  • Radiation damage in protein serial femtosecond crystallography using an x-ray free-electron laser
  • 2011
  • Ingår i: Physical Review B. Condensed Matter and Materials Physics. - 1098-0121 .- 1550-235X. ; 84:21, s. 214111-1-214111-6
  • Tidskriftsartikel (refereegranskat)abstract
    • X-ray free-electron lasers deliver intense femtosecond pulses that promise to yield high resolution diffraction data of nanocrystals before the destruction of the sample by radiation damage. Diffraction intensities of lysozyme nanocrystals collected at the Linac Coherent Light Source using 2 keV photons were used for structure determination by molecular replacement and analyzed for radiation damage as a function of pulse length and fluence. Signatures of radiation damage are observed for pulses as short as 70 fs. Parametric scaling used in conventional crystallography does not account for the observed effects.

  • CUBN Is a Gene Locus for Albuminuria
  • 2011
  • Ingår i: Journal of the American Society of Nephrology. - 1046-6673 .- 1533-3450. ; 22:3, s. 555-570
  • Tidskriftsartikel (refereegranskat)abstract
    • Identification of genetic risk factors for albuminuria may alter strategies for early prevention of CKD progression, particularly among patients with diabetes. Little is known about the influence of common genetic variants on albuminuria in both general and diabetic populations. We performed a meta-analysis of data from 63,153 individuals of European ancestry with genotype information from genome-wide association studies (CKDGen Consortium) and from a large candidate gene study (CARe Consortium) to identify susceptibility loci for the quantitative trait urinary albumin-to-creatinine ratio (UACR) and the clinical diagnosis microalbuminuria. We identified an association between a missense variant (I2984V) in the CUBN gene, which encodes cubilin, and both UACR (P = 1.1 x 10(-11)) and microalbuminuria (P = 0.001). We observed similar associations among 6981 African Americans in the CARe Consortium. The associations between this variant and both UACR and microalbuminuria were significant in individuals of European ancestry regardless of diabetes status. Finally, this variant associated with a 41% increased risk for the development of persistent microalbuminuria during 20 years of follow-up among 1304 participants with type 1 diabetes in the prospective DCCT/EDIC Study. In summary, we identified a missense CUBN variant that associates with levels of albuminuria in both the general population and in individuals with diabetes.

  • Rapport (övrigt vetenskapligt/konstnärligt)abstract
    • It is the main objective of the ARTIST4G Work Package 1 (WP1), to design innovative interference avoidance solutions by means of transmitter signal processing, scheduling and/or cross layer techniques. Such an approach will enable to identify optimal strategies also taking into account the practical implications on the real system. The major impacts of the WP1 innovations on the radio access network and on the mobile system interfaces, together with constraints due to practical implementations, will be analyzed in this deliverable. This deliverable will be a reference for the work in ARTIST4G Work Package 4 (WP4).A set of guiding questions used to precise the requirements for WP4 has been considered for each new cooperation/coordination functionality. Answers to these questions are provided in this deliverable by grouping the different classes of WP1 innovations into 4 categories with similar requirements on the radio access architecture. For each question, current, expected, minimum and maximum targets of the requirement will be given, helping to ensure that the range of all possible values for all the innovations are taken into account.

  • 2012
  • Rapport (övrigt vetenskapligt/konstnärligt)abstract
    • In this document we provide performance assessments of the most promising techniques that were studied within Work Package 1 (WP1) of the ARTIST4G project related to interference avoidance. The results are based on evolved techniques that were identified and classified in deliverable D1.1 and investigated in deliverable D1.2 and D1.3, as well as novel alternative techniques that are introduced and assessed in this document.Based on the insights from these performance results we provide a synthetic perspective over the most promising solutions for interference avoidance. We argue that these solutions can reach a satisfactory trade-off in terms of performance benefits vs complexity of implementation. Some of these techniques are also identified as complementary techniques towards an integrated interference avoidance concept.

  • Genetically Distinct Subsets within ANCA-Associated Vasculitis
  • 2012
  • Ingår i: New England Journal of Medicine. - : Massachusetts Medical Society. - 0028-4793 .- 1533-4406. ; 367:3, s. 214-223
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND less thanbrgreater than less thanbrgreater thanAntineutrophil cytoplasmic antibody (ANCA)-associated vasculitis is a severe condition encompassing two major syndromes: granulomatosis with polyangiitis (formerly known as Wegeners granulomatosis) and microscopic polyangiitis. Its cause is unknown, and there is debate about whether it is a single disease entity and what role ANCA plays in its pathogenesis. We investigated its genetic basis. less thanbrgreater than less thanbrgreater thanMETHODS less thanbrgreater than less thanbrgreater thanA genomewide association study was performed in a discovery cohort of 1233 U. K. patients with ANCA-associated vasculitis and 5884 controls and was replicated in 1454 Northern European case patients and 1666 controls. Quality control, population stratification, and statistical analyses were performed according to standard criteria. less thanbrgreater than less thanbrgreater thanRESULTS less thanbrgreater than less thanbrgreater thanWe found both major-histocompatibility-complex (MHC) and non-MHC associations with ANCA-associated vasculitis and also that granulomatosis with polyangiitis and microscopic polyangiitis were genetically distinct. The strongest genetic associations were with the antigenic specificity of ANCA, not with the clinical syndrome. Anti-proteinase 3 ANCA was associated with HLA-DP and the genes encoding alpha(1)-antitrypsin (SERPINA1) and proteinase 3 (PRTN3) (P = 6.2x10(-89), P = 5.6x10(-12), and P = 2.6x10(-7), respectively). Anti-myeloperoxidase ANCA was associated with HLA-DQ (P = 2.1x10(-8)). less thanbrgreater than less thanbrgreater thanCONCLUSIONS less thanbrgreater than less thanbrgreater thanThis study confirms that the pathogenesis of ANCA-associated vasculitis has a genetic component, shows genetic distinctions between granulomatosis with polyangiitis and microscopic polyangiitis that are associated with ANCA specificity, and suggests that the response against the autoantigen proteinase 3 is a central pathogenic feature of proteinase 3 ANCA-associated vasculitis. These data provide preliminary support for the concept that proteinase 3 ANCA-associated vasculitis and myeloperoxidase ANCA-associated vasculitis are distinct autoimmune syndromes. • Animal snoRNAs and scaRNAs with exceptional structures
  • 2011
  • Ingår i: RNA Biology. - : Informa UK Limited. - 1547-6286 .- 1555-8584. ; 8:6, s. 938-946
  • Tidskriftsartikel (refereegranskat)abstract
    • The overwhelming majority of small nucleolar RNAs (snoRNAs) fall into two clearly defined classes characterized by distinctive secondary structures and sequence motifs. A small group of diverse ncRNAs, however, shares the hallmarks of one or both classes of snoRNAs but differs substantially from the norm in some respects. Here, we compile the available information on these exceptional cases, conduct a thorough homology search throughout the available metazoan genomes, provide improved and expanded alignments, and investigate the evolutionary histories of these ncRNA families as well as their mutual relationships.

  • Synthesis, Structure, and Electronic Properties of RuN(6) Dinuclear Ru-Hbpp Complexes
  • 2012
  • Ingår i: Inorganic Chemistry. - : American Chemical Society (ACS). - 0020-1669 .- 1520-510X. ; 51:1, s. 320-327
  • Tidskriftsartikel (refereegranskat)abstract
    • A series of RuN(6) dinuclear Ru-Hbpp complexes (Hbpp is the dinucleating tetraaza ligand 3,5-bis(pyridyl)pyrazole) of general formula {[Ru(II)(R(2)-trpy)(MeCN)](2)(mu-R(1)-bpp)}3(+), 10(3+)-14(3+), (R(1) = H, Me, or NO(2). and R(2) = H, Me, MeO; see Scheme 1) has been prepared from their Cl(-) or AcO(-) bridged precursors. The complexes have been characterized by UV-vis, NMR, CV, and some by X-ray. Complexes 10(3+)-14(3+), Ru(2)(II,II), were oxidized by 1 equiv in solution, leading to the mixed valence Ru(2)(II,III) complexes 10(4+)-14(4+) containing one unpaired electron and were characterized by EPR and UV-vis-near-IR, which showed metal-centered spin and the presence of low-energy IVCT bands. The H(ab) parameter indicates a relatively strong electronic coupling between the two ruthenium centers (class II). Further two electron oxidation in solution of the 10(3+)-14(3+) led to the formation of EPR silent Ru(2)(II,III) complexes 10(5+)-14(5+), that were further characterized by UV-vis-NIR TD-DFT calculations are employed to assign the nature of the UV-vis transitions for the complexes in the various oxidation states, which are of metal to ligand charge transfer (MLCT) type for Ru(2)(II,II) and ligand to,m. metal charge transfer (LMCT) type for Ru(2)(III,II) and Ru(2)(III,III).
