    • Elevated As concentrations in groundwater of the Huhhot basin (HB), Inner Mongolia, China, and the western Bengal basin (WBB), India, have been known for decades. However, few studies have been performed to comprehend the processes controlling overall groundwater chemistry in the HB. In this study, the controls on solute chemistry in the HB have been interpreted and compared with the well-studied WBB, which has a very different climate, physiography, lithology, and aquifer characteristics than the HB. In general, there are marked differences in solute chemistry between HB and WBB groundwaters. Stable isotopic signatures indicate meteoric recharge in the HB in a colder climate, distant from the source of moisture, in comparison to the warm, humid WBB. The major-ion composition of the moderately reducing HB groundwater is dominated by a mixed-ion (Ca-Na-HCO3-Cl) hydrochemical facies with an evolutionary trend along the regional hydraulic gradient. Molar ratios and thermodynamic calculations show that HB groundwater has not been affected by cation exchange, but is dominated by weathering of feldspars (allitization) and equilibrium with gibbsite and anorthite. Mineral weathering and mobilization of As could occur as recharging water flows through fractured, argillaceous, metamorphic or volcanic rocks in the adjoining mountain-front areas, and deposits solutes near the center of the basin. In contrast, WBB groundwater is Ca-HCO3-dominated, indicative of calcite weathering, with some cation exchange and silicate weathering (monosiallitization).

  • IL-6 Regulates MMP-10 Expression via JAK2/STAT3 Signaling Pathway in a Human Lung Adenocarcinoma Cell Line

    • We previously reported that matrix metalloproteinase (MMP)-10 mRNA levels were significantly lower in tumor tissues than in adjacent normal tissues in human non-small cell lung cancer (NSCLC), whereas protein levels of MMP-10 were higher in the tumor tissues than the adjacent tissues. The mechanism of this divergence is still unknown. In the present stud), the role of Janus kinase 2/signal transducers and activators of transcription 3 (JAK2/STAT3) on interleukin (IL)-6 mediated regulation of MMP-10 expression was investigated in a human lung adenocarcinoma cell line (A549 cells) and the molecular regulatory mechanism of MMP-10 expression was explored. A549 cells were stimulated by different concentrations of IL-6 with or without AG490, a specific JAK2 inhibitor. It was demonstrated that IL-6 moderately reduced the MMP-10 mRNA levels, whereas it significantly enhanced the MMP-10 protein mass in the A549 cells. This phenomenon mimicked the divergence of mRNA level and protein mass of MMP-10 in human NSCLC. Moreover, the present study indicated that IL-6 regulation of MMP-10 expression was via the JAK2/STAT3 pathway. STAT3 mRNA levels were significantly increased when the cells were treated with IL-6, whereas when AG490 (50 mu M) was added to the cell cultures, IL-6-induced increase of STAT3 mRNA levels was abolished. Meanwhile, AG490 blocked the IL-6-induced inhibition of MMP-10 mRNA as well as blocking the IL-6-induced increase of MMP-10 protein mass in the A549 cells. Neither IL-6 nor AG490 influenced JAK2 mRNA levels in the A549 cell cultures. It is concluded that the JAK2/STAT3 pathway is involved in the IL-6-mediated regulation of MMP-10, and IL-6 can moderately reduce MMP-10 mRNA levels and strongly increase MMP-10 protein mass in human lung adenocarcinoma A549 cells. Contrasting effects of IL-6 on MMP-10 mRNA level and protein concentration in A549 cells may partially explain the divergence of MMP-10 mRNA level and protein mass in human NSCLC.

    • Mass fabrication of directly accessible, ultralong, uniform Si nanowires is realized by employing a controllable and reproducible method based on standard Si technology. High-conductivity polycrystalline Ni-silicide nanowires around 30 nm by 30 nm in cross section, able to support extremely high currents at similar to 10(8) A/cm(2), are obtained by means of solid-state reaction of the Si nanowires with subsequently deposited Ni films. By properly adjusting the Ni film thickness, NiSi, Ni2Si, and Ni31Si12 nanowires characterized with distinct resistivity and temperature coefficient of resistance are obtained. Upon annealing, the electrical continuity of the nanowires breaks at temperatures about 0.7 times the melting points of the silicides.

  • Family-based association study of the MCF2L2 gene and polycystic ovary syndrome

    • <i>Objective:</i> The aim of the study was to determine the association between three single nucleotide polymorphism (SNP) variants (rs35368790, rs35069869 and rs684846) of the MCF2 cell line-derived transforming sequence-like 2 <i>(MCF2L2)</i> gene and polycystic ovary syndrome (PCOS) in PCOS family trios. <i>Methods:</i> Genotyping was done by TaqMan assay that incorporates minor groove-binding probe technology for allelic discrimination. One hundred and fifty-two unrelated PCOS probands and their biological parents were recruited. All subjects were of Han Chinese origin and from Shandong Province. <i>Results:</i> The transmission disequilibrium test (TDT) for allelic association demonstrated that a weak association was detected in SNP rs35368790 with p = 0.008. However, we found no significant transmission distortion of the other two SNPs (rs35069869, χ<sup>2</sup> = 3.645, p = 0.056; rs684846, χ<sup>2</sup> = 1.429, p = 0.232, respectively). <i>Conclusions:</i> These results suggest that the genetic polymorphisms within <i>MCF2L2</i> are likely to confer an increased susceptibility to PCOS in the Chinese population. Our present data may provide a basis for further studies of the role of the <i>MCF2L2</i> gene in the etiology of PCOS.
