    • Context: TGFBR1*6A is a common polymorphism of the type I transforming growth factor 0 receptor (TGFBR1). Epidemiological studies suggest that TGFBR1*6A may act as a tumor susceptibility allele. How TGFBR1*6A contributes to cancer development is largely unknown.. Objectives: To determine whether TGFBR1*6A is somatically acquired by primary tumors and metastases during cancer development and whether the 3-amino acid deletion that differentiates TGFBR1*6A from TGFBR1 is part of the mature receptor or part of the signal sequence and to investigate TGFBR1*6A signaling in cancer cells. Design, Setting, and Patients: Tumor And germline tissues from 531 patients with a diagnosis of head and neck, colorectal, or breast cancer recruited from 3 centers in the United States and from 1 center in Spain from June 1, 1994, through June 30, 2004, In vitro translation assays, MCF-7 breast cancer cells stably transfected with TGFBR1*6A, TGFBR1, or the vector alone, DLD-1 colorectal cancer cells that endogenously carry TGFBR1*6A, and SW48 colorectal cancer cells that do not carry TGFBR1*6A. Main Outcome Measures: TGFBR1*6A somatic acquisition in cancer. Determination of the amino terminus of the mature TGFBR1*6A and TGFBR1 receptors. Determination of TGF-beta-dependent cell proliferation. Results: TGFBR1*6A was somatically acquired in 13 of 44 (29.5%) colorectal cancer metastases, in 4 of 157 (2.5%) of colorectal tumors, in 4 of 226 (1.8%) head and neck primary tumors, and in none of the 104 patients with breast cancer. TGFBR1*6A somatic acquisition is not associated with loss of heterozygosity, microsatellite instability, or a mutator phenotype. The signal sequences of TGFBR1 and TGFBR1*6A are cleaved at the same site resulting in identical mature receptors. TGFBR1*6A may switch TGF-beta growth inhibitory signals into growth stimulatory signals in MCF-7 breast cancer cells and in DLD-1 colorectal cancer cells. Conclusions: TGFBR1*6A is somatically acquired in 29.5% of liver metastases from colorectal cancer and may bestow cancer cells with a growth advantage in the presence of TGF-beta. The functional consequences of this conversion appear to be mediated by the TGFBR1*6A signal sequence rather than by the mature receptor. The results highlight a new facet of TGF-beta signaling in cancer and suggest that TGFBR1*6A may represent a potential therapeutic target in cancer.

    • A high performance apparatus has been designed and built by the H8-RD22 collaboration for the study of channeling and volume reflection phenomena in the interaction of 400 GeVc protons with bent silicon crystals, during the 2006 data taking in the external beamline H8 of the CERN SPS. High-quality silicon short crystals were bent by either anticlastic or quasimosaic effects. Alignment with the highly parallel (8 μrad divergence) proton beam was guaranteed through a submicroradian goniometric system equipped with both rotational and translational stages. Particle tracking was possible by a series of silicon microstrip detectors with high-resolution and a parallel plate gas chamber, triggered by various scintillating detectors located along the beamline. Experimental observation of volume reflection with 400 GeVc protons proved true with a deflection angle of (10.4±0.5) μrad with respect to the unperturbed beam, with a silicon crystal whose (111) planes were parallel to the beam. © 2008 American Institute of Physics.

    • Work is in progress to completely replace, in 2008/9, the existing JET CFC tiles with a configuration of plasma facing materials consistent with the ITER design. The ITER-like wall (ILW) will be created with a combination of beryllium ( Be), tungsten ( W), W-coated CFC and Be-coated inconel tiles, with the material depending on the local anticipated heat flux and geometry. It is part of an integrated package of JET enhancements whose aim is to develop an understanding of the ITER materials issues and develop the techniques required to operate with inductive and advanced scenarios as close as possible to ITER parameters. Over 4000 tiles will be replaced and the ILW will accommodate additional heating up to at least 50 MW for 10 s. This paper describes the scientific background to the project, the technical objectives, the material configuration selected, the R&D behind the practical realization of the objectives and the generic problems associated with the Be tiles ( power handling capacity and disruption induced eddy currents). One of the objectives is to maintain or improve the existing CFC tile power handling performance which has been achieved in most cases by hiding bolt holes, optimizing tile size and profile and introducing castellations on plasma facing surfaces.

    • Currently, the primary ITER materials choice is a full beryllium main wall with carbon fibre composite at the divertor strike points and tungsten on the upper vertical targets and dome. The full tungsten divertor option is a possibility for the subsequent D-T phase. Neither of the ITER material combinations of first wall and divertor materials has ever been tested in a tokamak. To collect operational experience at JET with ITER relevant material combination (Be, C and W) would reduce uncertainties and focus the preparation for ITER operations. Therefore, the ITER-like wall project has been launched to install in JET a tungsten divertor and a beryllium main wall. This paper describes the R&D activities carried out for the project to develop an inertially cooled bulk tungsten divertor tile, to fully characterise tungsten coating technologies for CFC divertor tiles and to develop erosion markers for use as diagnostics on beryllium tiles.

    • ITER-Like Wall Project has been launched at the JET tokamak in order to study a tokamak operation with beryllium components on the main chamber wall and tungsten in the divertor. To perform this first comprehensive test of both materials in a thermonuclear fusion environment, a broad program has been undertaken to develop plasma-facing components and assess their performance under high power loads. The paper provides a concise report on scientific and technical issues in the development of a beryllium first wall at JET.

    • This paper presents a detailed study of the deflection phenomena of a 400GeV/c proton beam impinging on a new generation of bent silicon crystals; the tests have been performed at the CERN Super Proton Synchrotron H8 beam line. Channeling and volume reflection angles are measured with an extremely precise goniometer and with high resolution silicon microstrip detectors. Volume reflection has been observed and measured for the first time at this energy, with a single-pass efficiency as large as 98%, in good agreement with the simulation results. This efficiency makes volume reflection a possible candidate for collimation with bent crystals at the CERN Large Hadron Collider. © 2008 The American Physical Society.
