  • Genome-wide association study identifies a variant in HDAC9 associated with large vessel ischemic stroke

    • Genetic factors have been implicated in stroke risk, but few replicated associations have been reported. We conducted a genome-wide association study (GWAS) for ischemic stroke and its subtypes in 3,548 affected individuals and 5,972 controls, all of European ancestry. Replication of potential signals was performed in 5,859 affected individuals and 6,281 controls. We replicated previous associations for cardioembolic stroke near PITX2 and ZFHX3 and for large vessel stroke at a 9p21 locus. We identified a new association for large vessel stroke within HDAC9 (encoding histone deacetylase 9) on chromosome 7p21.1 (including further replication in an additional 735 affected individuals and 28,583 controls) (rs11984041; combined P = 1.87 x 10(-11); odds ratio (OR) = 1.42, 95% confidence interval (CI) = 1.28-1.57). All four loci exhibited evidence for heterogeneity of effect across the stroke subtypes, with some and possibly all affecting risk for only one subtype. This suggests distinct genetic architectures for different stroke subtypes.

    • Calls for more broad-based, integrated, useful knowledge now abound in the world of global environmental change science. They evidence many scientists' desire to help humanity confront the momentous biophysical implications of its own actions. But they also reveal a limited conception of social science and virtually ignore the humanities. They thereby endorse a stunted conception of 'human dimensions' at a time when the challenges posed by global environmental change are increasing in magnitude, scale and scope. Here, we make the case for a richer conception predicated on broader intellectual engagement and identify some preconditions for its practical fulfilment. Interdisciplinary dialogue, we suggest, should engender plural representations of Earth's present and future that are reflective of divergent human values and aspirations. In turn, this might insure publics and decision-makers against overly narrow conceptions of what is possible and desirable as they consider the profound questions raised by global environmental change.

  • Communities in the Digital Age : Towards a Theoretical Model of Communities of Practice and Information Technology

    • The global change towards a knowledge economy forces organizations to seek new and better ways to manage knowledge work and learning. Organizations have recognized that dfferent information technologies can play an important role in their quest to make the desired changes take place. Along with this increasing interest in information technologies we have also noticed a growing interest in a specific type of community, referred to as communities of practice (CoP). A considerable body of research that studies communities of practice in relation to IT continues to grow. Despite this we are far from a theoretical model of the relation between communities of practice and IT that may guide future research and understanding in a fruitful way.The basis for this thesis are the lack of a theoretical model to describe a dialectical relation between communities of practice and information technologies and how these relations come into being. The purpose of this thesis and research study is to contribute to existing theories of communities of practice by developing a theoretical model of a dialectical relation between information technology and communities of practice. In line with this, the study addresses the following research question How do a community of practice and information technology constitute and shape each other? Furthermore, the research study adheres to an interpretative philosophy, a qualitative research design, and longitudinal research case study conducted at Siemens Sweden.This research study was divided in two parts. Part One presented the theoretical chapters and a theoretical working model, referred to as the CoPIT model (Community of Practice model of Information Technology). In Part Two the initial CoPIT model was confronted with empirical data and used to analyze and illustrate the relation between CoP and IT. The main conclusion drawn from the research study is that the CoPIT model makes sense and is relevant, and also that the general principles upon which it is founded hold true. Furthermore, from the empirical confrontation of the CoPIT model we can draw the conclusion that relations exist both between IT in general and the CoP component’s specific constructs and between CoP in general and the IT component’s specific constructs. These key findings validate the point of departure of this study that there is a dialectical relation between CoP and IT, and that they mutually constitute each other over time.

  • Biological effects of a disposable, canisterless negative pressure wound therapy system.

    • Objective: Recent developments of negative pressure wound therapy (NPWT) systems have focused on making pumps smaller, lighter, and more portable. The recently introduced PICO system manages wound fluid through a highly breathable film within the dressing, thereby negating the need for a canister, which allows greater mobility and patient concordance. The aim of this study is to compare the biological effects of this system compared to a traditional NPWT system. Methods: Laboratory tests were carried out to demonstrate the fluid handling properties of the PICO™ system. Porcine full thickness defect wounds and sutured incisional wounds were used to compare the biological effects. Wounds were treated with PICO dressings or traditional NPWT dressings and connected to either a PICO device or a traditional NPWT device. Results: The PICO dressing manages exudate predominantly through evaporative loss (up to 85% of all fluid entering the dressing). Both traditional NPWT and the PICO system maintained therapeutic levels of negative pressure in all wounds. Both NPWT systems produced similar effects on wound edge contraction and microvascular blood flow in defect wounds. No significant changes in blood flow or wound contraction were noted in incision wounds for any NPWT combinations tested. Conclusions: The disposable, canisterless PICO NPWT system functions in the same manner as the traditional NPWT systems with regard to fluid handling, pressure transmission to the wound bed, tissue contraction, and changes in blood flow.

    • Quantitative ultrasound of the heel captures heel bone properties that independently predict fracture risk and, with bone mineral density (BMD) assessed by X-ray (DXA), may be convenient alternatives for evaluating osteoporosis and fracture risk. We performed a meta-analysis of genome-wide association (GWA) studies to assess the genetic determinants of heel broadband ultrasound attenuation (BUA; n = 14 260), velocity of sound (VOS; n = 15 514) and BMD (n = 4566) in 13 discovery cohorts. Independent replication involved seven cohorts with GWA data (in silico n = 11 452) and new genotyping in 15 cohorts (de novo n = 24 902). In combined random effects, meta-analysis of the discovery and replication cohorts, nine single nucleotide polymorphisms (SNPs) had genome-wide significant (P < 5 x 10(-8)) associations with heel bone properties. Alongside SNPs within or near previously identified osteoporosis susceptibility genes including ESR1 (6q25.1: rs4869739, rs3020331, rs2982552), SPTBN1 (2p16.2: rs11898505), RSPO3 (6q22.33: rs7741021), WNT16 (7q31.31: rs2908007), DKK1 (10q21.1: rs7902708) and GPATCH1 (19q13.11: rs10416265), we identified a new locus on chromosome 11q14.2 (rs597319 close to TMEM135, a gene recently linked to osteoblastogenesis and longevity) significantly associated with both BUA and VOS (P < 8.23 x 10(-14)). In meta-analyses involving 25 cohorts with up to 14 985 fracture cases, six of 10 SNPs associated with heel bone properties at P < 5 x 10(-6) also had the expected direction of association with any fracture (P < 0.05), including three SNPs with P < 0.005: 6q22.33 (rs7741021), 7q31.31 (rs2908007) and 10q21.1 (rs7902708). In conclusion, this GWA study reveals the effect of several genes common to central DXA-derived BMD and heel ultrasound/DXA measures and points to a new genetic locus with potential implications for better understanding of osteoporosis pathophysiology.

    • The fission yeast clade-comprising Schizosaccharomyces pombe, S. octosporus, S. cryophilus, and S. japonicus-occupies the basal branch of Ascomycete fungi and is an important model of eukaryote biology. A comparative annotation of these genomes identified a near extinction of transposons and the associated innovation of transposon-free centromeres. Expression analysis established that meiotic genes are subject to antisense transcription during vegetative growth, which suggests a mechanism for their tight regulation. In addition, trans-acting regulators control new genes within the context of expanded functional modules for meiosis and stress response. Differences in gene content and regulation also explain why, unlike the budding yeast of Saccharomycotina, fission yeasts cannot use ethanol as a primary carbon source. These analyses elucidate the genome structure and gene regulation of fission yeast and provide tools for investigation across the Schizosaccharomyces clade.

    • This is a history of beginnings, but it doesn't come from 1879. Rather it is about the politics of the World Centennial of National Parks in 1972. Not in 1964 - 100 years after the first national park in the world, Yosemite. Nor in 1979 - 100 years after Royal National Park was legislated as the first national park in the world. The politics of the decision to rewrite history to make Yellowstone National Park the signature ‘first’ national park in the world is entangled with the perception of RNP's history. This decision reveals much about the workings of Earth Day politics in USA in 1970, and also the networks between national parks managers in Australia and in the United States in this period. The story speaks to the tensions between the national and global in the politics of biological diversity at a time when national parks were seen as the primary solution to a crisis of extinctions. It also reflects the tensions in Australia inherent in the constitutional responsibility for ‘national’ parks being vested in the states and the issues this still raises for Australia's international responsibilities for global biodiversity.
