    • This initiative examined systematically the extent to which a large set of archival research findings generalizes across contexts. We repeated the key analyses for 29 original strategic management effects in the same context (direct reproduction) as well as in 52 novel time periods and geographies; 45% of the reproductions returned results matching the original reports together with 55% of tests in different spans of years and 40% of tests in novel geographies. Some original findings were associated with multiple new tests. Reproducibility was the best predictor of generalizability-for the findings that proved directly reproducible, 84% emerged in other available time periods and 57% emerged in other geographies. Overall, only limited empirical evidence emerged for context sensitivity. In a forecasting survey, independent scientists were able to anticipate which effects would find support in tests in new samples.

  • Prolonged SARS-CoV-2 RNA virus shedding and lymphopenia are hallmarks of COVID-19 in cancer patients with poor prognosis

    • Patients with cancer are at higher risk of severe coronavirus infectious disease 2019 (COVID-19), but the mechanisms underlying virus–host interactions during cancer therapies remain elusive. When comparing nasopharyngeal swabs from cancer and noncancer patients for RT-qPCR cycle thresholds measuring acute respiratory syndrome coronavirus-2 (SARS-CoV-2) in 1063 patients (58% with cancer), we found that malignant disease favors the magnitude and duration of viral RNA shedding concomitant with prolonged serum elevations of type 1 IFN that anticorrelated with anti-RBD IgG antibodies. Cancer patients with a prolonged SARS-CoV-2 RNA detection exhibited the typical immunopathology of severe COVID-19 at the early phase of infection including circulation of immature neutrophils, depletion of nonconventional monocytes, and a general lymphopenia that, however, was accompanied by a rise in plasmablasts, activated follicular T-helper cells, and non-naive Granzyme B+FasL+, EomeshighTCF-1high, PD-1+CD8+ Tc1 cells. Virus-induced lymphopenia worsened cancer-associated lymphocyte loss, and low lymphocyte counts correlated with chronic SARS-CoV-2 RNA shedding, COVID-19 severity, and a higher risk of cancer-related death in the first and second surge of the pandemic. Lymphocyte loss correlated with significant changes in metabolites from the polyamine and biliary salt pathways as well as increased blood DNA from Enterobacteriaceae and Micrococcaceae gut family members in long-term viral carriers. We surmise that cancer therapies may exacerbate the paradoxical association between lymphopenia and COVID-19-related immunopathology, and that the prevention of COVID-19-induced lymphocyte loss may reduce cancer-associated death.

  • Managing hypertension in frail oldest-old-The role of guideline use by general practitioners from 29 countries

    • BACKGROUND: The best management of hypertension in frail oldest-old (≥80 years of age) remains unclear and we still lack guidelines that provide specific recommendations. Our study aims to investigate guideline use in general practitioners (GPs) and to examine if guideline use relates to different decisions when managing hypertension in frail oldest-old. DESIGN/SETTING: Cross-sectional study among currently active GPs from 29 countries using a case-vignettes survey. METHODS: GPs participated in a survey with case-vignettes of frail oldest-olds varying in systolic blood pressure (SBP) levels and cardiovascular disease (CVD). GPs from 26 European countries and from Brazil, Israel and New Zealand were invited. We compared the percentage of GPs reporting using guidelines per country and further stratified on the most frequently mentioned guidelines. To adjust for patient characteristics (SBP, CVD and GPs' sex, years of experience, prevalence of oldest-old and location of their practice), we used a mixed-effects regression model accounting for clustering within countries. RESULTS: Overall, 2,543 GPs from 29 countries were included. 59.4% of them reported to use guidelines. Higher guideline use was found in female (p = 0.031) and less-experienced GPs (p<0.001). Across countries, we found a large variation in self-reported guideline use, ranging from 25% to 90% of the GPs, but there was no difference in hypertension treatment decisions in frail oldest-old patients between GPs that did not use and GPs that used guidelines, irrespective of the guidelines they used. CONCLUSION: Many GPs reported using guidelines to manage hypertension in frail oldest-old patients, however guideline users did not decide differently from non-users concerning hypertension treatment decisions. Instead of focusing on the fact if GPs use guidelines or not, we as a scientific community should put an emphasis on what guidelines suggest in frail and oldest-old patients.

  • Morphometric Control on Dissolved Organic Carbon in Subarctic Streams

    • Climate change has the potential to alter hydrological regimes and to expand saturated areas in permafrost environments, which are important sources of organic carbon. The sources, transfer zones, and delivery mechanisms of carbon into the stream network are controlled by the morphometric properties of the catchment; however, the utility and limitations of these properties as predictors of dissolved organic carbon concentrations have rarely been systematically evaluated. This study tested the relationships between 18 morphometric indicators and observed dissolved organic carbon (DOC) concentrations in the Stordalen catchment, Sweden. Geospatial and explorative statistics were combined to assess the topographical, areal, and linear indicators influencing the distribution of DOC in the catchment. The results suggest that catchment morphometric indicators can be used as proxies to predict DOC concentrations along a longitudinal continuum in subarctic climate regions (R2 up to 0.52). Morphometry indicators that best served as predictors of DOC concentration in the model were as follows: relief, slope length and steepness factor (LS‐factor), sediment transport capacity, and catchment area. Due to the influence that catchment form exerts in DOC spatial patterns and processing, a morphometric approach can serve as a first approximation of DOC spatial patterns within a catchment. The initial step in identifying carbon sources based on the catchment topography has the potential to allow for quick and multilevel comparison within and between catchments.

  • Cutover Peat Limits Methane Production Causing Low Emission at a Restored Peatland

    • Peatland degradation due to human activities is contributing to rising atmospheric CO2 levels. Restoring the carbon (C) sink function in degraded peatlands and preventing further stored C losses is a key climate mitigation strategy, given the global scale of peatland disturbance. Active restoration involving a combination of rewetting and vegetation reestablishment at a post-extraction peatland in Canada has been shown to successfully re-establish net CO2 uptake rates similar to undisturbed peatlands within a decade or two. However, lower than expected CH4 emissions suggest recovery of belowground C cycling processes may lag behind the recovery of the surface net flux. Using closed chamber measurements over a warm season, we determined that restored Sphagnum, which covers two thirds of the site, was a null source of CH4. Emissions from the restored site were primarily attributed to vascular plant substrate inputs, measured as acetate, and plant-mediated transport. The C isotopic fractionation factor for CH4 and CO2 in the pore water from the restored former peat field suggested reduced hydrogenotrophic CH4 production deeper in the cutover peat profile (0.8 m depth). In contrast, isotopic fractionation in the former drainage ditches showed a balance of acetoclastic and hydrogenotrophic methanogenesis deeper in the profile, indicative of some bulk peat C turnover. This study suggests that the legacy of substrate quality in the cutover peat can reduce the climate warming impact of newly restored peatlands through a reduction in CH4 production and thus emission.
