    • Molecular diagnostics can offer early detection of disease, improved diagnostic accuracy, and qualified follow-up. Moreover, the use of microfluidic devices can in principle render these analyses quickly and user-friendly, placing them within the reach of the general practitioner and maybe even in households. However, the progress launching such devices has been limited so far. We propose that an important limiting factor has been the difficulty of establishing molecular assays suitable for microfabricated formats. The assays should be capable of monitoring a wide range of molecules, including genomic DNA, RNA and proteins with secondary modifications and interaction partners, and they must exhibit excellent sensitivity and specificity. We discuss these problems and describe a series of molecular tools that may present new opportunities for lab-on-a-chip devices at the point-of-care.

    • We have developed a multipurpose microfluidic platform that allows for sensitive fluorescence detection on inexpensive disposable chips. The fabrication scheme involves rapid injection molding of thermoplastics, followed by silica deposition and covalent attachment of an unstructured flexible lid. This combines the virtues of elastomer technology with high-throughput compact disk injection molding. Using this technique, the time to produce 100 chips using a single master can be lowered from more than 1 week by standard PDMS technologies to only a couple hours. The optical properties of the fabricated chips were evaluated by studying individual fluorescence-labeled DNA molecules in a microchannel. Concatemeric DNA molecules were generated through rolling circle replication of circular DNA molecules, which were labeled by hybridization of fluorescence-tagged oligonucleotides. Rolling circle products (RCPs) were detected after as little as 5 min of DNA polymerization, and the RCPs in solution showed no tendency for aggregation. To illustrate the versatility of the platform, we demonstrate two additional applications: The flexible property of the lid was used to create a peristaltic pump generating a flow rate of 9 nL/s. Biocompatibility of the platform was illustrated by culturing Chinese hamster ovary cells for 7 days in the microfluidic channels.

  • Tangential velocity measurements in a conical hydrocyclone operated with a fibre suspension

    • The tangential velocity flow field in a conical hydrocyclone was measured using a self-cleaning pitometer. The influence of pulp fibre concentration on the tangential velocity profile was of particular interest. The measurement, showed that the pulp fibres had a strong influence on the tangential velocity profile. When operating the hydrocyclone with pure water, the velocity profile showed the typical combination of free-vortex-like rotation close to the hydrocyclone wall and solid-body-like rotation in the proximity of the hydrocyclone axis. This profile changed significantly when pulp fibres were added. At a low fibre concentration (1.2 g/l), the fibres smoothened the transition between the free-vortex-like and the solid-body-like region of the velocity profile. The location of the maximum tangential velocity moved to a larger radius. At higher fibre concentration (7.5 and 11 g/l), the free-vortex-like behaviour in the outer area was virtually suppressed. Due to networking, the fibre suspension in the entire hydrocyclone behaved as a solid body. The radial acceleration profile and tangential rate-of-strain profile were determined. Based on these profiles, a hypothesis was proposed explaining the well-known observation that fractionation efficiency decreases significantly at higher fibre concentrations.

    • We give conjectures on the possible graded Betti numbers of Cohen-Macaulay modules up to multiplication by positive rational numbers. The idea is that the Betti diagrams should be nonh negative linear combinations of pure diagrams. The conjectures are verified in the cases where the structure of resolutions is known, that is: for modules of codimension two, for Glorenstein algebras of codimension three and for complete intersections. The motivation for proposing the conjectures comes from the Multiplicity conjecture of Herzog, Huneke and Srinivasan.

    • BACKGROUND: Recently, the dual-side readout technique has been introduced in computed radiography, leading to an increase in detective quantum efficiency (DQE) compared with the single-side readout technique. PURPOSE: To evaluate if the increase in DQE with the dual-side readout technique results in a higher clinical image quality in chest radiography of premature neonates at no increase in radiation dose. MATERIAL AND METHODS: Twenty-four chest radiographs of premature neonates were collected from both a single-side readout technique system and a double-side readout technique system. The images were processed in the same image-processing station in order for the comparison to be only dependent on the difference in readout technique. Five radiologists rated the fulfillment of four image quality criteria, which were based on important anatomical landmarks. The given ratings were analyzed using visual grading characteristics (VGC) analysis. RESULTS: The VGC analysis showed that the reproduction of the carina with the main bronchi and the thoracic vertebrae behind the heart was better with the dual-side readout technique, whereas no significant difference for the reproduction of the central vessels or the peripheral vessels could be observed. CONCLUSIONS: The results indicate that the higher DQE of the dual-side readout technique leads to higher clinical image quality in chest radiography of premature neonates at no increase in radiation dose.
