  • Guidelines for the use and interpretation of assays for monitoring autophagy

    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • State of the climate in 2013

    • © 2014, American Meteorological Society. All rights reserved. In 2013, the vast majority of the monitored climate variables reported here maintained trends established in recent decades. ENSO was in a neutral state during the entire year, remaining mostly on the cool side of neutral with modest impacts on regional weather patterns around the world. This follows several years dominated by the effects of either La Niña or El Niño events. According to several independent analyses, 2013 was again among the 10 warmest years on record at the global scale, both at the Earth’s surface and through the troposphere. Some regions in the Southern Hemisphere had record or near-record high temperatures for the year. Australia observed its hottest year on record, while Argentina and New Zealand reported their second and third hottest years, respectively. In Antarctica, Amundsen-Scott South Pole Station reported its highest annual temperature since records began in 1957. At the opposite pole, the Arctic observed its seventh warmest year since records began in the early 20th century. At 20-m depth, record high temperatures were measured at some permafrost stations on the North Slope of Alaska and in the Brooks Range. In the Northern Hemisphere extratropics, anomalous meridional atmospheric circulation occurred throughout much of the year, leading to marked regional extremes of both temperature and precipitation. Cold temperature anomalies during winter across Eurasia were followed by warm spring temperature anomalies, which were linked to a new record low Eurasian snow cover extent in May. Minimum sea ice extent in the Arctic was the sixth lowest since satellite observations began in 1979. Including 2013, all seven lowest extents on record have occurred in the past seven years. Antarctica, on the other hand, had above-average sea ice extent throughout 2013, with 116 days of new daily high extent records, including a new daily maximum sea ice area of 19.57 million km2 reached on 1 October. ENSO-neutral conditions in the eastern central Pacific Ocean and a negative Pacific decadal oscillation pattern in the North Pacific had the largest impacts on the global sea surface temperature in 2013. The North Pacific reached a historic high temperature in 2013 and on balance the globally-averaged sea surface temperature was among the 10 highest on record. Overall, the salt content in nearsurface ocean waters increased while in intermediate waters it decreased. Global mean sea level continued to rise during 2013, on pace with a trend of 3.2 mm yr-1 over the past two decades. A portion of this trend (0.5 mm yr-1) has been attributed to natural variability associated with the Pacific decadal oscillation as well as to ongoing contributions from the melting of glaciers and ice sheets and ocean warming. Global tropical cyclone frequency during 2013 was slightly above average with a total of 94 storms, although the North Atlantic Basin had its quietest hurricane season since 1994. In the Western North Pacific Basin, Super Typhoon Haiyan, the deadliest tropical cyclone of 2013, had 1-minute sustained winds estimated to be 170 kt (87.5 m s-1) on 7 November, the highest wind speed ever assigned to a tropical cyclone. High storm surge was also associated with Haiyan as it made landfall over the central Philippines, an area where sea level is currently at historic highs, increasing by 200 mm since 1970. In the atmosphere, carbon dioxide, methane, and nitrous oxide all continued to increase in 2013. As in previous years, each of these major greenhouse gases once again reached historic high concentrations. In the Arctic, carbon dioxide and methane increased at the same rate as the global increase. These increases are likely due to export from lower latitudes rather than a consequence of increases in Arctic sources, such as thawing permafrost. At Mauna Loa, Hawaii, for the first time since measurements began in 1958, the daily average mixing ratio of carbon dioxide exceeded 400 ppm on 9 May.   • Survey on European methodologies in the risk assessment of chemical exposures in emergency response situations

    • Survey on European methodologies in the risk assessment of chemical exposures in emergency response situations. Prevention or mitigation of human health effects is often the major determinant underlying chemical incident prevention policy and emergency response decisions. The ability to perform a human health risk assessment is a prerequisite for effective chemical incident prevention, preparedness and response. To identify knowledge gaps, needs and concerns relating to health risks from chemical incidents, a web-based survey was sent to various groups of stakeholders. The release of acutely toxic substances and irritating/corrosive substances appeared to be the most important risk scenario. Almost 40% of the respondents also expected a future increase of chemical terrorism or sabotage. Developments in nanotechnology were perceived as potential future risk drivers although more information is needed on the health hazards of nanoparticles. A high number of respondents also expressed concern for the consequences of globalization, international trade and higher industry efficiency demands on health risks through chemical incidents. Acute Exposure Reference Values (AERVs) were considered important cornerstones but a need was expressed for recommendations on their use for the management of chemical emergencies. Based on this survey, it is advised to develop European consensus on an authoritative methodology to derive AERVs, to design a process for their implementation and to provide guidance and training on their practical application. Attention should be paid to the widely used acutely toxic and irritating/corrosive substances, to specific endpoints such as carcinogenicity and reproductive toxicity and new and emerging chemicals. Research should focus on developing plausible scenarios for emerging human health risks from chemical incidents to allow better prioritisation of future risk assessments.

  • Human risk assessment of single exposure in chemical incidents : present situation and new and increasing chemical incident scenarios

    • The release of chemicals from their containment, either accidentally or deliberately, is one of the most relevant risk scenarios in Europe. A human health risk assessment is a prerequisite for chemical incident prevention, preparedness and response. European guidance and harmonized Acute Exposure Reference Values (AERVs) are urgently needed for effective human health risk assessment in the context of chemical incidents.At present, no broad European consensus is available on guidance for risk assessment, risk management and risk communication purposes in case of chemical incidents. A review of legislation, existing or currently under revision, suggests that harmonized European guidance is not expected to be developed in the short term. An increasing number of European countries are developing their own procedures to assess the human health risk of chemical incident scenarios. The AERVs thus produced serve different purposes and are not interchangeable. Lack of international harmonization seriously obstructs a consistent response in chemical emergencies with transboundary effects within and beyond the EU, will hamper multinational companies attempting to make consistent risk assessments worldwide and will hinder consistent and transparent assessment, and management and communication of risks by different stakeholders.Emerging chemical incident risk scenarios and risk drivers have been identified. It is recommended to monitor more frequently at an early stage for new trends in chemicals, scenarios and risks from chemical incidents. A need for a specific approach to deal with single exposure to mixtures of chemicals is identified, as well as for specific guidance to adequately protect professional first responders.

  • The molecular gas reservoir of 6 low-metallicity galaxies from the Herschel Dwarf Galaxy Survey A ground-based follow-up survey of CO(1-0), CO(2-1), and CO(3-2)

    • Context. Observations of nearby starburst and spiral galaxies have revealed that molecular gas is the driver of star formation. However, some nearby low-metallicity dwarf galaxies are actively forming stars, but CO, the most common tracer of this reservoir, is faint, leaving us with a puzzle about how star formation proceeds in these environments. Aims. We aim to quantify the molecular gas reservoir in a subset of 6 galaxies from the Herschel Dwarf Galaxy Survey with newly acquired CO data and to link this reservoir to the observed star formation activity. Methods. We present CO(1-0), CO(2-1), and CO(3-2) observations obtained at the ATNE Mopra 22-m, APEX, and IRAM 30-m telescopes, as well as [CII] 157 mu m and [OI] 63 mu m observations obtained with the Herschel/PACS spectrometer in the 6 low-metallicity dwarf galaxies: Haro 11, Mrk 1089, Mrk 930, NGC 4861, NGC 625, and UM 311. We derived their molecular gas masses from several methods, including using the CO-to-H-2 conversion factor X-CO (both Galactic and metallicity-scaled values) and dust measurements. The molecular and atomic gas reservoirs were compared to the star formation activity. We also constrained the physical conditions of the molecular clouds using the non-LTE code RADEX and the spectral synthesis code Cloudy. Results. We detect CO in 5 of the 6 galaxies, including first detections in Haro 11 (Z similar to 0.4 Z(circle dot)), Mrk 930 (0.2 Z(circle dot)), and UM 311 (0.5 Z(circle dot)), but CO remains undetected in NGC 4861 (0.2 Z(circle dot)). The CO luminosities are low, while [CII] is bright in these galaxies, resulting in [CII]/CO(1-0) >= 10 000. Our dwarf galaxies are in relatively good agreement with the Schmidt-Kennicutt relation for total gas. They show short molecular depletion timescales, even when considering metallicity-scaled X-CO factors. Those galaxies are dominated by their HI gas, except Haro 11, which has high star formation efficiency and is dominated by ionized and molecular gas. We determine the mass of each ISM phase in Haro 11 using Cloudy and estimate an equivalent X-CO factor that is 10 times higher than the Galactic value. Overall, our results confirm the emerging picture that CO suffers from significant selective photodissociation in low-metallicity dwarf galaxies.

  • Switching clotting factor concentrates: considerations in estimating the risk of immunogenicity

    • The development of neutralizing antibodies to factor VIII (FVIII) is the most serious complication of therapy for haemophilia A. There is now excellent documentation that a large number of both genetic and environmental factors contribute to the risk of FVIII inhibitor incidence. One of the environmental factors that has been proposed as an influence on this complication is the occurrence of FVIII product switching. There are only a small number of clinical studies that have addressed this question, and thus, the amount of objective information available to assess this association is limited. In this review, in addition to summarizing past evidence pertinent to this subject, we present the results of a complementary strategy, a Delphi analysis, to add to the considerations of product switching and FVIII immunogenicity. With the imminent arrival in the clinic of several new FVIII products, the haemophilia community must be prepared to collect prospectively controlled data to better address this important management issue.

  • SPECTRAL AND MORPHOLOGICAL ANALYSIS OF THE REMNANT OF SUPERNOVA 1987A WITH ALMA AND ATCA

    • We present a comprehensive spectral and morphological analysis of the remnant of supernova (SN) 1987A with the Australia Telescope Compact Array (ATCA) and the Atacama Large Millimeter/submillimeter Array (ALMA). The non-thermal and thermal components of the radio emission are investigated in images from 94 to 672 GHz (lambda 3.2 mm to 450 mu m), with the assistance of a high-resolution 44 GHz synchrotron template from the ATCA, and a dust template from ALMA observations at 672 GHz. An analysis of the emission distribution over the equatorial ring in images from 44 to 345 GHz highlights a gradual decrease of the east-to-west asymmetry ratio with frequency. We attribute this to the shorter synchrotron lifetime at high frequencies. Across the transition from radio to far infrared, both the synchrotron/dust-subtracted images and the spectral energy distribution (SED) suggest additional emission beside the main synchrotron component (S-nu proportional to nu(-0.73)) and the thermal component originating from dust grains at T similar to 22 K. This excess could be due to free-free flux or emission from grains of colder dust. However, a second flat-spectrum synchrotron component appears to better fit the SED, implying that the emission could be attributed to a pulsar wind nebula (PWN). The residual emission is mainly localized west of the SN site, as the spectral analysis yields -0.4 less than or similar to alpha less than or similar to -0.1 across the western regions, with alpha similar to 0 around the central region. If there is a PWN in the remnant interior, these data suggest that the pulsar may be offset westward from the SN position.

    • A scientifically sound assessment of the risk to human health resulting from acute chemical releases is the cornerstone for chemical incident prevention, preparedness and response. Although the general methodology to identify acute toxicity of chemicals has not substantially changed in the last decades, there is ongoing debate on the current approaches for human health risk assessment in scenarios involving acute chemical releases.A survey was conducted to identify 1) the most important present and potential future chemical incident scenarios and anticipated changes in chemical incidents or their management; 2) information, tools and guidance used in different countries to assess health risks from acute chemical releases; and 3) needs for new information, tools, guidance and expertise to enable the valid and rapid health risk assessment of acute chemical exposures.According to the results, there is an obvious variability in risk assessment practices within Europe. The multiplicity of acute exposure reference values appears to result in variable practices. There is a need for training especially on the practical application of acute exposure reference values. Although acutely toxic and irritating/corrosive chemicals will remain serious risks also in future the development of plausible scenarios for potential emerging risks is also needed. This includes risks from new mixtures and chemicals (e.g. nanoparticles).
