    • We synthesised results on soil carbon (C) and nitrogen (N) fluxes and the accumulation of soil organic C and N under adjacent 50-year-old Norway spruce, Scots pine and silver birch stands growing on similar soils and evaluated the different processes involved. C and N budgets were calculated. Spruce stands had larger stocks of C and N in biomass and soil than birch stands, with pine intermediate. The differences in soil stocks were mainly found in the organic layer, whereas differences in the mineral soil were small. The study showed that there is no simple answer to what is causing the differences in soil C and N stocks, because several processes are interacting. Spruce and pine trees had higher biomass and litter production than birch trees, but total litter inputs showed no significant difference between stands, because the rich ground vegetation under pine and birch contributed with substantial litter inputs, in contrast to the poor ground vegetation under spruce. Decomposition rate (per g of C) was markedly higher under birch than under spruce and pine resulting in lower C and N stocks in the organic layer. This effect was amplified by higher abundance and biomass of earthworms, favoured by higher pH and palatable litter under birch. Earthworm bioturbation probably both increased decomposition rate and damaged the ectomycorrhizal network with negative consequences for the formation of mycorrhizal litter and C storage. In conclusion, the direct effects of spruce, pine and birch litter on C and N pools and fluxes were modified by indirect effects of understorey structure, pH and earthworm responses.     • Soil properties were compared in adjacent 50-year-old Norway spruce, Scots pine and silver birch stands 26 growing on similar soils in south-west Sweden. The effects of tree species were most apparent in the 27 humus layer and decreased with soil depth. At 20–30 cm depth in the mineral soil, species differences 28 in soil properties were small and mostly not significant. Soil C, N, K, Ca, Mg, and Na content, pH, base sat- 29 uration and fine root biomass all significantly differed between humus layers of different species. Since 30 the climate, parent material, land use history and soil type were similar, the differences can be ascribed to 31 tree species. Spruce stands had the largest amounts of carbon stored down to 30 cm depth in mineral soil 32 (7.3 kg C m??2), whereas birch stands, with the lowest production, smallest amount of litterfall and lowest 33 C:N ratio in litter and humus, had the smallest carbon pool (4.1 kg C m??2), with pine intermediate 34 (4.9 kg C m??2). Similarly, soil nitrogen pools amounted to 349, 269, and 240 g N m??2 for spruce, pine, 35 and birch stands, respectively.   • Towards the development of a bioengineered uterus : Comparison of different protocols for rat uterus decellularization

    • Uterus transplantation (UTx) may be the only possible curative treatment for absolute uterine factor infertility, which affects 1 in every 500 females of fertile age. We recently presented the 6-month results from the first clinical UTx trial, describing nine live-donor procedures. This routine involves complicated surgery and requires potentially harmful immune suppression to prevent rejection. However, tissue engineering applications using biomaterials and stem cells may replace the need for a live donor, and could prevent the required immunosuppressive treatment. To investigate the basic aspects of this, we developed a novel whole-uterus scaffold design for uterus tissue engineering experiments in the rat. Decellularization was achieved by perfusion of detergents and ionic solutions. The remaining matrix and its biochemical and mechanical properties were quantitatively compared from using three different protocols. The constructs were further compared with native uterus tissue composition. Perfusion with Triton X-100/dimethyl sulfoxide/H2O led to a compact, weaker scaffold that showed evidence of a compromised matrix organization. Sodium deoxycholate/dH2O perfusion gave rise to a porous scaffold that structurally and mechanically resembled native uterus better. An innovative combination of two proteomic analyses revealed higher fibronectin and versican content in these porous scaffolds, which may explain the improved scaffold organization. Together with other important protocol-dependent differences, our results can contribute to the development of improved decellularization protocols for assorted organs. Furthermore, our study shows the first available data on decellularized whole uterus, and creates new opportunities for numerous in vitro and in vivo whole-uterus tissue engineering applications.

    • Clinical management guidelines on malignant disorders are generally based on data from clinical trials with selected patient cohorts. In Sweden, more than 95% of all patients diagnosed with chronic myeloid leukemia (CML) are reported to the national CML registry, providing unique possibilities to compile population-based information. This report is based on registry data from 2002 to 2010, when a total of 779 patients (425 men, 354 women; median age, 60 years) were diagnosed with CML (93% chronic, 5% accelerated, and 2% blastic phase) corresponding to an annual incidence of 0.9/100 000. In 2002, approximately half of the patients received a tyrosine kinase inhibitor as initial therapy, a proportion that increased to 94% for younger (andlt;70 years) and 79% for older (andgt;80 years) patients during 2007-2009. With a median follow-up of 61 months, the relative survival at 5 years was close to 1.0 for patients younger than 60 years and 0.9 for those aged 60 to 80 years, but only 0.6 for those older than 80 years. At 12 months, 3% had progressed to accelerated or blastic phase. Sokal, but not European Treatment and Outcome Study, high-risk scores were significantly linked to inferior overall and relative survival. Patients living in university vs nonuniversity catchment areas more often received tyrosine kinase inhibitors up front but showed comparable survival.

    • BACKGROUND: Infiltrating T-helper cells, cytotoxic T-cells, B-cells and monocytes are thought to mediate the damage to myelin, oligodendrocytes and axons in multiple sclerosis (MS), which results in progressive disability. OBJECTIVE: The objective of this paper is to explore gene expression profiles of leukocytes in the cerebrospinal fluid (CSF) compartment of MS patients during relapse. METHODS: Global gene expression was analyzed by DNA microarray analysis of cells in CSF from MS patients and controls, and verifications were performed with real-time polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay (ELISA). RESULTS: Fifty percent of the recently described risk genes for MS and 28% of non-risk genes were differently expressed in MS patients compared to controls (χ(2)-test, p=7.7 × 10(-5)). Genes involved in T- and NK-cell processes were up-regulated, and genes involved in processes targeting innate immunity or B-cells were down-regulated in MS. Increased expression of EDN1 and CXCL11 and decreased expression of HMOX1 was verified with real-time PCR and increased expression of CXCL13 was verified with ELISA in CSF. CONCLUSION: DNA microarray analysis is useful in identifying differently expressed genes in CSF leukocytes, which may be important in MS in vivo. Our findings suggest that many of the risk genes for MS are differently expressed in the disease-mediating leukocytes that penetrate the blood-brain barrier.

  • The Scent of Disease : Human Body Odor Contains an Early Chemosensory Cue of Sickness

    • Observational studies have suggested that with time, some diseases result in a characteristic odor emanating from different sources on the body of a sick individual. Evolutionarily, however, it would be more advantageous if the innate immune response were detectable by healthy individuals as a first line of defense against infection by various pathogens, to optimize avoidance of contagion. We activated the innate immune system in healthy individuals by injecting them with endotoxin (lipopolysaccharide). Within just a few hours, endotoxin-exposed individuals had a more aversive body odor relative to when they were exposed to a placebo. Moreover, this effect was statistically mediated by the individuals' level of immune activation. This chemosensory detection of the early innate immune response in humans represents the first experimental evidence that disease smells and supports the notion of a "behavioral immune response" that protects healthy individuals from sick ones by altering patterns of interpersonal contact.

    • The present study describes the use of reactive UV-absorbers (UVAs) that can be covalently attached to a wood substrate to obtain an improved exterior durability. It has previously been demonstrated that the photostability of wood is increased if the UVA is grafted to the wood surface. The grafting of an epoxide-functional UVA, 2-hydroxy-4(2,3-epoxypropoxy)-benzophenone (HEPBP), to thin veneer wood specimens had a larger photostabilizing effect than if the corresponding UVA without the reactive handle was added to the wood coating. The present study shows that not only the chemical structure of the UVA is important but also how the modification is performed i.e. the reaction procedure. A successful attachment of the UVA depends on both the reaction conditions as well as if other reactive species are present at the same time. The presence of for example a vegetable oil (epoxy functionalized soy bean oil) affects the grafting reaction of the UVA. Details on the effect of reaction conditions, reaction sequences, and presence of co-reactants will be presented.
