  • Manipulation of CuO morphology for efficient potentiometric detection of urea via slow nucleation/growth kinetics exerted by mixed solvents

    • Controlling the reaction kinetics during the nucleation/growth of cupric oxide (CuO) nanostructures is very critical in order to achieve a specific and well-defined morphology. For this purpose, we have slowed down the reaction speed using a mixed solvent concept and successfully obtained a chain-like morphology of CuO nanostructures using hydrothermal method. The CuO chain-like morphology was synthesized using a 1:1 (v/v) ratio of ethylene glycol and water. The morphology and crystalline features of CuO were studied by scanning electron microscopy (SEM) and powder X-ray diffraction techniques. The high resolution transmission electron microscopy revealed 5 nm crystallite size for the CuO material prepared in the mixed solvents. The obtained results have shown that the prepared CuO chains had a monocline phase, containing only Cu and O as main elements as confirmed by energy dispersive spectroscopy. This unique morphology obtained from mixed solvent process has provided a better surface for the loading of urease enzyme, thus it enabled the development of sensitive and selective urea biosensor in phosphate buffer solution of pH 7.4. The physical adsorption method was used to immobilize urease enzyme onto the nano surface of CuO. The fabricated biosensor based on urease/CuO chains has shown a dynamic linear range from 0.0005 to15 mM with a low limit of detection 0.0001 mM. Additionally, a fast response time aroudn1s, h high selectivity, stability, repeatability, storage time, and reproducibility were observed. The effect of pH and temperature on the potentiometric signal of the developed biosensor was also examined. Importantly, the practical aspects of the fabricated urea biosensor were probed and the obtained percent recovery results revealed an outstanding performance. The strategy of using mixed solvent with equal volume ratio would be useful for the preparation of other metal oxides with improved catalytic properties for a wide range of clinical, biomedical and other related applications.

  • Sport-related concussion in soccer –a scoping review of available guidelines and a call for action to FIFA & soccer governing bodies

    • Introduction: Sport-related concussions (SRC) have been a concern in all sports, including soccer. The long-term effects of soccer-related head injuries are a public health concern. The Concussion in Sport Group (CISG) released a consensus statement in 2017 and several soccer governing associations have published their own SRC guidelines while referring to it but it is unclear whether this has been universally adopted. Research question: We aimed to investigate whether guidelines published by soccer associations have any discrepancies; and the extent to which they follow the CISG recommendations. Materials and methods: A scoping review of available soccer-specific SRC guidelines was performed via databases PubMed, Google Scholar, and official soccer association websites via web browser Google. The inclusion criteria were soccer-specific SRC guidelines. Comparisons between guidelines were made concerning the following index items: initial (on-site) assessment, removal from play, re-evaluation with neuroimaging, return-to-sport protocol, special populations, and education. Results: Nine soccer associations with available guidelines were included in this review. Guidelines obtained were from official associations in the United Kingdom, United States of America, Canada, Australia, and New Zealand. When compared to each other and the CISG recommendations, discrepancies were found within guidelines regarding the index items. Additionally, major soccer associations in some countries famous for soccer were found to have not published any publicly available guidelines. Discussion and conclusion: SRC guidelines from different soccer associations contain discrepancies which may be detrimental to athletes, both short and long-term. We recommend that all major soccer governing associations publish guidelines that are standardised and accessible to all athletes.

  • Individual and synergistic effects of different fertilizers and gibberellin on growth and morphology of chili seedlings

    • Gibberellins (GA3), as well as the basic elements phosphorus (P), nitrogen (N), and potassium (K), are crucial to the growth of chili. This study investigates the effect of different fertilizers and plant growth regulator on the growth and morphology of chili seedlings. Soil application of NPK, urea, SOP, and DAP (2.5 g/L) was applied during sowing, and N in two splits at sowing and after twenty days of sowing while foliar application of GA3 (50 mg/L) was applied after fifteen days of germination. The result of five seedlings' traits plant height (PH), plant girth (PG), plant spread (PS), number of leaves (NOF), and root length (RL) demonstrated a significant difference among growth-related traits in chili seedlings owing to the use of fertilizers, GA3, and their combinations. An optimum level of K and P alone or in combination with GA3 had a significant effect on all traits. PH was particularly influenced by the combination of GA3 with K and P whereas other traits like PG, NOF, PS, and RL are greatly influenced by the application of NPK, urea, SOP, DAP, and their combination with GA3. The study results showed an increase in chili seedlings' growth and morphology in response to various fertilizers and GA3.

  • CoMPARA : Collaborative Modeling Project for Androgen Receptor Activity

    • BACKGROUND: Endocrine disrupting chemicals (EDCs) are xenobiotics that mimic the interaction of natural hormones and alter synthesis, transport, or metabolic pathways. The prospect of EDCs causing adverse health effects in humans and wildlife has led to the development of scientific and regulatory approaches for evaluating bioactivity. This need is being addressed using high-throughput screening (HTS) in vitro approaches and computational modeling.OBJECTIVES: In support of the Endocrine Disruptor Screening Program, the U.S. Environmental Protection Agency (EPA) led two worldwide consortiums to virtually screen chemicals for their potential estrogenic and androgenic activities. Here, we describe the Collaborative Modeling Project for Androgen Receptor Activity (CoMPARA) efforts, which follows the steps of the Collaborative Estrogen Receptor Activity Prediction Project (CERAPP).METHODS: The CoMPARA list of screened chemicals built on CERAPP's list of 32,464 chemicals to include additional chemicals of interest, as well as simulated ToxCast (TM) metabolites, totaling 55,450 chemical structures. Computational toxicology scientists from 25 international groups contributed 91 predictive models for binding, agonist, and antagonist activity predictions. Models were underpinned by a common training set of 1,746 chemicals compiled from a combined data set of 11 ToxCast (TM)/Tox21 HTS in vitro assays.RESULTS: The resulting models were evaluated using curated literature data extracted from different sources. To overcome the limitations of single-model approaches, CoMPARA predictions were combined into consensus models that provided averaged predictive accuracy of approximately 80% for the evaluation set.DISCUSSION: The strengths and limitations of the consensus predictions were discussed with example chemicals; then, the models were implemented into the free and open-source OPERA application to enable screening of new chemicals with a defined applicability domain and accuracy assessment. This implementation was used to screen the entire EPA DSSTox database of similar to 875,000 chemicals, and their predicted AR activities have been made available on the EPA CompTox Chemicals dashboard and National Toxicology Program's Integrated Chemical Environment.

    • The development of a PEG-decorated lipid-polymer hybrid system camouflaged with natural and synthetic chemotherapeutic moieties is an influential approach melding the biomimetic properties of long-circulating vesicles to utilize different mechanisms to dwindle the tumor growth. Therefore, a safe and efficient lipid-coated nano-particulate system (LCNPs) was proposed to investigate the in-vitro, ex-vivo and in-vivo demeanors of such amalgamation.Docetaxel loaded PLGA nanoparticles (DTX-NPs) were prepared by solvent evaporation while curcumin liposomes were mapped out using the film hydration method. Physicochemical characterizations were executed in terms of size, surface morphology, differential scanning calorimetry (DSC) and fourier-transform infrared spectroscopy (FTIR). In-vitro cytotoxicity was effectuated using MCF-7 cell line. Hemolysis, erythrocyte aggregation and acute in-vivo toxicity were carried out to establish the biocompatibility. The hydrodynamic diameters of samples were in the nano-range and corresponded to the findings of scanning electron microscopy (SEM) and atomic force microscopy (AFM). The absence of distinctive peaks of DTX-NPs in FTIR and DSC analysis of LCNPs depicts the shielding of the lipid bilayer over the nanoparticle. Cytotoxicity induced by the LCNPs represented the efficient delivery of cargo to the tumor cells. LCNPs also exhibited the least toxicity under ex-vivo and in-vivo circumstances compared to free drugs. Additionally, histological studies showed no evidence of substantial necrosis. Additionally, histological studies showed no evidence of notable necrosis.
