  • Phenome-wide association analysis of LDL-cholesterol lowering genetic variants in PCSK9

    • Background: We characterised the phenotypic consequence of genetic variation at the PCSK9 locus and compared findings with recent trials of pharmacological inhibitors of PCSK9. Methods: Published and individual participant level data (300,000+ participants) were combined to construct a weighted PCSK9 gene-centric score (GS). Seventeen randomized placebo controlled PCSK9 inhibitor trials were included, providing data on 79,578 participants. Results were scaled to a one mmol/L lower LDL-C concentration. Results: The PCSK9 GS (comprising 4 SNPs) associations with plasma lipid and apolipoprotein levels were consistent in direction with treatment effects. The GS odds ratio (OR) for myocardial infarction (MI) was 0.53 (95% CI 0.42; 0.68), compared to a PCSK9 inhibitor effect of 0.90 (95% CI 0.86; 0.93). For ischemic stroke ORs were 0.84 (95% CI 0.57; 1.22) for the GS, compared to 0.85 (95% CI 0.78; 0.93) in the drug trials. ORs with type 2 diabetes mellitus (T2DM) were 1.29 (95% CI 1.11; 1.50) for the GS, as compared to 1.00 (95% CI 0.96; 1.04) for incident T2DM in PCSK9 inhibitor trials. No genetic associations were observed for cancer, heart failure, atrial fibrillation, chronic obstructive pulmonary disease, or Alzheimer's disease - outcomes for which large-scale trial data were unavailable. Conclusions: Genetic variation at the PCSK9 locus recapitulates the effects of therapeutic inhibition of PCSK9 on major blood lipid fractions and MI. While indicating an increased risk of T2DM, no other possible safety concerns were shown; although precision was moderate.

    • This paper proposes a distributed continuous-time epidemic model, called networked SIWS (Susceptible-Infected-Water-Susceptible) model, for an SIS type waterborne disease spreading over a network of multiple groups of individuals sharing a water source. A sufficient condition is obtained for the healthy state, at which all individuals are not infected and the water is not contaminated, to be globally asymptotically stable. The effects of the shared water source on the disease spreading are analyzed through the comparison of the basic reproduction number with the networked SIS model without water and demonstrated via simulations.

  • Modeling and Stability of Prosumer Heat Networks

    • The energy sector is going through a large transformation due to public demands of renewable energy sources. However, a major issue is that these energy sources are intermittent. If designed correctly, district heating systems can naturally contain energy storing units, for example by storing heat in the isolated pipes that make up the heat grid. Additionally, this makes it easier to reuse and transport already generated heat to other users. This paper proposes a mathematical model of such a grid, where excess energy can be retracted from one user and distributed to other users using a network of heat pumps. In some cases, one can balance residual heat production with the heat consumption, temporarily eliminating the need for a centralized heating plant. Existence conditions for stable steady states of such a network with general topology are given. Finally, energy optimal stable steady states are obtained through convex optimization.

    • This paper studies a distributed continuous-time bi-virus model in which two competing viruses spread over a network consisting of multiple groups of individuals. Limiting behaviors of the network are characterized by analyzing the equilibria of the system and their stability. Specifically, when the two viruses spread over possibly different directed infection graphs, the system may have the following: first, a unique equilibrium, the healthy state, which is globally stable, implying that both viruses will eventually be eradicated, second, two equilibria including the healthy state and a dominant virus state, which is almost globally stable, implying that one virus will pervade the entire network causing a single-virus epidemic while the other virus will be eradicated, or third, at least three equilibria including the healthy state and two dominant virus states, depending on certain conditions on the healing and infection rates. When the two viruses spread over the same directed infection graph, the system may have zero or infinitely many coexisting epidemic equilibria, which represents the pervasion of the two viruses. Sensitivity properties of some nontrivial equilibria are investigated in the context of a decentralized control technique, and an impossibility result is given for a certain type of distributed feedback controller.

    • We show that two widely accepted model reduction techniques, balanced truncation (BT) and balanced singular perturbation approximation (BSPA), can be derived as limiting approximations of a carefully constructed parameterization of linear time invariant systems by employing the model boundary approximation method (MBAM) [1]. We also show that MBAM provides a novel way to interpolate between BT and BSPA, by exploring the set of approximations on the boundary of the "model manifold," which is associated with the specific choice of model parameterization and initial condition and is embedded in a sample space of measured outputs, between the elements that correspond to the two model reduction techniques. This paper suggests similar types of approximations may be obtainable in topologically similar places (i.e., on certain boundaries) on the associated model manifold of nonlinear systems if analogous parameterizations can be achieved, therefore extending these widely accepted model reduction techniques to nonlinear systems.(1)

    • This paper proposes a layered networked SIWS (Susceptible-Infected-Water-Susceptible) model, for an SIS-type waterborne disease spreading over a human contact network connected to a water distribution network that has a pathogen spreading in it. Conditions for local and global stability of the healthy state, where no one is sick and the water network is not contaminated, are provided. We also pose an observability problem, and show under certain conditions if you observe some of the human contact network you can recover the pathogen levels in the water network.

    • This paper proposes a continuous-time virus spreading model that includes a human-contact network and a water component that may be contaminated. The main motivating applications are modeling infectious waterborne diseases such as cholera, SARS, and amoebiasis (hand-to-mouth). We present the model and its derivation, explore the equilibria of the model, and analyze the healthy equilibria. We illustrate the behavior of the model via simulation, and demonstrate how the proposed model captures the behavior of Dr. John Snow's pioneering cholera dataset.

  • Networked Model for Cooperative Adaptive Cruise Control

    • This paper proposes a cooperative adaptive cruise control model, adding a communication network structure to an existing model that has been shown to capture real commercial adaptive cruise control vehicle behavior. The proposed model is interesting because it only requires minimal information sharing, facilitating the creation of platoons comprised of vehicles from different manufacturers. We prove the stability of the model and discuss string stability. Algorithms for estimating the velocity of the vehicles locally and for estimating the velocities of all the vehicles in the platoon are presented. We simulate vehicle platoon control with the lead vehicle following an experimentally collected trajectory, showing that adding communication can cause a string unstable platoon to become stable.
