The draft genome of the microscopic Nemertoderma westbladi sheds light on the evolution of Acoelomorpha genomes

- Background: Xenacoelomorpha is a marine clade of microscopic worms that is an important model system for understanding the evolution of key bilaterian novelties, such as the excretory system. Nevertheless, Xenacoelomorpha genomics has been restricted to a few species that either can be cultured in the lab or are centimetres long. Thus far, no genomes are available for Nemertodermatida, one of the group’s main clades and whose origin has been dated more than 400 million years ago.Methods: DNA was extracted from a single specimen and sequenced with HiFi following the PacBio Ultra-Low DNA Input protocol. After genome assembly, decontamination, and annotation, the genome quality was benchmarked using two acoel genomes and one Illumina genome as reference. The gene content of three cnidarians, three acoelomorphs, four deuterostomes, and eight protostomes was clustered in orthogroups to make inferences of gene content evolution. Finally, we focused on the genes related to the ultrafiltration excretory system to compare patterns of presence/absence and gene architecture among these clades.Results: We present the first nemertodermatid genome sequenced from a single specimen of Nemertoderma westbladi. Although genome contiguity remains challenging (N50: 60 kb), it is very complete (BUSCO: 80.2%, Metazoa; 88.6%, Eukaryota) and the quality of the annotation allows fine-detail analyses of genome evolution. Acoelomorph genomes seem to be relatively conserved in terms of the percentage of repeats, number of genes, number of exons per gene and intron size. In addition, a high fraction of genes present in both protostomes and deuterostomes are absent in Acoelomorpha. Interestingly, we show that all genes related to the excretory system are present in Xenacoelomorpha except Osr, a key element in the development of these organs and whose acquisition seems to be interconnected with the origin of the specialised excretory system.Conclusion: Overall, these analyses highlight the potential of the Ultra-Low Input DNA protocol and HiFi to generate high-quality genomes from single animals, even for relatively large genomes, making it a feasible option for sequencing challenging taxa, which will be an exciting resource for comparative genomics analyses.

- The microstructure response of laser-powder bed fusion (L-PBF)-processed ferritic stainless steel (AISI 441) during post-heat treatments is studied in detail. Focus is on the precipitation kinetics of the Nb-rich phases: Laves (Fe2Nb) and the cubic carbo-nitride (NbC), as well as the grain structure evolution. The evolution of the precipitates is characterized using scanning and transmission electron microscopy (SEM and TEM) and the experimental results are used to calibrate precipitation kinetics simulations using the precipitation module (TC-PRISMA) within the Thermo-Calc Software package. The calculations reproduce the main trend for both the mean radii for the Laves phase and the NbC, and the amount of Laves phase, as a function of temperature. The calibrated model can be used to optimize the post-heat treatment of additively manufactured ferritic stainless steel components and offer a creator tool for process and structure linkages in an integrated computational materials engineering (ICME) framework for alloy and process development of additively manufactured ferritic steels.

Low-grade intestinal inflammation two decades after pelvic radiotherapy.

- Radiotherapy is effective in the treatment of cancer but also causes damage to non-cancerous tissue. Pelvic radiotherapy may produce chronic and debilitating bowel symptoms, yet the underlying pathophysiology is still undefined. Most notably, although pelvic radiotherapy causes an acute intestinal inflammation there is no consensus on whether the late-phase pathophysiology contains an inflammatory component or not. To address this knowledge gap, we examined the potential presence of a chronic inflammation in mucosal biopsies from irradiated pelvic cancer survivors.We biopsied 24 cancer survivors two to 20 years after pelvic radiotherapy, and four non-irradiated controls. Using tandem mass tag (TMT) mass spectrometry and mRNA sequencing (mRNA-seq), we charted proteomic and transcriptomic profiles of the mucosal tissue previously exposed to a high or a low/no dose of radiation. Changes in the immune cell populations were determined with flow cytometry. The integrity of the protective mucus layers were determined by permeability analysis and 16S rRNA bacterial detection.942 proteins were differentially expressed in mucosa previously exposed to a high radiation dose compared to a low radiation dose. The data suggested a chronic low-grade inflammation with neutrophil activity, which was confirmed by mRNA-seq and flow cytometry and further supported by findings of a weakened mucus barrier with bacterial infiltration.Our results challenge the idea that pelvic radiotherapy causes an acute intestinal inflammation that either heals or turns fibrotic without progression to chronic inflammation. - Does the in-between exist? And if so, does it have any significance for our wellbeing? This article takes on the vital process of recovery from work, and we argue that to understand that process there is a great potential in what we define as spaces in-between. The type of stressors we are exposed to in working life, or due to conflicts between work and other roles in life, mostly have the character of everyday often recurring strain and irritation. This form of low-intense stress can lead to mental deterioration. When resources are consumed we need time to reboot and recreate energy. We need time for recovery, which ought to take place somewhere. Depending on organizing processes in our working life, these “whereabouts” are more or less accessible. We argue that the way space is controlled, managed, experienced and constructed affects its potential of being a place for recovery. Through these avenues and with the help of a fictional story, we show how an increase in surveillance, control and transparency rationalize and streamlines the spaces in-between “turning them” in to formal and corporate space. However, we also uncover the potential in the space in-between and illustrate when these spaces are successfully constructed and made use of as recovery from work. This potential is associated with the process of re-constructing these particular spaces to humanized meaningful places.

- Pandemin satte i gång en diskussion om arbetsplatsens betydelse och sådde även ett frö om eventuella alternativ, där rummet skulle omförhandlas, där flexibilitet och frihetsgrader skulle öka. Var kunde arbetet utföras? Var borde arbetet utföras? Är det på ”plats” eller ”from anywhere”? Under denna period var inte sällan samtalets fokus på hur de ”mänskliga resurserna” påverkades av digitalisering och det distala arbetsutförandet. Både avseende den psykosociala arbetsmiljön, den nödvändiga kompetensutvecklingen samt förutsättningar för prestation. En framträdande funktion i organisationer för att hantera dessa frågor är HRM/HRD. Den tolkning och analys som HR-staben gjorde av arbetets organisering under pandemin kommer förmodligen ha betydande inflytande på en framtida riktning för arbetets utförande och organisering. Det vill säga det faktiska innehållet av ”det nya normala”. Det är således ett sådant perspektiv som föreliggande artikel har för avsikt att fördjupa. Syftet med artikeln är att kasta ljus över hur HRM förstod och berättade om arbetets utförande och organisering under pandemin. Utsagorna tolkas genom en konstruktionistisk lins och teorier om rummets och mötets betydelse. Teorier vars hjälp får analysen att gå bortom pandemin, och ta höjd för ett framtida arbetsliv. Det empiriska bidraget i denna studie baseras på sju intervjuer med HR-direktörer på större svenska företag. Deltagarnas utsagor kretsade kring en balansgång och ambivalent hållning till organisationens olika rum, hur dessa rum görs och om rummen eventuellt kan eller inte kan styras. Deltagarnas utsagor diskuteras utifrån begreppen organisationens legitima sida och skuggsida, och vad dessa sidor representerar. En svårhanterlig spänning visade sig vara den mellan kontroll och rummets potentiella kraft, något som visade sig ha en inverterad relation. Där ökad kontroll minskade den potentiella kraften och vice versa. Vi avslutar artikeln med att diskutera ett framtida arbetsliv utifrån ett HRM-perspektiv och temana: mellanrummens betydelse, kreativitet och innovation samt ”strategy of the void”.
