  • Factory-in-a-Box - Solutions for Availability and Mobility of Flexible Production Capacity

    • The objective of this paper is to present examples of how to realize a flexible and reconfigurable production system. An ongoing research project in Sweden called Factory-in-a-Box will be presented which is one research initiative within this area. The purpose of the Factory-in-a-Box project is to develop solutions for mobile production capacity on demand. Three key features have been identified as enablers for these kinds of production capabilities: mobility, flexibility, and speed. The concept consists of standardized modules that can be installed in e.g. containers and easily transported by trucks, rail vehicles, and boats. The modules can easily be combined into complete production systems and reconfigured for new products and/or scaled to handle new volumes. The goal of the Factory-in-a-Box project is to build fully operative production modules that are developed in close cooperation between different academic and industrial partners. This paper will present the results from these demonstrators giving examples of the usability of the Factory-in-a-Box concept in industry.

    • We give conjectures on the possible graded Betti numbers of Cohen-Macaulay modules up to multiplication by positive rational numbers. The idea is that the Betti diagrams should be nonh negative linear combinations of pure diagrams. The conjectures are verified in the cases where the structure of resolutions is known, that is: for modules of codimension two, for Glorenstein algebras of codimension three and for complete intersections. The motivation for proposing the conjectures comes from the Multiplicity conjecture of Herzog, Huneke and Srinivasan.

    • Analytical reactions in which short DNA strands are used in combination with DNA ligases have proven useful for measuring, decoding, and locating most classes of macromolecules. Given the need to accumulate large amounts of precise molecular information from biological systems in research and in diagnostics, ligation reactions will continue to offer valuable strategies for advanced analytical reactions. Here, we provide a basis for further development of methods by reviewing the history of analytical ligation reactions, discussing the properties of ligation reactions that render them suitable for engineering novel assays, describing a wide range of successful ligase-based assays, and briefly considering future directions.

    • The adipocyte-derived hormone leptin is associated with insulin resistance and reduced fibrinolytic status - or dysfibrinolysis - in humans. As leptin associates differentially to the development of cardiovascular disease and diabetes in men and women, we hypothesized that leptin and insulin sensitivity are related to dysfibrinolysis in a sex-dependent manner. Thirty-two men and 40 women were recruited from the Monitoring of trends and determinants in Cardiovascular disease (MONICA) population sample, representing the highest and lowest quartiles of fasting insulin levels. Lipids, fibrinolytic status [plasminogen activator inhibitor 1 (PAI-1) activity, tissue plasminogen activator (tPA) mass and activity, and tPA-PAI complex], leptin, testosterone and sex-hormone-binding globulin were measured. Insulin sensitivity was estimated using the euglycaemic clamp technique. Body composition was determined by bioimpedance. Determinants for circulating levels of fibrinolytic factors were explored in a multivariate linear regression analysis. Levels of fibrinolytic variables and estimated insulin sensitivity did not differ between men and women. Leptin was independently associated with reduced fibrinolytic status high PAI-1 activity, low tPA activity, high tPA mass, and high tPA-PAI complex) in men (P<0.001-0.002). In women, fat mass and/or insulin sensitivity were related to these factors (P<0.001-0.03), and leptin only to reduced tPA activity (P = 0.002). Hyperleptinemia, dysfibrinolysis, insulin sensitivity and androgenicity associate differentially in men and women.     • Patients diagnosed with diffuse large B-cell lymphoma (DLBCL) show varying responses to conventional therapy, and this might be contributed to the differentiation stage of the tumor B-cells. The aim of the current study was to evaluate a panel of kinases (ZAP70, PKC-β I and II and phosphorylated PKB/Akt) and phosphatases (PTEN, SHP1 and SHP2) known to be frequently deregulated in lymphoid malignancies. De novo DLBCL cases were divided into two subgroups, the germinal center (GC) group (14/28) and the non-germinal center (non-GC) or activated B-cell (ABC) group (14/28). ZAP70 and PKC-β II were expressed in a significantly higher percentage of tumor cells in the clinically more aggressive non-GC group compared with the prognostically favourable GC group. Also, the subcellular localization of PKC-β I and II differed in DLBCL cells, with the PKC-β I isoform being expressed in both the cytoplasm and nucleus, while PKC-β II was found exclusively in the cytoplasm. Loss of nuclear PTEN correlated with poor survival in cases from both subgroups. In addition, five cell lines of DLBCL origin were analyzed for protein expression and for mRNA levels of PTEN and SHP1. For the first time, we show that ZAP70 is expressed in a higher percentage of tumor cells in the aggressive non-GC subgroup of DLBCL and that PKC-β I and II are differently distributed in the two prognostic subgroups of de novo DLBCL.

  • Land-Use and Land-Cover Dynamics in the Central Rift Valley of Ethiopia

    • Understanding the complexity of land-use and land-cover (LULC) changes and their driving forces and impacts on human and environmental security is important for the planning of natural resource management and associated decision making. This study combines and compares participatory field point sampling (pfps) and remote sensing to explore local LULC dynamics. The study was conducted in two peasant associations located in the central Ethiopian Rift Valley, which is a dry-land mixed farming area exposed to rapid deforestation. From 1973-2006, the area of cropland doubled at the expense of woodland and wooded-grassland in both of the study sites. Major deforestation and forest degradation took place from 1973-1986; woodland cover declined from 40% to 9% in one of the study sites, while the other lost all of its original 54% woodland cover. Our study concludes that assessing LULC dynamics using a combination of remote sensing and pfps is a valuable approach. The two methods revealed similar LULC trends, while the pfps provided additional details on how farmers view the changes. This study documents dramatic trends in LULC over time, associated with rapid population growth, recurrent drought, rainfall variability and declining crop productivity. The alarming nature of these trends is reflected in a decrease in the livelihood security of local communities and in environmental degradation. Given these dry-land conditions, there are few opportunities to improve livelihoods and environmental security without external support. If negative changes are to be halted, action must be taken, including building asset bases, instituting family planning services, and creating opportunities outside these marginal environments.
