  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • (Re)Planning of green infrastructure and nature-based solutions for sustainable urban transition

    • Landscape approaches are important for planning of urban sprawl in peri-urban landscapes, continuously emerging in many metropolitan regions. In the case of Stockholm Region, land-take and incremental urbanisation is a continouous process, while the regional development plan has ambitions to steer the overall development in a sustainable direction. This plan contains a green infrastructure effort building on a set of green wedges, mainly serving as support to the needs of the city and suburbs and their citizens. This initiative differ from the later green infrastructure action plan provided by the county administrative board, related to the EU biodiversity strategy and guidelines. The latter has a different approach, mainly targeting biodiversity goals as well as ecosystem services. These approaches differ from each other in several ways while both have unclear roles when it comes to municipal planning on different levels. Furthermore, the municipalities have their own initiatives when it comes to green infrastructure and nature-based solutions and it is not clear how the different planning tiers are linked to each other, to planning and management, and to multifunctional landscapes. The aim of the REPLAN project is to investigate how the different green infrastructure initiatives are linked to planning, to each other on different scales, and whether they can serve multi-functional landscapes when it comes to biodiversity and different ecosystem services. The REPLAN project involves stakeholders and practitioners on different planning levels for co-producing knowledge, methods and strategies for green infrastructure and nature-based solutions to serve as tools for sustainable transition of metropolitan areas and their peri-urban landscapes.

  • Organo-ptii complexes for potent photodynamic inactivation of multi-drug resistant bacteria and the influence of configuration

    • PtII based organometallic photosensitizers (PSs) have emerged as novel potent photodynamic inactivation (PDI) reagents through their enhanced intersystem crossing (ISC) processes. Currently, few PtII PSs have been investigated as antibacterial materials, with relatively poor performances reported and with structure-activity relationships not well described. Herein, a pair of configurational isomers are reported of Bis-BODIPY (4,4-difluoro-boradizaindacene) embedded PtII PSs. The cis-isomer (cis-BBP) displayed enhanced 1O2 generation and better bacterial membrane anchoring capability as compared to the trans-isomer (trans-BBP). The effective PDI concentrations (efficiency > 99.9%) for cis-BBP in Acinetobacter baumannii (multi-drug resistant (MDR)) and Staphylococcus aureus are 400 nM (12 J cm−2) and 100 nM (18 J cm−2), respectively; corresponding concentrations and light doses for trans-BBP in the two bacteria are 2.50 µM (30 J cm−2) and 1.50 µM (18 J cm−2), respectively. The 50% and 90% minimum inhibitory concentration (MIC50 and MIC90) ratio of trans-BBP to cis-BBP is 22.22 and 24.02 in A. baumannii (MDR); 21.29 and 22.36 in methicillin resistant S. aureus (MRSA), respectively. Furthermore, cis-BBP displays superior in vivo antibacterial performance, with acceptable dark and photoinduced cytotoxicity. These results demonstrate cis-BBP is a robust light-assisted antibacterial reagent at sub-micromolecular concentrations. More importantly, configuration of PtII PSs should be an important issue to be considered in further PDI reagents design.

    • With the publication of the European Green Deal, the European Union has committed to reaching carbon neutrality by 2050. The envisaged reductions of direct greenhouse gases emissions are seen as technically feasible, but if a wrong path is pursued, significant unintended impacts across borders, sectors, societies and ecosystems may follow. Without the insights gained from an impact assessment framework reaching beyond the techno-economic perspective, the pursuit of direct emission reductions may lead to counterproductive outcomes in the long run. We discuss the opportunities and challenges related to the creation and use of an integrated assessment framework built to inform the European Commission on the path to decarbonisation. The framework is peculiar in that it goes beyond existing ones in its scope, depth and cross-scale coverage, by use of numerous specialised models and case studies. We find challenges of consistency that can be overcome by linking modelling tools iteratively in some cases, harmonising modelling assumptions in others, comparing model outputs in others. We find the highest added value of the framework in additional insights it provides on the technical feasibility of decarbonisation pathways, on vulnerability aspects and on unintended environmental and health impacts on national and sub-national scale.

  • Bioinspired Co-Design of Tactile Sensor and Deep Learning Algorithm for Human-Robot Interaction

    • Robots equipped with bionic skins for enhancing the robot perception capability are increasingly deployed in wide applications ranging from healthcare to industry. Artificial intelligence algorithms that can provide bionic skins with efficient signal processing functions further accelerate the development of this trend. Inspired by the somatosensory processing hierarchy of humans, the bioinspired co-design of a tactile sensor and a deep learning-based algorithm is proposed herein, simplifying the sensor structure while providing computation-enhanced tactile sensing performance. The soft piezoresistive sensor, based on the carbon black-coated polyurethane sponge, offers a continuous sensing area. By utilizing a customized deep neural network (DNN), it can detect external tactile stimulus spatially continuously. Besides, a novel data augmentation method is developed based on the sensor's hexagonal structure that has a sixfold rotation symmetry. It can significantly enhance the generalization ability of the DNN model by enriching the collected training data with generated pseudo-data. The functionality of the sensor and the robustness of the proposed data augmentation strategy are verified by precisely recognizing five touch modalities, illustrating a well-generalized performance, and providing a promising application prospect in human-robot interaction.

    • Multiple Human-Robot Collaboration (HRC) requires self-organising task allocation to adapt to varying operation goals and workspace changes. However, nowadays an HRC system relies on predefined task arrangements for human and robot agents, which fails to accomplish complicated manufacturing tasks consisting of various operation sequences and different mechanical parts. To overcome the bottleneck, this paper proposes a temporal subgraph reasoning-based method for self-organising HRC task planning between multiple agents. Firstly, a tri-layer Knowledge Graph (KG) is defined to depict task-agent-operation relations in HRC tasks. Then, a subgraph mechanism is introduced to learn node embeddings from subregions of the HRC KG, which distills implicit information from local object sets. Thirdly, a temporal reasoning module is leveraged to integrate features from previous records and update the HRC KG for forecasting humans' and robots' subsequent operations. Finally, a car engine assembly task is demonstrated to evaluate the effectiveness of the proposed method, which outperforms other benchmarks in experimental results.

    • The demand for timber, bioenergy feedstock and other forest products, leading to intensified forest harvesting, is expected to increase in the coming decades. A reduction in the delivery of forest ecosystem services and, specifically, biodiversity, including the provision of habitats for mature-forest-dwelling raptors, has been attributed to the intensification of forest exploitation. Therefore, in order to adopt adequate conservation measures to create a timely buffer against the consequences of increased harvesting, it is critically important to understand how the availability of nesting habitats for protected species will fluctuate in the future landscape. In this study, using the LandSim tool, we modelled the dynamics of the forests and nesting habitat availability for the forest-nesting raptor, the lesser spotted eagle Clanga pomarina, for the next 50 years in eastern Lithuania, Central Europe. Our findings indicate that the share of forests available for final harvesting is expected to increase rapidly in the coming decades due to a large amount of forest stands reaching a mature age, if current forest management practices, despite them being considered as relatively conservative, are continued. As a consequence, the availability of nesting habitats will constantly decrease in nesting territories, as well as elsewhere in the landscape, in the coming decades. We suggest that species conservation strategies should not only incorporate directly targeted measures to protect nest sites from destruction and disturbance, but also, at the very least, preserve a sufficient amount of nesting habitats in areas inhabited by eagle pairs and, at best, at the landscape scale.

  • Sustainability assessment of intensified forestry - Forest bioenergy pathways versus forest biodiversity targeting forest birds

    • Intensified forestry can be seen as a solution to climate change mitigation and securing energy supply, increasing the production of forest bioenergy feedstock as a substitution for fossil fuels. However, it may come with detrimental impacts on forest biodiversity, especially related to older forests. The aim of this study was to assess the sustainability of intensified forestry from climate-energy and biodiversity perspectives, targeting forest bird species. For this purpose, we applied the Landscape simulation and Ecological Assessment (LEcA) tool to the study area of Lithuania, having high ambitions for renewables and high forest biodiversity. With LEcA, we simulated forest growth and management for 100 years with two forest management strategies: Business As Usual (BAU) and Intensive forestry (INT), the latter with the purpose to fulfil renewable energy goals. With both strategies, the biomass yields increased well above the yields of the reference year, while the biodiversity indicators related to forest bird habitat to different degrees show the opposite, with lower levels than for the reference year. Furthermore, Strategy INT resulted in small-to-no benefits in the long run concerning potential biomass harvesting, while substantially affecting the biodiversity indicators negatively. The model results have the potential to inform policy and forest management planning concerning several sustainability goals simultaneously.

  • Dual-Dimensional Manufacturing Service Collaboration Optimization Toward Industrial Internet Platforms

    • An Industrial Internet platform is acknowledged to be a requisite promoter for smart manufacturing, enabling physical manufacturing resources to be virtualized and permitting resources to collaborate in the form of services. As a central function of the platform, manufacturing service collaboration optimization is dedicated to establishing high-quality service collaboration solutions for manufacturing tasks. Such optimization is inseparable from the functional and amount requirements of a task, which must be satisfied when orchestrating services. However, existing manufacturing service collaboration optimization methods mainly focus on horizontal collaboration among services for functional demands and rarely consider vertical collaboration to cover the needed amounts. To address this gap, this paper proposes a dual-dimensional service collaboration methodology that combines functional and amount collaboration. First, a multi-granularity manufacturing service modeling method is presented to describe services. On this basis, a dual-dimensional manufacturing service collaboration optimization (DMSCO) model is formulated. In the vertical dimension, multiple functionally equivalent services form a service cluster to fulfill a subtask; in the horizontal dimension, complementary service clusters collaborate for the entire task. Service selection and amount distribution to the selected services are critical issues in the model. To solve the problem, a multi-objective memetic algorithm with multiple local search operators is tailored. The algorithm embeds a competition mechanism to dynamically adjust the selection probabilities of the local search operators. The experimental results demonstrate the superiority of the algorithm in terms of convergence, solution quality, and comprehensive metrics, in comparison with commonly used algorithms.

  • Theoretical Study of the Catalytic Mechanism of the Cu-Only Superoxide Dismutase

    • The catalytic mechanisms for the wild-type and the mutated Cu-only superoxide dismutase were studied using the hybrid density functional B3LYP and a quantum chemical cluster approach. Optimal protonation states of the active site were examined for each stage of the catalytic cycle. For both the reductive and the oxidative half-reactions, the arrival of the substrate O-2(center dot-) was found to be accompanied by a chargecompensating H+ with exergonicities of -15.4 kcal center dot mol and -4.7 kcal center dot mol, respectively. The second-sphere Glu-110 and first-sphere His-93 were suggested to be the transient protonation site for the reductive and the oxidative half-reactions, respectively, which collaborates with the hydrogen bonding water chain to position the substrate near the redox-active copper center. For the reductive half-reaction, the rate-limiting step was found to be the inner-sphere electron transfer from the partially coordinated O-2(center dot-) to Cu-II with a barrier of 8.1 kcal center dot mol. The formed O-2 is released from the active site with an exergonicity of -14.9 kcal center dot mol. For the oxidative half-reaction, the inner-sphere electron transfer from CuI to the partially coordinated O-2(center dot-) was found to be accompanied by the proton transfer from the protonated His-93 and barrierless. The rate-limiting step was found to be the second proton transfer from the protonated Glu-110 to HO2 with a barrier of 7.3 kcal center dot mol. The barriers are reasonably consistent with experimental activities, and a proton-transfer rate-limiting step in the oxidative half-reaction could explain the experimentally observed pH-dependence. For the E110Q CuSOD, Asp-113 was suggested to be likely to serve as the transient protonation site in the reductive half-reaction. The rate-limiting barriers were found to be 8.0 and 8.6 kcal center dot mol, respectively, which could explain the slightly lower performance of E110X mutants. The results were found to be stable, with respect to the percentage of exact exchange in B3LYP.
