  • Unraveling the importance of solid and adsorbed phase mercury speciation for methylmercury formation, evasion and bioaccumulation

    • Monomethylmercury, MeHg, is formed under anoxic conditions in waters, sediments and soils and then bioaccumulated and biomagnified in aquatic food webs, negatively effecting both human and wildlife health. It is generally accepted that precipitation of mercury, Hg, and adsorption of Hg to e.g. organic matter and mineral surfaces are important processes limiting the reactivity of Hg mobilized in the environment by natural and anthropogenic activities. However, knowledge concerning the role of different solid and adsorbed chemical forms of Hg for MeHg formation, evasion and bioaccumulation is missing. Such information is vital for the understanding of environmental processes controlling MeHg formation and bioaccumulation, as well as for predicting how changes in e.g. loading rates of atmospheric Hg and the outcome of climate change scenarios and anthropogenic land use could alter Hg concentrations in biota.In this thesis, a novel experimental approach, using isotopically enriched solid and adsorbed phases of inorganic Hg, HgII, as tracers, was developed. Using this approach, we successfully determined rates of MeHg formation from solid and adsorbed Hg species in sediment slurries and in mesocosm systems under conditions closely resembling those in field. We conclude that the solid/adsorbed phase speciation of HgII is a major controlling factor for MeHg net formation rates. Microcosm experiments revealed that newly formed MeHg was a major contributor to the evasion of MeHg from the water‒sediment system, emphasizing the importance of MeHg formation rate, rather than MeHg concentration, in the sediment for this process. From mesocosm systems, we provide experimental evidence, as well as quantitate data, for that terrestrial and atmospheric sources of HgII and MeHg are more available for methylation and bioaccumulation processes than HgII and MeHg stored and formed in sediments. This suggests that the contribution from terrestrial and atmospheric sources to the accumulation of Hg in fish may have been underestimated. As a consequence, in regions where climate change is expected to further increase land runoff, terrestrial MeHg sources may have even higher negative effects on biota than previously thought. Data and concepts presented in this thesis lay the basis for unprecedented in-depth modeling of processes in the Hg biogeochemical cycle that will improve our understanding and the predicting power on how aquatic ecosystems may respond to environmental changes or differences in loading rates for atmospheric Hg.

    • Growing InGaN quantum dots (QDs) at the apex of hexagonal GaN pyramids is an elegant approach to achieve a deterministic positioning of QDs. Despite similar synthesis procedures by metal–organic chemical vapor deposition, the optical properties of the QDs reported in the literature vary drastically. The QDs tend to exhibit either narrow or broad emission lines in the micro-photoluminescence spectra. By coupled microstructural and optical investigations, the QDs giving rise to narrow emission lines were concluded to nucleate in association with a (0001) facet at the apex of the GaN pyramid.

    • In the cities of La Paz and El Alto, 573 tons of organic material are disposed in landfills every day. These residues can be used to produce biogas and recycle nutrients, thus alleviating environmental impacts related to waste management. Technical solutions are evaluated through a multicriteria analysis with the purpose of defining a strategy for implementing waste-to-biogas in the two cities. As a result, the development for waste-to-biogas-system is defined in three steps. Step 1 consists of an active extraction system of landfill gas in the already existing landfills. Step 2 implies the establishment of a dry-digestion biogas facility based on present waste collection practices, that is, not segregated waste. Step 3 consists of a biogas plant using dry digestion for processing source segregated bio-waste. The economic feasibility of these three steps is evaluated. Despite prevailing fossil fuels subsidies in the country, implementing waste-to-biogas turn out feasible in the country provided the digestate is commercialized as bio-fertilizer or erosion control material and additional services such as waste collection and deposition are computed in the total economy of the biogas production plant.

    • PURPOSE:According to the American Association of Physicists in Medicine Task Group No. 43 (TG-43) formalism used for dose calculation in brachytherapy treatment planning systems, the absolute level of absorbed dose is determined through coupling with the measurable quantity air-kerma strength or the numerically equal reference air-kerma rate (RAKR). Traceability to established standards is important for accurate dosimetry in laying the ground for reliable comparisons of results and safety in adoption of new treatment protocols. The purpose of this work was to compare the source strength for a high-dose rate (HDR) (192)Ir source as measured using equipment traceable to different standard laboratories in Europe and the United States.METHODS AND MATERIALS:Source strength was determined for one HDR (192)Ir source using four independent systems, all with traceability to different primary or interim standards in the United States and Europe.RESULTS:The measured HDR (192)Ir source strengths varied by 0.8% and differed on average from the vendor value by 0.3%. Measurements with the well chambers were 0.5% ± 0.1% higher than the vendor-provided source strength. Measurements with the Farmer chamber were 0.7% lower than the average well chamber results and 0.2% lower than the vendor-provided source strength. All of these results were less than the reported source calibration uncertainties (k=2) of each measurement system.CONCLUSIONS:In view of the uncertainties in ion chamber calibration factors, the maximum difference in source strength found in this study is small and confirms the consistency between calibration standards in use for HDR (192)Ir brachytherapy.

  • The Effect of Baseline Testosterone on the Efficacy of Degarelix and Leuprolide: Further Insights From A 12-Month, Comparative, Phase III Study in Prostate Cancer Patients

    • OBJECTIVE To investigate the effects of baseline testosterone on testosterone control and prostate-specific antigen (PSA) suppression using data from a phase III trial (CS21) comparing degarelix and leuprolide in prostate cancer. METHODS In CS21, patients with histologically confirmed prostate cancer (all stages) were randomized to degarelix 240 mg for 1 month followed by monthly maintenance doses of 80 or 160 mg, or leuprolide 7.5 mg/month. Patients receiving leuprolide could receive antiandrogens for flare protection. Treatment effects on testosterone and PSA reduction, testosterone surge, and microsurges were investigated in 3 baseline testosterone subgroups: <3.5, 3.5-5.0, and >5.0 ng/mL. Data are presented for the groups receiving degarelix 240/80 mg (the approved dose) and leuprolide 7.5 mg. RESULTS Higher baseline testosterone delayed castration with both treatments. However, castrate testosterone levels and PSA suppression occurred more rapidly with degarelix irrespective of baseline testosterone. With leuprolide, the magnitude of testosterone surge and microsurges increased with increasing baseline testosterone. There was no overall correlation between baseline testosterone and initial PSA decrease in either treatment group, although PSA suppression tended to be slowest with leuprolide and fastest with degarelix in the high baseline testosterone subgroup. CONCLUSION Patients with high baseline testosterone may have greater risk of tumor stimulation (clinical flare) and mini-flares during gonadotrophin-releasing hormone agonist treatment and so the need for flare protection with antiandrogens in these patients is obvious, especially in metastatic disease. Although higher baseline testosterone delays castration, castrate testosterone and PSA suppression occur more rapidly with degarelix, irrespective of baseline testosterone, without the need for flare protection.
