  • With Reference to Reference Genes : A Systematic Review of Endogenous Controls in Gene Expression Studies

    • The choice of reference genes that are stably expressed amongst treatment groups is a crucial step in real-time quantitative PCR gene expression studies. Recent guidelines have specified that a minimum of two validated reference genes should be used for normalisation. However, a quantitative review of the literature showed that the average number of reference genes used across all studies was 1.2. Thus, the vast majority of studies continue to use a single gene, with beta-actin (ACTB) and/or glyceraldehyde 3-phosphate dehydrogenase (GAPDH) being commonly selected in studies of vertebrate gene expression. Few studies (15%) tested a panel of potential reference genes for stability of expression before using them to normalise data. Amongst studies specifically testing reference gene stability, few found ACTB or GAPDH to be optimal, whereby these genes were significantly less likely to be chosen when larger panels of potential reference genes were screened. Fewer reference genes were tested for stability in non-model organisms, presumably owing to a dearth of available primers in less well characterised species. Furthermore, the experimental conditions under which real-time quantitative PCR analyses were conducted had a large influence on the choice of reference genes, whereby different studies of rat brain tissue showed different reference genes to be the most stable. These results highlight the importance of validating the choice of normalising reference genes before conducting gene expression studies.

  • Microstructure evolution-based design of thermal post-treatments for EBM-built Alloy 718

    • Alloy 718 samples were fabricated by electron beam melting (EBM) additive manufacturing process. The work focused on systematic investigation of response of the material to various thermal post-treatments, involving hot isostatic pressing (HIPing), solution treatment (ST) and two-step aging, to tailor post-treatment procedure for EBM-built Alloy 718. Results showed that HIPing at lowered temperature can be used for attaining desired defect closure while preserving grain size. Subjecting the material to ST, with or without prior HIPing, mainly caused precipitation of δ phase at the grain boundaries with prior HIPing decreasing the extent of δ phase precipitation. Moreover, results suggest that the utility of ST, with prior HIPing, could be dictated by the need to achieve a certain δ phase content, as the typically targeted homogenization after ST had already been achieved through HIPing. Detailed investigation of microstructural evolution during subsequent aging with and without prior HIPing showed that a significantly shortened aging treatment (‘4 + 1’ h), compared to the ‘standard’ long treatment (‘8 + 8’ h) traditionally developed for conventionally produced Alloy 718, might be realizable. These results can have significant techno-economic implications in designing tailored post-treatments for EBM-built Alloy 718.

  • Prior shared international experience and the survival of international new ventures

    • This paper shows how organizational members’ prior shared experience in international firms affects the survival of the international new ventures (INVs) to which they belong. I propose that as the length of prior shared international experience (PSIE) increases, these ventures are more likely to draw greater survival-enhancing benefits from the pre-existing routines and capabilities that their members previously developed while working together in the same international firm. However, for high PSIE length, INVs may find it difficult and costly to revise existing routines and capabilities and to develop new ones to achieve survival. Using a unique sample of Swedish INVs, I find that PSIE has an inverted U-shaped relationship with survival. Further, I theorize and show that contextual familiarity between the contexts in which PSIE was acquired and those in which it is applied through the INV is an important contingency of the PSIE-survival relationship. This study has valuable implications for research on international entrepreneurship and shared experience.

    • In this paper we will investigate and document some of the practices that a group of children use to form an activity of creating a screen-based interactive play world using a tangible programming system. A specific focus is on the resources that the children use to create and maintain a sense of shared understanding of what they are building. We emphasise the embodied aspects of interaction in the activity and how physical resources and bodily action must be viewed as intrinsically intertwined with the children's process of argumentation and articulation.

  • Struggle for Mobility : Risk, hope and community of knowledge in Eritrean and Ethiopian migration pathways towards Sweden

    • On the basis of the ethnographic fieldwork conducted in Sweden, Italy, Sudan and Ethiopia during 2013–2015, this study examines the motivations, organizations and impact of overland migratory journeys from Ethiopia and Eritrea across the Sahara Desert and Mediterranean Sea to Sweden. The analysis involves the exploring of how migrants strive to prepare, manage and survive the multiple risks and structural barriers they encounter: the exits from Eritrea and Ethiopia, negotiations and contacts with various brokers and facilitators, organized crime and violence, restrictive border controls, passage through the Desert and high Sea and finally, ‘managing the asylum system in Sweden’. Further, it maps how the process of contemporary refugee mobility and multiple transitions is facilitated by the entanglement of transnational social relations and smuggling practices. The study argues for a perspective wherein migration journeys are embedded in and affected by the process of dynamic intergenerational, translocal and transnational social relations, material practices and knowledge productions. It depicts how practices and facilitations of irregular migratory mobility reproduce collective knowledge that refugees mobilize to endure risks during their journey, establishing a community and creating a home after arriving at the destination location.

  • Comparing Calculated Nutrient Intakes Using Different Food Composition Databases : Results from the European Prospective Investigation into Cancer and Nutrition (EPIC) Cohort

    • This study aimed to compare calculated nutrient intakes from two different food composition databases using data from the European prospective investigation into cancer and nutrition (EPIC) cohort. Dietary intake data of the EPIC cohort was recently matched to 150 food components from the U.S. nutrient database (USNDB). Twenty-eight of these nutrients were already included in the EPIC nutrient database (ENDB-based upon country specific food composition tables), and used for comparison. Paired sample t-tests, Pearson's correlations (r), weighted kappa's (kappa) and Bland-Altman plots were used to compare the dietary intake of 28 nutrients estimated by the USNDB and the ENDB for 476,768 participants. Small but significant differences were shown between the USNDB and the ENDB for energy and macronutrient intakes. Moderate to very strong correlations (r = 0.60-1.00) were found for all macro- and micronutrients. A strong agreement (kappa > 0.80) was found for energy, water, total fat, carbohydrates, sugar, alcohol, potassium and vitamin C, whereas a weak agreement (kappa < 0.60) was found for starch, vitamin D and vitamin E. Dietary intakes estimated via the USNDB compare adequately with those obtained via the ENDB for most macro- and micronutrients, although the agreement was weak for starch, vitamin D and vitamin E. The USNDB will allow exposure assessments for 150 nutrients to investigate associations with disease outcomes within the EPIC cohort.

    • The majority of smokers begin their habit during adolescence, which often precedes experimentation with alcohol. Interestingly, very little preclinical work has been done examining how exposure to nicotine during periadolescence impacts the affective properties of alcohol in adulthood. Understanding how periadolescent nicotine exposure influences the aversive effects of alcohol might help to explain why it becomes more acceptable to this preexposed population. Thus, Experiment 1 exposed male Sprague Dawley rats to either saline or nicotine (0.4mg/kg, IP) from postnatal days 34 to 43 (periadolescence) and then examined changes in the aversive effects of alcohol (0, 0.56, 1.0 and 1.8g/kg, IP) in adulthood using the conditioned taste aversion (CTA) design. Changes in blood alcohol concentration (BAC) as well as alcohol-induced hypothermia and locomotor suppression were also assessed. To determine if changes seen were specific to nicotine exposure during periadolescence, the procedures were replicated in adults (Experiment 2). Preexposure to nicotine during periadolescence attenuated the acquisition of the alcohol-induced CTAs (at 1.0g/kg) and the hypothermic effects of alcohol (1.0g/kg). Adult nicotine preexposure produced similar attenuation in alcohol's aversive (at 1.8g/kg) and hypothermic (1.8g/kg) effects. Neither adolescent nor adult nicotine preexposure altered BACs or alcohol-induced locomotor suppression. These results suggest that nicotine may alter the aversive and physiological effects of alcohol, regardless of the age at which exposure occurs, possibly increasing its overall reinforcing value and making it more likely to be consumed.
