  • A comprehensive map coupling histone modifications with gene regulation in adult dopaminergic and serotonergic neurons

    • The brain is composed of hundreds of different neuronal subtypes, which largely retain their identity throughout the lifespan of the organism. The mechanisms governing this stability are not fully understood, partly due to the diversity and limited size of clinically relevant neuronal populations, which constitute a technical challenge for analysis. Here, using a strategy that allows for ChIP-seq combined with RNA-seq in small neuronal populations in vivo, we present a comparative analysis of permissive and repressive histone modifications in adult midbrain dopaminergic neurons, raphe nuclei serotonergic neurons, and embryonic neural progenitors. Furthermore, we utilize the map generated by our analysis to show that the transcriptional response of midbrain dopaminergic neurons following 6-OHDA or methamphetamine injection is characterized by increased expression of genes with promoters dually marked by H3K4me3/H3K27me3. Our study provides an in vivo genome-wide analysis of permissive/repressive histone modifications coupled to gene expression in these rare neuronal subtypes.

    • For an animal to escape an attacking predator, speed and manoeuvrability are likely to be crucial factors. Previous studies on reptiles and birds have revealed that gaining weight, due to for instance egg load or lipid accumulation, leads to impaired escape ability and possibly increases the risk of being caught by predators. Here we tested whether the flight performance of an insect, the small tortoiseshell butterfly (Aglais urticae), is affected by variations in body mass due to feeding by comparing flight performance parameters between individuals that (i) took flight spontaneously or after being subjected to a simulated predator attack and (ii) varied in flight muscle ratio (FMR: thorax mass/total body mass). The results show that butterflies that were subjected to a simulated predator attack flew at higher velocities and straighter than butterflies that were allowed to take off spontaneously. Furthermore, our study suggests, for the first time, that butterflies may experience impaired escape ability after feeding, which directly leads to a decrease in FMR; a reduction from 0.5 to 0.3 reduced escape flight velocity by about 37% at 0.5 m from the start. Finally, this study shows that FMR affects take-off angle and sinuosity, with steeper angles and more turning in butterflies with low FMR.

    • Recent advances in information and communication technologies have made the development and operation of complex disease surveillance systems technically feasible, and many systems have been proposed to interpret diverse data sources for health-related signals. Implementing these systems for daily use and efficiently interpreting their output, however, remains a technical challenge. This thesis presents a method for understanding disease surveillance systems structurally, examines four existing systems, and discusses the implications of developing such systems. The discussion is followed by two papers. The first paper describes the design of a national outbreak detection system for daily disease surveillance. It is currently in use at the Swedish Institute for Communicable Disease Control. The source code has been licenced under GNU v3 and is freely available. The second paper discusses methodological issues in computational epidemiology, and presents the lessons learned from a software development project in which a spatially explicit micro-meso-macro model for the entire Swedish population was built based on registry data.

    • During 2011, a policy for scholarly publishing was implemented at KTH.1 The policy states that researchers at KTH should strive to publish open access, either through well-reputed open accessjournals or by self-archiving in KTH publication database DiVA. Publications published by KTH – i.e dissertations, bachelor and master theses, reports – are to be self-archived in DiVA as a general rule. In this report, shares and trends of open access publishing at KTH are analysed starting from the same year the policy was implemented, in 2011. A report that analyses open access at KTH will then be released annually

    • I denna studie beräknas våld vid busshållplatser samt våld vid busshållplatser relativt antalet busspassagerare i Malmö. En riskterrängmodell (RTM) genomförs också för att beräkna vad som kännetecknar busshållplatser med mycket våld. Resultaten visar att det är mest våld i centrum, men att risken är högst i utsatta områden med låg kollektiv förmåga.

    • Ecomodernists have argued that rather than imposing an “ethic of sustainability”, publicly funded breakthrough innovation can make possible both liberal freedom and expanding opportunity in the Anthropocene. Yet, just like the perfectionist social vision formulated by Randall Curren and Ellen Metzger, an ecomodern future would require far-reaching political commitment and coordination to an extent that may be at odds with real world political experiences. This commentary suggests that important insights into sustainability debates can be obtained by taking a longer look at human history.
