  • Testing a Home Solution for Preparing Young Children for an Awake MRI : A Promising Smartphone Application

    • Thanks to its non-invasive nature and high-resolution imaging capabilities, magnetic resonance imaging (MRI) is a valuable diagnostic tool for pediatric patients. However, the fear and anxiety experienced by young children during MRI scans often result in suboptimal image quality and the need for sedation/anesthesia. This study aimed to evaluate the effect of a smartphone application called COSMO@home to prepare children for MRI scans to reduce the need for sedation or general anesthesia. The COSMO@home app was developed incorporating mini-games and an engaging storyline to prepare children for learning goals related to the MRI procedure. A multicenter study was conducted involving four hospitals in Belgium. Eligible children aged 4–10 years were prepared with the COSMO@home app at home. Baseline, pre-scan, and post-scan questionnaires measured anxiety evolution in two age groups (4–6 years and 7–10 years). Eighty-two children participated in the study, with 95% obtaining high-quality MRI images. The app was well-received by children and parents, with minimal technical difficulties reported. In the 4–6-year-old group (N = 33), there was a significant difference between baseline and pre-scan parent-reported anxiety scores, indicating an increase in anxiety levels prior to the scan. In the 7–10-year-old group (N = 49), no significant differences were observed between baseline and pre-scan parent-reported anxiety scores. Overall, the COSMO@home app proved to be useful in preparing children for MRI scans, with high satisfaction rates and successful image outcomes across different hospitals. The app, combined with minimal face-to-face guidance on the day of the scan, showed the potential to replace or assist traditional face-to-face training methods. This innovative approach has the potential to reduce the need for sedation or general anesthesia during pediatric MRI scans and its associated risks and improve patient experience.

  • Locating place, crime and the fear of crime : methodological and theoretical considerations

    • Much previous research on the fear of crime has focused on why some individuals, with certain characteristics, experience more or less fear of crime than others. However, there is also a growing body of research examining the role that the neighbourhood context in which individuals reside plays in shaping such feelings and perceptions. At the same time, less research has been directed at understanding why certain small-scale micro-places evoke feelings of unsafety and fear of crime.The aim of this dissertation is to contribute to improving the current state ofthe research focused on place, the fear of crime, and related methodological issues. The dissertation includes four original empirical research papers. Study I is based on a case study evaluating the impact of camera surveillance and examines what role the operationalization of place may play for the results and interpretation of a given study.The findings show that different operationalizations may indeed produce different results, and that the choice of operationalization must be carefully considered in the context of study design. Study II uses responses to an open-ended survey question from three waves of the Malmö Community Survey (2012, 2015, 2018) to chart the spatial concentration and temporal stability of unsafe locations.The findings show that locations perceived as unsafe by city inhabitants are concentrated to a very small proportion of the urban space, and that there is a temporal stability in unsafe locations over time. Study III further explores unsafe locations by examining the spatial risk factors associated with these unsafe locations and the role played by neighbourhood collective efficacy and disorder. The results show that a number of spatial risk factors are correlated with the outcome, suggesting that the physical environment has a role to play in shaping people’s perceptions of unsafety at a given location. The findings also show that there are major between neighbourhood variations in unsafe locations, but that neighbourhood collective efficacy and disorder play only a limited role in the explanation of this variance. The final paper, Study IV, is a methodological study focused on the feasibility of using an alternative approach to studying fear of crime, as a momentary event, and uses an experience research framework implemented using a smart phone application (STUNDA). The general conclusion is that it is feasible to conduct research on the fear of crime using a smartphone application, but that emerging methods may also involve new methodological issues and challenges.The four studies have both methodological and theoretical implications, suggesting that the way place is defined and operationalized may have important impacts on the results and interpretations of research studies. In addition, the findings suggest that there is more to be learned about the fear of crime as a context-specific experience that is dependent on the immediate environment, and that alternative methodological approaches focused on surveying momentary experiences of fear of crime using smartphone applications seem to be feasible. A place-based approach to the fear of crime, supported by alternative measures and methods, may also be important in developing a broader understanding of how perceptions of fear of crime and unsafety are shaped. Such an understanding may in turn assist policymakers and practitioners to design knowledge-based interventions to reduce fear of crime and feelings of unsafety.

  • Positioning aquatic animals with acoustic transmitters

    • Geolocating aquatic animals with acoustic tags has been ongoing for decades, relying on the detection of acoustic signals at multiple receivers with known positions to calculate a 2D or 3D position, and ultimately recreate the path of an aquatic animal from detections at fixed stations.This method of underwater geolocation is evolving with new software and hardware options available to help investigators design studies and calculate positions using solvers based predominantly on time-difference-of-arrival and time-of-arrival.We provide an overview of the considerations necessary to implement positioning in aquatic acoustic telemetry studies, including how to design arrays of receivers, test performance, synchronize receiver clocks and calculate positions from the detection data. We additionally present some common positioning algorithms, including both the free open-source solvers and the ‘black-box’ methods provided by some manufacturers for calculating positions.This paper is the first to provide a comprehensive overview of methods and considerations for designing and implementing better positioning studies that will support users, and encourage further knowledge advances in aquatic systems.

  • Profiling of Extracellular Small RNAs Highlights a Strong Bias towards Non-Vesicular Secretion

    • The extracellular environment consists of a plethora of molecules, including extracellular miRNA that can be secreted in association with extracellular vesicles (EVs) or soluble protein complexes (non-EVs). Yet, interest in therapeutic short RNA carriers lies mainly in EVs, the vehicles conveying the great majority of the biological activity. Here, by overexpressing miRNA and shRNA sequences in parent cells and using size exclusion liquid chromatography (SEC) to separate the secretome into EV and non-EV fractions, we saw that >98% of overexpressed miRNA was secreted within the non-EV fraction. Furthermore, small RNA sequencing studies of native miRNA transcripts revealed that although the abundance of miRNAs in EVs, non-EVs and parent cells correlated well (R-2 = 0.69-0.87), quantitatively an outstanding 96.2-99.9% of total miRNA was secreted in the non-EV fraction. Nevertheless, though EVs contained only a fraction of secreted miRNAs, these molecules were stable at 37 degrees C in a serum-containing environment, indicating that if sufficient miRNA loading is achieved, EVs can remain delivery-competent for a prolonged period of time. This study suggests that the passive endogenous EV loading strategy might be a relatively wasteful way of loading miRNA to EVs, and active miRNA loading approaches are needed for developing advanced EV miRNA therapies in the future.
