  • Regulating the production of (R)-3-hydroxybutyrate in Escherichia coli by N or P limitation

    • The chiral compound (R)-3-hydroxybutyrate (3HB) is naturally produced by many wild type organisms as the monomer for polyhydroxybutyrate (PHB). Both compounds are commercially valuable and co-polymeric polyhydroxyalkanoates have been used e.g., in medical applications for skin grafting and as components in pharmaceuticals. In this paper we investigate cultivation strategies for production of 3HB in the previously described E. coil strain AF1000 pJBGT3RX. This strain produces extracellular 3HB by expression of two genes from the PHB pathway of Halomonas boliviensis. H. boliviensis is a newly isolated halophile that forms PHB as a storage compound during carbon excess and simultaneous limitation of another nutrient like nitrogen and phosphorous. We hypothesize that a similar approach can be used to control the flux from acetylCoA to 3HB also in E coli; decreasing the flux to biomass and favoring the pathway to the product. We employed ammonium- or phosphate-limited fed-batch processes for comparison of the productivity at different nutrient limitation or starvation conditions. The feed rate was shown to affect the rate of glucose consumption, respiration, 3HB, and acetic acid production, although the proportions between them were more difficult to affect. The highest 3HB volumetric productivity, 1.5 g L-1 h(-1), was seen for phosphate-limitation.

  • The effect of social protection and income maintenance policies on health and health inequalities

    • BackgroundAs a starting point we review the existing evidence on welfare states, health and health inequalities, from 2005 onwards. Three different approaches emerge in these previous studies – the welfare regime approach, the welfare institutions approach and the social spending approach. While no clear picture emerges for the welfare regime approach, summarising findings regarding the institutional and expenditure approach suggest that a higher degree of generosity and social spending benefits public health. These are therefore approaches we follow to arrive at a better understanding of what type of policies are linked to smaller inequalities in health across the life-course.MethodsA starting point in the analyses is the relation between income, poverty and mortality. The cross-national variation in poverty rates, both absolute (poverty threshold) and relative (60 per cent of median income) measure, and mortality rates in European 26 countries will be considered.The second step in the analysis focuses on the relationship between social rights and subjective health in Europe, with a focus on national variations and changes in social rights to levels and changes in subjective health outcomes across several countries. The data holds information regarding social rights and social expenditure, including individual data from EU-SILC.ResultsPreliminary results indicate that it is the totality of social protection that is important rather than individual policies. A sub-study regarding social rights and health among youth highlight also the importance of active and passive labour market policy in the 16 included countries.ConclusionsIn sum our diverse approach to analysing welfare state efforts and their links to health inequalities suggest that there is a clear relationship between more ambitious policies and smaller inequalities in health. These results are discussed in relation to previous findings.

  • Recovering Whooper Swans do not cause a decline in Eurasian Wigeon via their grazing impact on habitat

    • The Whooper Swan (Cygnus cygnus) is a good example of successful conservation, with rapidly growing numbers in Fennoscandia in recent decades. To the contrary, Eurasian Wigeon (Mareca penelope) shows a strong negative trend in breeding numbers, which raises conservation concerns. Previous research suggests a causal link between recent population trajectories of the two species. Both preferentially breed on wetlands with abundant horsetail (Equisetum spp.), a plant providing food for Whooper Swan and crucial feeding microhabitat for Eurasian Wigeon broods. We here test predictions based on the hypothesis that grazing on Equisetum by Whooper Swan reduces breeding habitat or breeding habitat quality for Eurasian Wigeon. We use data from 60 lakes in which waterfowl were counted in 1990–1991 and 2016, and Equisetum was mapped in 1990–1991 and 2013–2014. Lakes colonized by Whooper Swan typically had more abundant Equisetum vegetation in the past than lakes not colonized. Lake-specific decrease of Equisetum was not associated with colonization by Whooper Swan. The number of lakes occupied by Eurasian Wigeon decreased, but the decrease was not stronger on lakes colonized by Whooper Swan than on those that were not. Contrary to our prediction, current Eurasian Wigeon abundance was positively associated with Whooper Swan abundance. Moreover, Eurasian Wigeon did not decrease more on lakes from which Equisetum disappeared than on lakes in which there was still Equisetum left. This study does not support the idea that Whooper Swan affects Eurasian Wigeon negatively by grazing on Equisetum.

  • Utilizing venous occlusion plethysmography to assess vascular effects: A study with buloxibutid, an angiotensin II type 2 receptor agonist

    • Buloxibutid (also known as C21) is a potent and selective angiotensin II type 2 receptor (AT2R) agonist, in development for oral treatment of fibrotic lung disease. This phase I, open-label, pharmacodynamic study investigated vascular effects of buloxibutid in five healthy male volunteers. Subjects were administered intra-arterial infusions of buloxibutid for 5 min in ascending doses of 3, 10, 30, 100, and 200 mu g/min, infused sequentially in the forearm. Infusions of sodium nitroprusside (SNP) solution in doses of 0.8-3.2 mu g/min were administered as a positive control. Forearm blood flow (FBF) was measured by venous occlusion plethysmography. Safety and tolerability of intra-arterial administrations of buloxibutid were evaluated. Following infusion of buloxibutid in doses of 3-200 mu g/min, the range of increase in FBF was 27.8%, 17.2%, 37.0%, 28.5%, and 60.5%, compared to the respective baseline. The largest increase was observed in the highest dose group. Infusions of SNP as a positive control, increased FBF 230-320% compared to baseline. Three adverse events (AEs) of mild intensity, not related to buloxibutid or SNP, were reported for two subjects. Two of these AEs were related to study procedures. There were no clinically relevant changes in arterial blood pressure during the study period. Intra-arterial infusion of buloxibutid in low, ascending doses increased FBF, indicating that buloxibutid may be effective in conditions associated with endothelial dysfunction. Venous occlusion plethysmography was found to be a useful method to explore pharmacodynamic vascular effects of novel AT2R agonists, while avoiding systemic adverse effects.

  • A Measurement Based Shadow Fading Model for Vehicle-to-Vehicle Network Simulations

    • The vehicle-to-vehicle (V2V) propagation channel has significant implications on the design and performance of novel communication protocols for vehicular ad hoc networks (VANETs). Extensive research efforts have been made to develop V2V channel models to be implemented in advanced VANET system simulators for performance evaluation. The impact of shadowing caused by other vehicles has, however, largely been neglected in most of the models, as well as in the system simulations. In this paper we present a shadow fading model targeting system simulations based on real measurements performed in urban and highway scenarios. The measurement data is separated into three categories, line-of-sight (LOS), obstructed line-of-sight (OLOS) by vehicles, and non-line-of-sight due to buildings, with the help of video information recorded during the measurements. It is observed that vehicles obstructing the LOS induce an additional average attenuation of about 10 dB in the received signal power. An approach to incorporate the LOS/OLOS model into existing VANET simulators is also provided. Finally, system level VANET simulation results are presented, showing the difference between the LOS/OLOS model and a channel model based on Nakagami-m fading.

  • Statistical Sensor Calibration Algorithms

    • The use of sensors is ubiquitous in our IT-based society; smartphones, consumer electronics, wearable devices, healthcare systems, industries, and autonomous cars, to name but a few, rely on quantitative measurements for their operations. Measurements require sensors, but sensor readings are corrupted not only by noise but also, in almost all cases, by deviations resulting from the fact that the characteristics of the sensors typically deviate from their ideal characteristics.This thesis presents a set of methodologies to solve the problem of calibrating sensors with statistical estimation algorithms. The methods generally start with an initial statistical sensor modeling phase in which the main objective is to propose meaningful models that are capable of simultaneously explaining recorded evidence and the physical principle for the operation of the sensor. The proposed calibration methods then typically use training datasets to find point estimates of the parameters of these models and to select their structure (particularlyin terms of the model order) using suitable criteria borrowed from the system identification literature. Subsequently, the proposed methods suggest how to process the newly arriving measurements through opportune filtering algorithms that leverage the previously learned models to improve the accuracy and/or precision of the sensor readings.This thesis thus presents a set of statistical sensor models and their corresponding model learning strategies, and it specifically discusses two cases: the first case is when we have a complete training dataset (where “complete” refers to having some ground-truth informationin the training set); the second case is where the training set should be considered incomplete (i.e., not containing information that should be considered ground truth, which implies requiring other sources of information to be used for the calibration process). In doing so, we consider a set of statistical models consisting of both the case where the variance of the measurement error is fixed (i.e., homoskedastic models) and the case where the variance changes with the measured quantity (i.e., heteroskedastic models). We further analyzethe possibility of learning the models using closed-form expressions (for example, when statistically meaningful, Maximum Likelihood (ML) and Weighted Least Squares (WLS) estimation schemes) and the possibility of using numerical techniques such as Expectation Maximization (EM) or Markov chain Monte Carlo (MCMC) methods (when closed-form solutions are not available or problematic from an implementation perspective). We finally discuss the problem formulation using classical (frequentist) and Bayesian frameworks, and we present several field examples where the proposed calibration techniques are applied on sensors typically used in robotics applications (specifically, triangulation Light Detection and Rangings (Lidars) and Time of Flight (ToF) Lidars).

  • Optimal Design in Geodetic GNSS-based Networks

    • An optimal design of a geodetic network helps the surveying engineers maximise the efficiency of the network. A number of pre-defined quality requirements, i.e. precision, reliability, and cost, of the network are fulfilled by performing an optimisation procedure. Today, this is almost always accomplished by implementing analytical solutions, where the human intervention in the process cycle is limited to defining the requirements. Nevertheless, a trial and error method can be beneficial to some applications. In order to analytically solve an optimisation problem, it can be classified to different orders, where an optimal datum, configuration, and optimal observation weights can be sought such that the precision, reliability and cost criteria are satisfied.In this thesis, which is a compilation of six peer-reviewed papers, we optimised and redesigned a number of GNSS-based monitoring networks in Sweden by developing new methodologies. In addition, optimal design and efficiency of total station establishment with RTK-GNSS is investigated in this research.Sensitivity of a network in detecting displacements is of importance for monitoring purposes. In the first paper, a precision criterion was defined to enable a GNSS-based monitoring network to detect 5 mm displacements at each network point. Developing an optimisation model by considering this precision criterion, reliability and cost yielded a decrease of 17% in the number of observed single baselines implying a reliable and precise network at lower cost. The second paper concerned a case, where the precision of observations could be improved in forthcoming measurements. Thus a new precision criterion was developed to consider this assumption. A significant change was seen in the optimised design of the network for subsequent measurements. As yet, the weight of single baselines was subject to optimisation, while in the third paper, the effect of mathematical correlations between GNSS baselines was considered in the optimisation. Hence, the sessions of observations, including more than two receivers, were optimised. Four out of ten sessions with three simultaneous operating receivers were eliminated in a monitoring network with designed displacement detection of 5 mm. The sixth paper was the last one dealing with optimisation of GNSS networks. The area of interest was divided into a number of three-dimensional elements and the precision of deformation parameters was used in developing a precision criterion. This criterion enabled the network to detect displacements of 3 mm at each point.A total station can be set up in the field by different methods, e.g. free station or setup over a known point. A real-time updated free station method uses RTK-GNSS to determine the coordinates and orientation of a total station. The efficiency of this method in height determination was investigated in the fourth paper. The research produced promising results suggesting using the method as an alternative to traditional levelling under some conditions. Moreover, an optimal location for the total station in free station establishment was studied in the fifth paper. It was numerically shown that the height component has no significant effect on the optimal localisation.

    • Precision, reproducibility and lower limit of quantitation (LLOQ) are important characteristics of a quantitative method. We have investigated these properties for Ximelagatran (Xi), which has a high tendency to form doubly charged ions in electrospray ionization (ESI), by studying the percentage of doubly charged species formed when varying the formic acid (FA) concentration, analyte concentration, amount of organic modifier and flow rate. It was found that the percentage of [Xi + 2H]2+ can be controlled to be more than 90% or less than 10% by varying the amount of FA present, and that the change between these values is dramatic. Furthermore, the percentage of [Xi + 2H]2+ formed decreases with increased analyte concentration and increased flow rate. No apparent relationship with the amount of organic modifier was found. The results have the implication that, by carefully controlling the selected parameters, the LLOQ, precision and reproducibility can be improved. We have compared the fragmentation of the singly and doubly charged species and concluded that the [Xi + 2H]2+ ion is more inclined to undergo fragmentation than [Xi + H]+. As a consequence, unusual instrumental settings had to be used for the experiments. The fragmentation patterns are to a great extent similar, but the doubly charged species is more inclined to generate low-mass product ions.
