- The application of quantitative methods in electron probe X-ray microanalysis of biological specimens expands the scope of this technique. Accurate quantitation in biological X-ray microanalysis is not easy and a number of difficulties is typical for biological specimens. This thesis presents: (1) a method to optimize the instrumental configuration by minimizing the contribution of extraneous sources to the X-ray spectrum through the use of a carbon specimen holder, (2) methods to prepare standards for analysis of thin cryosections, thin sections of resin-embedded tissue and bulk specimens, (3) practical methods to carry out the quantitative determinations and calculations. The applications of quantitative electron probe X-ray microanalysis include: distribution of K and Na in the mould Neurospora crassa, and of Br in the marine red alga Chondrus crispus, and three applications in the field of human pathology: X-ray microanalysis of single muscle fibres, diagnosis of cystic fibrosis, and the demonstration of copper in 'green hair'.
