  • Particle size : A missing factor in risk assessment of human exposure to toxic chemicals in settled indoor dust

    • For researches on toxic chemicals in settled indoor dust, selection of dust fraction is a critical influencing factor to the accuracy of human exposure risk assessment results. However, analysis of the selection of dust fraction in recent studies revealed that there is no consensus. This study classified and presented researches on distribution of toxic chemicals according to dust particle size and on relationship between dust particle size and human exposure possibility. According to the literature, beyond the fact that there were no consistent conclusions on particle size distribution of adherent fraction, dust with particle size less than 100 mu m should be paid more attention and that larger than 250 mu m is neither adherent nor proper for human exposure risk assessment. Calculation results based on literature data show that with different selections of dust fractions, analytical results of toxic chemicals would vary up to 10-fold, which means that selecting dust fractions arbitrarily will lead to large errors in risk assessment of human exposure to toxic chemicals in settled dust. Taking into account the influence of dust particle size on risk assessment of human exposure to toxic chemicals, a new methodology for risk assessment of human exposure to toxic chemicals in settled indoor dust is proposed and human exposure parameter systems to settled indoor dust are advised to be established at national and regional scales all over the world.

    • IntroductionEthylenediamine-grafted multiwalled carbon nanotubes (MWCNTs-EDA-I and MWCNTs-EDA-II) are optimized and employed to investigate the preconcentration of lead ions (Pb(II)) in trace level.ResultsThe results show that Pb(II) can be adsorbed quantitatively on the optimized MWCNTs in the range of pH 4–7 and MWCNTs-EDA-I has a higher maximum Pb(II) adsorption capacity (157.19 mg/g) than MWCNTs-EDA-II (89.16 mg/g). The adsorbed Pb(II) can be eluted completely using 5 mL of 1 mol/L HNO3.DiscussionA new approach using a microcolumn packed with the obtained MWCNTs-EDA-I has been developed for the preconcentration of trace amount of Pb(II). Parameters influencing the preconcentration of Pb(II), such as pH of the sample, sample volume, elution solution, and interfering ions, have been examined and optimized in detail. Under optimum experimental conditions, the limit of detection is 0.30 ng/mL with the enrichment factor of 60. The relative standard deviation (R.S.D) was 2.6% at the 20 ng/mL Pb(II) level.ConclusionThe method has been applied for the preconcentration of trace amount of Pb(II) in environmental water samples with satisfying results.

  • Arabidopsis STO/BBX24 negatively regulates UV-B signaling by interacting with COP1 and repressing HY5 transcriptional activity

    • UV-B (280-315 nm) is an integral part of solar radiation and can act either as a stress inducer or as a developmental signal. In recent years, increasing attention has been paid to the low-fluence UV-B-induced photomorphogenic response and several key players in this response have been identified, which include UVR8 (a UV-B-specific photoreceptor), COP1 (a WD40-repeat-containing RING finger protein), HY5 (a basic zipper transcription factor), and RUP1/2 (two UVR8-interacting proteins). Here we report that Arabidopsis SALT TOLERANCE (STO/BBX24), a known regulator for light signaling in plants, defines a new signaling component in UV-B-mediated photomorphogenesis. The bbx24 mutant is hypersensitive to UV-B radiation and becomes extremely dwarfed under UV-B treatment. By contrast, BBX24 overexpression transgenic lines respond much more weakly to UV-B than the bbx24 and wild-type plants. BBX24 expression is UV-B-inducible and its accumulation under UV-B requires COP1. Co-immunoprecipitation experiments indicate that BBX24 interacts with COP1 in planta upon UV-B illumination. Moreover, BBX24 interacts with HY5 and acts antagonistically with HY5 in UV-B-induced inhibition of hypocotyl elongation. Furthermore, BBX24 attenuates UV-B-induced HY5 accumulation and suppresses its transcription-activation activity. Taken together, our results reveal a previously uncharacterized function of the light-regulated BBX24 in UV-B responses and demonstrate that BBX24 functions as a negative regulator of photomorphogenic UV-B responses by interacting with both COP1 and HY5. The UV-B-inducible expression pattern and its suppression of HY5 activity suggest that BBX24 could be a new component of the feedback regulatory module of UV-B signaling in plants.

    • Daylight UV-B (UV-B) radiation (280–315 nm) is, because of its photochemical effects and potential destructive impact, an important environmental factor for plants. After decades of fruitless attempts, a receptor molecule, UVR8, for sensing of ambient UV-B radiation by plants has been characterized, and the initial steps in signal transduction have been identified. There are, however, other signaling pathways, and there are apparent contradictions in the literature. There is still much to find out about the complex signaling network in plants for processing of information about the daylight surrounding them.

  • Establishment and assessments of a new model for the postoperative fatigue syndrome by major small intestinal resection in rats

    • Objective. Postoperative fatigue syndrome (POFS) is a general and main complication after surgery. However, there is no stable and standardized animal model for POFS. The aim of the present study was to establish a rodent model of POFS by small intestinal resection, with POFS evaluated by acknowledged physical and behavioral methods. Material and Methods. Forty-two Sprague-Dawley rats were randomly divided into four groups according to the length of a "middle" small intestinal resection: 0% (sham group; i.e., laparotomy alone), 10%, 40% and 70% groups, with corresponding lengths of small intestinal resections. Following surgery, the general state of health was evaluated. Tail suspension test, open field test and Morris water maze test were used to evaluate the degree of POFS. Serum albumin, transferrin, prealbumin and fibronectin were measured to assess the nutritional status, and superoxide dismutase (SOD) and malondialdehyde (MDA) were also measured. Results. As compared with the other three groups, the 70% small intestinal resection group showed the worst general state of health, decreased strength of the tail suspension test and decreased score of Morris water maze test (p < 0.05) after operation. All rats in whom the small intestinal resection was done demonstrated a certain degree of malnutrition and behavior of depression, and the 70% resection group had the lowest levels of transferrin, prealbumin and fibronectin as compared with the other groups (p < 0.05), as well as decreased SOD and increased MDA in serum (p < 0.05). Conclusions. Resection of 70% of the small intestine resulted in typical characteristics of POFS. As this procedure is simple, stable and easily reproducible, it may serve as a model for research on POFS.
