  • Addressing model complexity in automotive system development : Selection of system model elements for allocation of requirements

    • Modern automotive embedded systems are developed by Original Equipment Manufacturers (OEM) together with multiple suppliers. A key problem for a supplier is to allocate an OEM’s requirements specification to their own subsystem design. This is a difficult manual task especially on complex systems and it requires expert knowledge about the system design. To address this problem, this paper presents a design science research to develop and evaluate a Requirements Allocation Assistant tool (RAA). The tool provides functionality to search through and filter requirements and system models to enable efficient requirements allocation even in the presence of complexity. RAA is built on top of the EATOP/Eclipse framework using EAST-ADL as system modelling language. The tool was evaluated and validated during a qualitative usability study with 17 engineers active in the Swedish automotive industry. Key findings are that searching is used to learn about a system, whereas filtering is used to narrow down a set of candidate elements of the system design. Engineers request further support in narrowing down a set of candidate elements and in checking that an allocation is correct.

  • Ketogenic diet attenuates hepatopathy in mouse model of respiratory chain complex III deficiency caused by a Bcs1l mutation

    • Mitochondrial disorders are among the most prevalent inborn errors of metabolism but largely lack treatments and have poor outcomes. High-fat, low-carbohydrate ketogenic diets (KDs) have shown beneficial effects in mouse models of mitochondrial myopathies, with induction of mitochondrial biogenesis as the suggested main mechanism. We fed KD to mice with respiratory chain complex III (CIII) deficiency and progressive hepatopathy due to mutated BCS1L, a CIII assembly factor. The mutant mice became persistently ketotic and tolerated the KD for up to 11 weeks. Liver disease progression was attenuated by KD as shown by delayed fibrosis, reduced cell death, inhibition of hepatic progenitor cell response and stellate cell activation, and normalization of liver enzyme activities. Despite no clear signs of increased mitochondrial biogenesis in the liver, CIII assembly and activity were improved and mitochondrial morphology in hepatocytes normalized. Induction of hepatic glutathione transferase genes and elevated total glutathione level were normalized by KD. Histological findings and transcriptome changes indicated modulation of liver macrophage populations by the mutation and the diet. These results reveal a striking beneficial hepatic response to KD in mice with mitochondrial hepatopathy and warrant further investigations of dietary modification in the management of these conditions in patients.

  • Techno-economic analysis of integrating first and second-generation ethanol production using filamentous fungi : An industrial case study

    • The 2nd generation plants producing ethanol from lignocelluloses demand risky and high investment costs. This paper presents the energy- and economical evaluations for integrating lignocellulose in current 1st generation dry mill ethanol processes, using filamentous fungi. Dry mills use grains and have mills, liquefactions, saccharifications, fermentation, and distillation to produce ethanol, while their stillage passes centrifugation, and evaporation to recycle the water and dry the cake and evaporated syrup into animal feed. In this work, a bioreactor was considered to cultivate fungi on the stillage either before or after the centrifugation step together with pretreated lignocellulosic wheat bran. The results showed that the integrated 1st and 2nd generation ethanol process requires a capital investment of 77 million USD, which could yield NPV of 162 million USD after 20 years. Compared to the fungal cultivation on thin stillage modified 1st generation process, the integrated process resulted in 53 million USD higher NPV. The energy analysis showed that the thin stillage modified 1st generation process could reduce the overall energy consumption by 2.5% and increase the ethanol production by 4%. Such modifications in the 1st generation processes and integration concepts could be interesting for the ethanol industries, as integrating lignocelluloses to their existing setup requires less capital investment.
