    • Many populations of brown trout are managed by hatchery breeding and supportive release. Using a limited number of individuals as founders creates a bottleneck, which can lead to loss of genetic diversity in a population. In this study 17 populations of hatchery-reared brown trout were examined genetically using microsatellite markers. Genetic variation measured as gene diversity and allelic richness were analysed and the effects of effective population size of founders and time since founding on these parameters were explored. Allelic richness and gene diversity decreased with increasing time since founding of the stock, and there was a negative relationship between current effective population size and time since founding. Allelic richness was positively correlated with effective population size at founding. The results indicate that considerations concerning effective population size in hatcheries must be taken seriously to promote high levels of genetic variation among individuals and minimise loss of genetic diversity.

  • Creation and Detection of Single Photons

    • A growing number of technologies employ quantum properties in order to produce solutions that surpass the performance of conventional devices, or to execute operations that are fundamentally impossible with classical systems alone. In the field of optical quantum information science, photons are utilized to encode, communicate and manipulate information, making them vitally important. While photon production always constitutes the first step in any optical experiment, in the field of quantum information science, the recording of data through the process of photon detection is an equally crucial final step.This thesis deals with both the single photons generation (based on diamond color defects) and their detection, utilizing a novel type of superconducting detectors. In particular, part one of this thesis is devoted to the construction of custom designed microscope setup, and the development of laboratory experiments, to enable the generation of single photons as well as the investigation of the optical and spin properties of diamond color centers. Confocal microscopy is used for this purpose, as it allows for the identification and addressing of individual color centers that emit only single photons. This microscope also feature an integrated self-built microwave and magnetic hardware setup, which allows for a wide range of spin environment spectroscopy studies. Single photon emission is demonstrated through both photon anti-bunching and Rabi oscillations at room temperature.The second part of the thesis offers an exploration of superconducting single photon detectors through experiment. Since electronics are an essential part of these detectors, the possibility of using a novel alternative scheme based on capacitive readout combined with fast gating to enable simplified readout is demonstrated. This scheme overcomes the limitations of conventional readout schemes, which require large bandwidth amplification and complex counting electronics. Besides photon detection, the capabilities of these detectors are also expanded to include high-energy particles in the MeV energy range, and the detectors are demonstrated to not only detect single α- and β-particles, but to do so with near unity efficiency. Finally, a multipurpose testing station for superconducting detectors is demonstrated with a central objective of optimizing the coupling efficiency of light to the active area of the detector, as well as to allow for a fast exchange of the optical fiber, thereby facilitating an efficient characterization of the detector. The optimization of this coupling efficiency was demonstrated through proof-of-principle experiments.

    • A new birkeniid anaspid, Cowielepis ritchiei gen. et sp. nov., from the mid Silurian Cowie Harbour fish bed in Scotland is described on the basis of three specimens. Although sharing characters with various well-known anaspids, it possesses a unique combination of features that justifies the establishment of a new genus. Cowielepis is characterized by a single row of dorsolateral scales and a distinctive skull roof pattern with a large pineal plate and elongated posterior plates. The presence of paired ventrolateral fins in C. ritchiei supports previous suggestions that all anaspids possess such fins, but leaves open the question of homology with the gnathostome pectoral fin.

    • Photodynamic therapy (PDT) with topical methyl aminolevulinate ( MAL) administered in two treatment sessions separated by 1 week is an effective treatment for actinic keratoses. This open prospective study compared the efficacy and safety of MAL-PDT given as a single treatment with two treatments of MAL-PDT 1 week apart. Two hundred and eleven patients with 413 thin to moderately thick actinic keratoses were randomized to either a single treatment with PDT using topical MAL ( regimen I; n=105) or two treatments 1 week apart ( regimen II; n=106). Each treatment involved surface debridement, application of Metvix(R) cream (160 mg/g) for 3 h, followed by illumination with red light using a light-emitting diode system ( peak wavelength 634 +/- 3 nm, light dose 37 J/cm(2)). Thirty-seven lesions (19%) with a non-complete response 3 months after a single treatment were re-treated. All patients were followed up 3 months after the last treatment. A total of 400 lesions, 198 initially treated once and 202 treated twice, were evaluable. Complete response rate for thin lesions after a single treatment was 93% (95% CI = 87 - 97%), which was similar to 89% ( 82 - 96%) after repeated treatment. Response rates were lower after single treatment of thicker lesions (70% ( 60 - 78%) vs 84% ( 77 - 91%)), but improved after repeated treatment (88% ( 82 - 94%)). The conclusion of this study is that single treatment with topical MAL-PDT is effective for thin actinic keratosis lesions; however, repeated treatment is recommended for thicker or non-responding lesions.

  • Consolidating Bus Charger Deployment and Fleet Management for Public Transit Electrification: A Life-Cycle Cost Analysis Framework

    • Despite rapid advances in urban transit electrification, the progress of systematic planning and management of the electric bus (EB) fleet is falling behind. In this research, the fundamental issues affecting the nascent EB system are first reviewed, including charging station deployment, battery sizing, bus scheduling, and life-cycle analysis. At present, EB systems are planned and operated in a sequential manner, with bus scheduling occurring after the bus fleet and infrastructure have been deployed, resulting in low resource utilization or waste. We propose a mixed-integer programming model to consolidate charging station deployment and bus fleet management with the lowest possible life-cycle costs (LCCs), consisting of ownership, operation, maintenance, and emissions expenses, thereby narrowing the gap between optimal planning and operations. A tailored branch-and-price approach is further introduced to reduce the computational effort required for finding optimal solutions. Analytical results of a real-world case show that, compared with the current bus operational strategies and charging station layout, the LCC of one bus line can be decreased significantly by 30.4%. The proposed research not only performs life-cycle analysis but also provides transport authorities and operators with reliable charger deployment and bus schedules for single- and multi-line services, both of which are critical requirements for decision support in future transit systems with high electrification penetration, helping to accelerate the transition to sustainable mobility.
