  • Variations in soil characteristics affecting the occurrence of Aphanomyces root rot of sugar beet - Risk evaluation and disease control

    • Damping off and chronic root rot of sugar beet caused by Aphanomyces cochlioides is a major constraint in cultivation of sugar beet, with occurrence of the disease in Sweden being concentrated to specific areas. This study examined soil factors that can be used for risk assessment of Aphanomyces root rot. Soils from 134 field plots were assessed over three years for Aphanomyces root rot potential in bioassays and analysed for easily measured soil factors such as soluble nutrients, pH and soil electrical conductivity (EC). Classification of the field plots into four groups with increasing disease severity index (DSI) according to the bioassay revealed that the group with the lowest DSI (<39) had an average soil calcium (Ca) content of 430 mg/100 g and a soil EC of 1.12 mS/cm, which were significantly higher than in the groups with DSI >40. From these results, we concluded that soil Ca concentration is an easily measured factor that can be used to identify soils with an increased risk of Aphanomyces root rot. We suggest that the Ca content should be above 250 mg Ca/100 g soil to avoid problems with Aphanomyces root rot in sugar beet. To gain a more thorough understanding of the geographical variation in Aphanomyces root rot and its connection to the geological origin of the soils, a number of other soil factors were analysed in the field plots, including clay mineralogy, CEC, and particle size distribution. Aphanomyces root rot was very rare in soils with a high proportion of smectite and vermiculite relative to illite and kaolin minerals, here predominantly calcareous soils developed on clay till in south-western Scania.     • Irrigation water is an important vehicle for dissemination of human pathogens to plants. As contamination in an early stage of the production chain cannot necessarily be counteracted later, cultural measures to reduce the contamination risk need to be adopted during primary production. In a two-factorial greenhouse experiment, we studied the impact of inoculum density and the interval between irrigation and haivest on the prevalence of an inoculated gfp-tagged non-pathogenic strain of Escherichia coli O157:H7. The strain was inoculated with the irrigation water at a density of log 5.6, log 6.6 and log 7.6 CFU ml(-1) into the phyllosphere of fully grown crops of rocket and spinach (BBCH 49). The crops were then harvested after 3, 24,48 and 72 h. The introduced strain decreased exponentially in numbers within 72 h, to 49.6%, 52.6% and 50.6%, respectively, in the spinach and to 58.5%, 67.4% and 73.4% in the rocket. No differences were found in the number of the total viable count of aerobic bacteria and of Enterobacteriaceae as assessed on tryptic soy agar (TSA) and violet red bile dextrose agar (VRBD), respectively. Sequencing of the 16S rRNA genes of randomly selected isolates from VRBD were identified as Enterobacter cloaceae, Enterobacter ludwigii, Pantoea sp. and Raoultella planticola as the dominant Enterobacteriaceae species in the rocket and spinach phyllosphere. We found that cessation of irrigation for three days seems not to be an adequate sanitisation treatment to exclude the possibility of viable E. coil 0157:H7 cells on spinach or rocket.   • Green tea powder and Lactobacillus plantarum affect gut microbiota, lipid metabolism and inflammation in high-fat fed C57BL/6J mice

    • Background: Type 2 diabetes is associated with obesity, ectopic lipid accumulation and low-grade inflammation. A dysfunctional gut microbiota has been suggested to participate in the pathogenesis of the disease. Green tea is rich in polyphenols and has previously been shown to exert beneficial metabolic effects. Lactobacillus plantarum has the ability to metabolize phenolic acids. The health promoting effect of whole green tea powder as a prebiotic compound has not been thoroughly investigated previously. Methods: C57BL/6J mice were fed a high-fat diet with or without a supplement of 4% green tea powder (GT), and offered drinking water supplemented with Lactobacillus plantarum DSM 15313 (Lp) or the combination of both (Lp + GT) for 22 weeks. Parameters related to obesity, glucose tolerance, lipid metabolism, hepatic steatosis and inflammation were examined. Small intestinal tissue and caecal content were collected for bacterial analysis. Results: Mice in the Lp + GT group had significantly more Lactobacillus and higher diversity of bacteria in the intestine compared to both mice in the control and the GT group. Green tea strongly reduced the body fat content and hepatic triacylglycerol and cholesterol accumulation. The reduction was negatively correlated to the amount of Akkermansia and/or the total amount of bacteria in the small intestine. Markers of inflammation were reduced in the Lp + GT group compared to control. PLS analysis of correlations between the microbiota and the metabolic variables of the individual mice showed that relatively few components of the microbiota had high impact on the correlation model. Conclusions: Green tea powder in combination with a single strain of Lactobacillus plantarum was able to promote growth of Lactobacillus in the intestine and to attenuate high fat diet-induced inflammation. In addition, a component of the microbiota, Akkermansia, correlated negatively with several metabolic parameters known to be risk factors for the development of type 2 diabetes.

    • Abstract Background: Lactobacillus species are the most often found inhabitants of vaginal ecosystem of fertile women. In postmenopausal women with low oestrogen levels, Lactobacillus flora is diminishing or absent. However, no studies have been performed to investigate the correlation between oestrogen levels and the lactobacilli in the gut. The aim of the present study was to investigate the relation in healthy women between vaginal and rectal microbial flora as well as possible variations with hormone levels. Methods: Vaginal and rectal smears were taken from 20 healthy fertile women, average 40 years (range 28-49 years), in two different phases of the menstrual cycle, and from 20 postmenopausal women, average 60 years (range 52-85 years). Serum sex hormone levels were analyzed. Bacteria from the smears isolated on Rogosa Agar were grouped by Randomly Amplified Polymorphic DNA and identified by multiplex PCR and partial 16S rRNA gene sequencing. Results: Lactobacillus crispatus was more often found in the vaginal flora of fertile women than in that of postmenopausal (p = 0.036). Fifteen of 20 fertile women had lactobacilli in their rectal smears compared to 10 postmenopausal women (p = 0.071). There was no correlation between the number of bacteria in vagina and rectum, or between the number of bacteria and hormonal levels. Neither could any association between the presence of rectal lactobacilli and hormonal levels be found. Conclusion: Lactobacillus crispatus was more prevalent in the vaginal flora of fertile women, whereas the Lactobacillus flora of rectum did not correlate to the vaginal flora nor to hormonal levels.

    • Living with fibromyalgia (FM) means living with a chronic pain condition that greatly influences daily life. The majority of people with FM are middle-aged women. The aim of this study was to elucidate meanings of pain for women with FM. Fifteen women with FM were interviewed about their pain experiences and a phenomenological hermeneutic interpretation was used to analyse the interview texts. The findings show that meanings of pain for women with FM can be understood as living with a double burden; living with an aggressive, unpredictable pain and being doubted by others in relation to the invisible pain. The ever-present pain was described as unbearable, overwhelming, and dominated the women's whole existence. Nevertheless, all the women tried to normalize life by doing daily chores in an attempt to alleviate the pain. In order to support the women's needs and help them to feel well despite their pain, it is important that nurses and health care personnel acknowledge and understand women with FM and their pain experiences. © 2011 P.     • Fibromyalgia (FM) is a common chronic pain syndrome that mostly affects middle-aged women. Our aim with this study was to elucidate meanings of being received and met by others as experienced by women with FM. Interviews with a narrative approach were conducted with 9 women. We analyzed the transcribed interviews with a phenomenological hermeneutical interpretation. The findings revealed two themes: being seen as a malingerer and being acknowledged. Meanings of being received and met by others, as experienced by women with FM, can be understood as a movement between the two perspectives. When they were acknowledged, their feelings of security and trust increased, but the women could not rely on this because others received and met them in such an unpredictable manner. © The Author(s) 2014.

    • The researchers focus in this study was to elucidate meanings of feeling well for women with fibromyalgia (FM). We obtained narrative interviews with thirteen women with FM and used a phenomenological-hermeneutic interpretation to analyse the interview texts. Our interpretation of the findings show that for women with FM meanings of feeling well can be understood as having strength to be involved. The women's experiences of feeling well meant being in control, having power, finding one's own pace and feelings of belonging.

    • Purpose: Living with fibromyalgia (FM) means living with a long-term pain syndrome that is invisible to others. Support and understanding from others seem to be important to managing the affected daily life. The aim of this study was to describe the significance of FM associations for women with FM. Methods: Data collection was carried out through focus group discussions with seventeen women with FM. Data were analyzed through thematic content analysis. Results: The findings show that women experienced associations for people with FM as important as they gave access to contacts with others with similar experiences. Their need of togetherness was fulfilled at the association and they described being strengthened by the support received. Because of the lack of information and knowledge about FM, the association was described as an important venue for getting and mediating information about the illness. Conclusions: At the association the women seem to be empowered, which increases their ability to manage their daily lives despite the limitations imposed by FM. Healthcare personnel could not satisfy the women’s needs and to manage to support women with FM.     • BACKGROUND: Probiotics may exert beneficial effects in the gastrointestinal tract. This randomized trial investigated the effect of the probiotic Lactobacillus plantarum 299v on the intestinal load of potentially pathogenic bacteria, bacterial translocation, and cell proliferation in elective colon surgery. METHODS: Seventy-five patients were randomized to pre- and postoperative oral intake of Lactobacillus plantarum 299v or placebo. Rectal swabs and mucosal biopsies were taken before the start of intake, after 1 week, at surgery, and after 6 days, weeks, and months. Viable counts were quantified for clostridia, Enterobacteriaceae, Gram-negative anaerobes, and lactobacilli. Bacterial translocation was determined by the analysis of bacterial DNA genes in mesenteric lymph nodes. Ki-67 was used as a marker of cell proliferation in normal mucosa and tumor. RESULTS: Lactobacillus plantarum 299v was given without adverse effects. Lactobacillus plantarum 299v as well as Enterobacteriaceae and Gram-negative anaerobes increased in the colon 1 week after the administration of Lactobacillus plantarum 299v. There were no significant differences between patients receiving Lactobacillus plantarum 299v and placebo in the incidence of bacterial translocation (27 vs. 13 %) and postoperative complications (16 vs. 31 %). CONCLUSIONS: Lactobacillus plantarum 299v was established in the intestine, but no inhibitory effect on enteric bacteria, bacterial translocation, or postoperative complications was found. The mechanism behind the protective effects of probiotics found in animal and some human studies remain elusive and require further explorations. No adverse effects were recorded after the administration of high doses of Lactobacillus plantarum 299v.
