  • Genetic studies of body mass index yield new insights for obesity biology.

    • Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10(-8)), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20% of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

  • Is Unbleached Cotton Better Than Bleached? Exploring the Limits of Life-Cycle Assessment in the Textile Sector

    • The applicability of life-cycle assessment (LCA) for the textile industry is discussed with a special focus on environmental impact from chemicals. Together with issues of water depletion and energy use, the use of chemicals and their emissions are important environmental considerations for textile products. However, accounting for chemicals is a weak point in LCA methodology and practice. Two research questions were investigated in a case study of hospital garments: 1) whether LCA adds value to assessments of the chemical performance of textile products, and 2) whether inclusion of toxicity issues in LCA affects environmental performance rankings for textile products. It is concluded that the quantitative and holistic tool LCA is useful for environmental decision makers in the textile industry, and becomes more effective when chemical impacts are included. A flexible way forward is demonstrated to meet the challenge of accounting for chemicals in LCAs of textile products.

  • A century of trends in adult human height

    • Being taller is associated with enhanced longevity, and higher education and earnings. We reanalysed 1472 population-based studies, with measurement of height on more than 18.6 million participants to estimate mean height for people born between 1896 and 1996 in 200 countries. The largest gain in adult height over the past century has occurred in South Korean women and Iranian men, who became 20.2 cm (95% credible interval 17.522.7) and 16.5 cm (13.319.7) taller, respectively. In contrast, there was little change in adult height in some sub-Saharan African countries and in South Asia over the century of analysis. The tallest people over these 100 years are men born in the Netherlands in the last quarter of 20th century, whose average heights surpassed 182.5 cm, and the shortest were women born in Guatemala in 1896 (140.3 cm; 135.8144.8). The height differential between the tallest and shortest populations was 19-20 cm a century ago, and has remained the same for women and increased for men a century later despite substantial changes in the ranking of countries.

  • Which criteria are used to clear patients to return to sport after primary ACL reconstruction? A scoping review

    • Objective To describe the criteria used to clear athletes to return to sport (RTS) following primary ACL reconstruction. Design Scoping review. Data sources MEDLINE, Embase, CINAHL and SPORTDiscus electronic databases were searched using keywords related to ACL and RTS. Eligibility criteria Prospective or retrospective studies reporting at least one RTS criterion for athletes who had primary ACL reconstruction with an autograft. Results In total, 209 studies fulfilled the inclusion criteria. RTS criteria were categorised into six domains: time, strength, hop testing, clinical examination, patient-report and performance-based criteria. From the 209 included studies, time was used in 178 studies (85%), and in 88 studies (42%) was the sole RTS criterion. Strength tests were reported in 86 studies (41%). Sixteen different hop tests were used in 31 studies (15%). Clinical examination was used in 54 studies (26%), patient report in 26 studies (12%) and performance-based criteria in 41 studies (20%). Summary Time and impairment-based measures dominated RTS criteria, despite sport being a complex physical and biopsychosocial activity with demands across all aspects of function. Time was included as a criterion in 85% of studies, and over 80% of studies allowed RTS before 9 months. Whether RTS tests are valid-do they predict successful RTS?-is largely unknown.

Hypothesizing that genetic effects might be stronger for early- versus late-onset stroke, we conducted a 2-stage meta-analysis of genome-wide association studies, focusing on stroke cases with an age of onset <60 years.

    • Polymorphisms in coagulation genes have been associated with early-onset ischemic stroke. Here we pursue an a priori hypothesis that genetic variation in the endothelial-based receptors of the thrombomodulin-protein C system (THBD and PROCR) may similarly be associated with early-onset ischemic stroke. We explored this hypothesis utilizing a multi-stage design of discovery and replication.Discovery was performed in the Genetics-of-Early-Onset Stroke (GEOS) Study, a biracial population-based case-control study of ischemic stroke among men and women aged 15-49 including 829 cases of first ischemic stroke (42.2% African-American) and 850 age-comparable stroke-free controls (38.1% African-American). Twenty-four single-nucleotide-polymorphisms (SNPs) in THBD and 22 SNPs in PROCR were evaluated. Following LD pruning (r2≥0.8), we advanced uncorrelated SNPs forward for association analyses. Associated SNPs were evaluated for replication in an early-onset ischemic stroke population (onset-age<60 years) consisting of 3676 cases and 21118 non-stroke controls from 6 case-control studies. Lastly, we determined if the replicated SNPs also associated with older-onset ischemic stroke in the METASTROKE data-base.Among GEOS Caucasians, PROCR rs9574, which was in strong LD with 8 other SNPs, and one additional independent SNP rs2069951, were significantly associated with ischemic stroke (rs9574, OR = 1.33, p = 0.003; rs2069951, OR = 1.80, p = 0.006) using an additive-model adjusting for age, gender and population-structure. Adjusting for risk factors did not change the associations; however, associations were strengthened among those without risk factors. PROCR rs9574 also associated with early-onset ischemic stroke in the replication sample (OR = 1.08, p = 0.015), but not older-onset stroke. There were no PROCR associations in African-Americans, nor were there any THBD associations in either ethnicity.PROCR polymorphisms are associated with early-onset ischemic stroke in Caucasians.

  • Perception matters! Engagement in task orientated social robotics

    • Engagement in task orientated social robotics is a complex phenomenon, consisting of both task and social elements. Previous work in this area tends to focus on these aspects in isolation without consideration for the positive or negative effects one might cause the other. We explore both, in an attempt to understand how engagement with the task might effect the social relationship with the robot, and vice versa. In this paper, we describe the analysis of participant self-report data collected during an exploratory pilot study used to evaluate users’ “perception of engagement”. We discuss how the results of our analysis suggest that ultimately, it was the users’ own perception of the robots’ characteristics such as friendliness, helpfulness and attentiveness which led to sustained engagement with both the task and robot

    • Archaeological evidence indicates that pig domestication had begun by similar to 10,500 y before the present ( BP) in the Near East, and mitochondrial DNA ( mtDNA) suggests that pigs arrived in Europe alongside farmers similar to 8,500 y BP. A few thousand years after the introduction of Near Eastern pigs into Europe, however, their characteristic mtDNA signature disappeared and was replaced by haplotypes associated with European wild boars. This turnover could be accounted for by substantial gene flow from local European wild boars, although it is also possible that European wild boars were domesticated independently without any genetic contribution from the Near East. To test these hypotheses, we obtained mtDNA sequences from 2,099 modern and ancient pig samples and 63 nuclear ancient genomes from Near Eastern and European pigs. Our analyses revealed that European domestic pigs dating from 7,100 to 6,000 y BP possessed both Near Eastern and European nuclear ancestry, while later pigs possessed no more than 4% Near Eastern ancestry, indicating that gene flow from European wild boars resulted in a near-complete disappearance of Near East ancestry. In addition, we demonstrate that a variant at a locus encoding black coat color likely originated in the Near East and persisted in European pigs. Altogether, our results indicate that while pigs were not independently domesticated in Europe, the vast majority of human-mediated selection over the past 5,000 y focused on the genomic fraction derived from the European wild boars, and not on the fraction that was selected by early Neolithic farmers over the first 2,500 y of the domestication process.

  • Choosing a rain coating – life cycle assessment to guide substitution of hazardous durable water repellent chemistry

    • Many actors in the textile industry are currently substituting hazardous long-chain per- and polyfluoroalkyl substances (PFAS). One application of PFAS in the textile industry is in finishing to impart water (and oil) resistance to the textile (chemicals that provide this function are called durable water repellents; DWR). In a review of properties of the DWRs that are alternatives to DWRs containing long-chain PFASs, we could show that several of the alternatives also have hazardous properties and that a substitution of the DWR may lead to a shift in the environmental burden of the garment, e.g. by the need of more frequent wash or shorter life length. It is clear that to achieve an environmentally sound substitution thorough assessment of the alternatives is needed. The project Substitution in Practice of Prioritized Fluorinated Chemicals to Eliminate Diffuse Sources (SUPFES, www.supfes.eu, funded by the Swedish Research Council for Environment, Agricultural Sciences and Spatial Planning (FORMAS) under grant agreement No. 2012-2148) set out to guide this substitution by assessment of the DWR alternatives, both with regards to their functionality and the environmental consequences of their use. In life cycle assessments (LCA) the environmental performance of four different types of garments (two types of shell jackets, an ambulance jacket and a children’s overall), meeting different user protection needs, are investigated. The LCAs are intended to make possible a holistic assessment where toxicological hazards as well as risks of problem shifting (e.g. between impact categories) are addressed. Connected with technical performance testing of the DWR alternatives the LCAs allow for a unique possibility to include also functionality aspects. One important prerequisite for the LCAs is the availability of characterisation factors for (eco)toxicological effects and a review of existing characterisation factors (CF) for textile chemicals showed that CFs are missing for many DWR-related substances. The calculation of new CFs with UseTox 2.01 made it clear that several of the DWR-related substances are particularly difficult to model due to physical-chemical properties affecting their fate. In addition, many of the substances are persistent, bioaccumulative and toxic (PBT) or very persistent and very mobile (vPvM) and as such their environmental consequences may not be well captured by existing life cycle impact assessment (LCIA) methods, if for example relevant accumulation compartments are not included in the models. In this contribution we will report on our results from the SUPFES project’s screening LCAs on the four garments and these results’ implications for the DWR substitution case. Which DWR alternative will provide sufficient functionality and environmental soundness, making possible a substitution lasting in the long run? We will also use these cases as basis for the discussion about LCIA methods and practices suitable in substitution cases involving PBT or vPvM substances in general, and PFAS in particular.
