related serious adverse events occurred. Conclusions: Etranacogene dezaparvovec gene therapy was superior to prophylactic factor IX with respect to the annualized bleeding rate, and it had a favorable safety profile.

  • X-ray diffractive imaging of highly ionized helium nanodroplets

    • Finding the lowest energy configuration of N unit charges on a sphere, known as Thomson's problem, is a long-standing query which has only been studied via numerical simulations. We present its physical realization using multiply charged He nanodroplets. The charge positions are determined by x-ray coherent diffractive imaging with Xe as a contrast agent. In neutral droplets, filaments resulting from Xe atoms condensing on quantum vortices are observed. Unique to charged droplets, however, Xe clusters that condense on charges are distributed on the surface in lattice-like structures, introducing He droplets as experimental model systems for the study of Thomson's problem.

  • Genetic loci and prioritization of genes for kidney function decline derived from a meta-analysis of 62 longitudinal genome-wide association studies

    • Estimated glomerular filtration rate (eGFR) reflects kidney function. Progressive eGFR-decline can lead to kidney failure, necessitating dialysis or transplantation. Hundreds of loci from genome-wide association studies (GWAS) for eGFR help explain population cross section variability. Since the contribution of these or other loci to eGFR-decline remains largely unknown, we derived GWAS for annual eGFR-decline and meta-analyzed 62 longitudinal studies with eGFR assessed twice over time in all 343,339 individuals and in high-risk groups. We also explored different covariate adjustment. Twelve genomewide significant independent variants for eGFR-decline unadjusted or adjusted for eGFR- baseline (11 novel, one known for this phenotype), including nine variants robustly associated across models were identified. All loci for eGFR-decline were known for cross-sectional eGFR and thus distinguished a subgroup of eGFR loci. Seven of the nine variants showed variant- by-age interaction on eGFR cross section (further about 350,000 individuals), which linked genetic associations for eGFR-decline with agedependency of genetic cross- section associations. Clinically important were two to four-fold greater genetic effects on eGFR-decline in high-risk subgroups. Five variants associated also with chronic kidney disease progression mapped to genes with functional in- silico evidence (UMOD, SPATA7, GALNTL5, TPPP). An unfavorable versus favorable nine-variant genetic profile showed increased risk odds ratios of 1.35 for kidney failure (95% confidence intervals 1.03- 1.77) and 1.27 for acute kidney injury (95% confidence intervals 1.08-1.50) in over 2000 cases each, with matched controls). Thus, we provide a large data resource, genetic loci, and prioritized genes for kidney function decline, which help inform drug development pipelines revealing important insights into the age-dependency of kidney function genetics.

  • APOGEE Chemical Abundance Patterns of the Massive Milky Way Satellites

    • The SDSS-IV Apache Point Observatory Galactic Evolution Experiment (APOGEE) survey has obtained high-resolution spectra for thousands of red giant stars distributed among the massive satellite galaxies of the Milky Way (MW): the Large and Small Magellanic Clouds (LMC/SMC), the Sagittarius Dwarf Galaxy (Sgr), Fornax (Fnx), and the now fully disrupted Gaia Sausage/Enceladus (GSE) system. We present and analyze the APOGEE chemical abundance patterns of each galaxy to draw robust conclusions about their star formation histories, by quantifying the relative abundance trends of multiple elements (C, N, O, Mg, Al, Si, Ca, Fe, Ni, and Ce), as well as by fitting chemical evolution models to the [α/Fe]-[Fe/H] abundance plane for each galaxy. Results show that the chemical signatures of the starburst in the Magellanic Clouds (MCs) observed by Nidever et al. in the α-element abundances extend to C+N, Al, and Ni, with the major burst in the SMC occurring some 3-4 Gyr before the burst in the LMC. We find that Sgr and Fnx also exhibit chemical abundance patterns suggestive of secondary star formation epochs, but these events were weaker and earlier (∼5-7 Gyr ago) than those observed in the MCs. There is no chemical evidence of a second starburst in GSE, but this galaxy shows the strongest initial star formation as compared to the other four galaxies. All dwarf galaxies had greater relative contributions of AGB stars to their enrichment than the MW. Comparing and contrasting these chemical patterns highlight the importance of galaxy environment on its chemical evolution.

  • The chemical characterization of halo substructure in the Milky Way based on APOGEE

    • Galactic haloes in a Λ-CDM universe are predicted to host today a swarm of debris resulting from cannibalized dwarf galaxies. The chemodynamical information recorded in their stellar populations helps elucidate their nature, constraining the assembly history of the Galaxy. Using data from APOGEE and Gaia, we examine the chemical properties of various halo substructures, considering elements that sample various nucleosynthetic pathways. The systems studied are Heracles, Gaia-Enceladus/Sausage (GES), the Helmi stream, Sequoia, Thamnos, Aleph, LMS-1, Arjuna, I’itoi, Nyx, Icarus, and Pontus. Abundance patterns of all substructures are cross-compared in a statistically robust fashion. Our main findings include: (i) the chemical properties of most substructures studied match qualitatively those of dwarf Milky Way satellites, such as the Sagittarius dSph. Exceptions are Nyx and Aleph, which are chemically similar to disc stars, implying that these substructures were likely formed in situ; (ii) Heracles differs chemically from in situ populations such as Aurora and its inner halo counterparts in a statistically significant way. The differences suggest that the star formation rate was lower in Heracles than in the early Milky Way; (iii) the chemistry of Arjuna, LMS-1, and I’itoi is indistinguishable from that of GES, suggesting a possible common origin; (iv) all three Sequoia samples studied are qualitatively similar. However, only two of those samples present chemistry that is consistent with GES in a statistically significant fashion; (v) the abundance patterns of the Helmi stream and Thamnos are different from all other halo substructures.

  • Investigating the Dominant Environmental Quenching Process in UVCANDELS/COSMOS Groups

    • We explore how the fraction of quenched galaxies changes in groups of galaxies with respect to the distance to the center of the group, redshift, and stellar mass to determine the dominant process of environmental quenching in 0.2 < z < 0.8 groups. We use new UV data from the UVCANDELS project in addition to existing multiband photometry to derive new galaxy physical properties of the group galaxies from the zCOSMOS 20 k group catalog. Limiting our analysis to a complete sample of log (M*/M⊙) > 10.56 group galaxies, we find that the probability of being quenched increases slowly with decreasing redshift, diverging from the stagnant field galaxy population. A corresponding analysis on how the probability of being quenched increases with time within groups suggests that the dominant environmental quenching process is characterized by slow (∼Gyr) timescales. We find a quenching time of approximately  Gyr, consistent with the slow processes of strangulation and delayed-then-rapid quenching although more data are needed to confirm this result.

  • A Spatially Resolved Analysis of Star Formation Burstiness by Comparing UV and Hα in Galaxies at z ∼ 1 with UVCANDELS

    • The UltraViolet imaging of the Cosmic Assembly Near-infrared Deep Extragalactic Legacy Survey Fields (UVCANDELS) program provides Hubble Space Telescope (HST)/UVIS F275W imaging for four CANDELS fields. We combine this UV imaging with existing HST/near-IR grism spectroscopy from 3D-HST+AGHAST to directly compare the resolved rest-frame UV and Hα emission for a sample of 979 galaxies at 0.7 < z < 1.5, spanning a range in stellar mass of 108−11.5M⊙. Using a stacking analysis, we perform a resolved comparison between homogenized maps of rest-UV and Hα to compute the average UV-to-Hα luminosity ratio (an indicator of burstiness in star formation) as a function of galactocentric radius. We find that galaxies below stellar mass of ∼109.5M⊙, at all radii, have a UV-to-Hα ratio higher than the equilibrium value expected from constant star formation, indicating a significant contribution from bursty star formation. Even for galaxies with stellar mass ≳109.5M⊙, the UV-to-Hα ratio is elevated toward their outskirts (R/Reff > 1.5), suggesting that bursty star formation is likely prevalent in the outskirts of even the most massive galaxies, but is likely overshadowed by their brighter cores. Furthermore, we present the UV-to-Hα ratio as a function of galaxy surface brightness, a proxy for stellar mass surface density, and find that regions below ∼107.5M⊙ kpc−2 are consistent with bursty star formation, regardless of their galaxy stellar mass, potentially suggesting that local star formation is independent of global galaxy properties at the smallest scales. Last, we find galaxies at z > 1.1 to have bursty star formation, regardless of radius or surface brightness.

  • The Lazy Giants : APOGEE Abundances Reveal Low Star Formation Efficiencies in the Magellanic Clouds

    • We report the first APOGEE metallicities and alpha-element abundances measured for 3600 red giant stars spanning a large radial range of both the Large (LMC) and Small Magellanic Clouds, the largest Milky Way (MW) dwarf galaxies. Our sample is an order of magnitude larger than that of previous studies and extends to much larger radial distances. These are the first results presented that make use of the newly installed southern APOGEE instrument on the du Pont telescope at Las Campanas Observatory. Our unbiased sample of the LMC spans a large range in metallicity, from [Fe/H] = -0.2 to very metal-poor stars with [Fe/H] -2.5, the most metal-poor Magellanic Cloud (MC) stars detected to date. The LMC [alpha/Fe]-[Fe/H] distribution is very flat over a large metallicity range but rises by similar to 0.1 dex at -1.0 < [Fe/H] less than or similar to -0.5. We interpret this as a sign of the known recent increase in MC star formation activity and are able to reproduce the pattern with a chemical evolution model that includes a recent "starburst." At the metal-poor end, we capture the increase of [alpha/Fe] with decreasing [Fe/H] and constrain the "alpha-knee" to [Fe/H] less than or similar to -2.2 in both MCs, implying a low star formation efficiency of similar to 0.01 Gyr(-1). The MC knees are more metal-poor than those of less massive MW dwarf galaxies such as Fornax, Sculptor, or Sagittarius. One possible interpretation is that the MCs formed in a lower-density environment than the MW, a hypothesis that is consistent with the paradigm that the MCs fell into the MW's gravitational potential only recently.

  • The Seventeenth Data Release of the Sloan Digital Sky Surveys : Complete Release of MaNGA, MaStar, and APOGEE-2 Data

    • This paper documents the seventeenth data release (DR17) from the Sloan Digital Sky Surveys; the fifth and final release from the fourth phase (SDSS-IV). DR17 contains the complete release of the Mapping Nearby Galaxies at Apache Point Observatory (MaNGA) survey, which reached its goal of surveying over 10,000 nearby galaxies. The complete release of the MaNGA Stellar Library accompanies this data, providing observations of almost 30,000 stars through the MaNGA instrument during bright time. DR17 also contains the complete release of the Apache Point Observatory Galactic Evolution Experiment 2 survey that publicly releases infrared spectra of over 650,000 stars. The main sample from the Extended Baryon Oscillation Spectroscopic Survey (eBOSS), as well as the subsurvey Time Domain Spectroscopic Survey data were fully released in DR16. New single-fiber optical spectroscopy released in DR17 is from the SPectroscipic IDentification of ERosita Survey subsurvey and the eBOSS-RM program. Along with the primary data sets, DR17 includes 25 new or updated value-added catalogs. This paper concludes the release of SDSS-IV survey data. SDSS continues into its fifth phase with observations already underway for the Milky Way Mapper, Local Volume Mapper, and Black Hole Mapper surveys.
