  • Overview of the JET preparation for deuterium-tritium operation with the ITER like-wall

    • For the past several years, the JET scientific programme (Pamela et al 2007 Fusion Eng. Des. 82 590) has been engaged in a multi-campaign effort, including experiments in D, H and T, leading up to 2020 and the first experiments with 50%/50% D-T mixtures since 1997 and the first ever D-T plasmas with the ITER mix of plasma-facing component materials. For this purpose, a concerted physics and technology programme was launched with a view to prepare the D-T campaign (DTE2). This paper addresses the key elements developed by the JET programme directly contributing to the D-T preparation. This intense preparation includes the review of the physics basis for the D-T operational scenarios, including the fusion power predictions through first principle and integrated modelling, and the impact of isotopes in the operation and physics of D-T plasmas (thermal and particle transport, high confinement mode (H-mode) access, Be and W erosion, fuel recovery, etc). This effort also requires improving several aspects of plasma operation for DTE2, such as real time control schemes, heat load control, disruption avoidance and a mitigation system (including the installation of a new shattered pellet injector), novel ion cyclotron resonance heating schemes (such as the three-ions scheme), new diagnostics (neutron camera and spectrometer, active Alfven eigenmode antennas, neutral gauges, radiation hard imaging systems...) and the calibration of the JET neutron diagnostics at 14 MeV for accurate fusion power measurement. The active preparation of JET for the 2020 D-T campaign provides an incomparable source of information and a basis for the future D-T operation of ITER, and it is also foreseen that a large number of key physics issues will be addressed in support of burning plasmas.

  • The amyloid-β precursor protein (APP)-binding protein Fe65 and APP processing

    • Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by abnormal deposition of neurotoxic amyloid-β (Aβ) peptide. Aβ is generated by sequential cleavage of the amyloid-β precursor protein (APP) by β- and then γ-secretase. However, APP can also be processed by α- and γ-secretase, instead resulting in generation of neuroprotective sAPPα. Increased APP phosphorylation and altered expression levels of the brain enriched Fe65 protein have been observed in the brains of AD patients. Fe65 can not only interact with membrane tethered APP, but can also localized into the nucleus and act as a transcriptional regulator together with the APP intracellular domain (AICD), generated after γ-secretase processing. How APP processing, APP/Fe65 interaction, and the nuclear AICD/Fe65 complex is regulated has not yet been fully understood. The aim of this thesis was therefore to further elucidate how Fe65 is regulated and how APP Ser675 phosphorylation affects APP processing.We could identify several factors regulating Fe65. First, we identified that neuronal differentiation induces Fe65 phosphorylation (paper I), and that phosphorylated forms of Fe65 were preferentially localized outside the nucleus (paper II). Second, we found that the APP binding PTB2 domain of Fe65, rather than the previously proposed N-terminal WW domain, is important for the nuclear localization of Fe65 (paper II). In addition, we surprisingly found that mutation of S228 in the Fe65 N-terminus could increase the APP/Fe65 interaction (paper III). Third, both α- and γ-secretase inhibitors decreased Fe65 nuclear localization similarly, indicating an important role of α-secretase in regulating Fe65 nuclear localization (papers II and III). Lastly, we could in paper IV for the first time show that phosphorylation of APP at Ser675 regulates APP processing at the plasma membrane, resulting in reduced levels of sAPPα. These results, together with the observation that APP Ser675 phosphorylation occur in AD brains, suggest that Ser675 phosphorylation could contribute to AD pathology by decreasing α-secretase processing and instead increasing the levels of Aβ.In summary these studies have contributed to understanding of APP processing and the interplay between Fe65 and APP, two suggested key players in AD.

    • The addition of gelling polysaccharides to sport-drinks may provide improved tolerability of drinks with high concentration of digestible carbohydrates (CHO), otherwise known to increase the risk of gastro-intestinal complaints among athletes under prolonged exercise. The physico-chemical properties of a drink containing 14 wt% of digestible CHO (0.7:1 fructose and maltodextrin-ratio), 0.2 wt% of HM-pectin/alginate and 0.06 wt%. sodium chloride were examined under in vitro gastric conditions using rheology and large deformation testing. The in vivo gelling behaviour of the drink was studied using magnetic resonance imaging of subjects at rest together with blood glucose measurements. The in vivo results confirm gelation of the test drink, with no gel remaining in the stomach at 60 min and blood glucose values were similar to control. The physico-chemical characterisation of the acidified test drink confirms the formation of a weak gel through which low Mw CHO can diffuse.

    • This deliverable represents the final report on the METIS radio link research. It provides a comprehensive and self-contained summary of all investigated technology components (TeCs), including evaluation results and conclusions on their potential for 5G. The METIS radio link research covers three main areas, which are considered key aspects for developing a self-contained air interface design for 5G: 1) Flexible air interface, 2) Waveforms, coding & modulation and transceiver design and 3) Multiple access, Medium Access Control (MAC) and Radio Resource Management (RRM). TeCs with similar research context and objectives have been grouped into clusters, whereof five have been selected as the most promising for 5G systems: From research area 1, TeC clusters providing key enablers for an air interface for ultra-dense networks (UDN) and for moving networks; from research area 2, multi-carrier transmission schemes with filtering; and from research area 3, novel access schemes for massive machine access as well as for non-orthogonal access.

    • Despite a worldwide increase in the presence of methicillin-resistant Staphylococcus pseudintermedius (MRSP) in dogs and its potential to cause serious canine health problem, the understanding of the transmission and long-term carriage of MRSP is limited. The objective of this study was to investigate the transmission of MRSP to contact dogs living in multiple dog households where one or more of the dogs had been diagnosed with a clinically apparent infection with MRSP. MRSP carriage was investigated over several months in 11 dogs living in four separate multiple dog households where an MRSP infection in a dog had been diagnosed. Whole-genome sequencing was used for genotypic characterization. Contact dogs were only MRSP-positive if the index dog was positive on the same sample occasion. Three contact dogs were consistently MRSP-negative. The data from whole genome sequencing showed similarities between isolates within each family group, indicating that MRSP was transmitted within each family. The results show that the risk of MRSP-colonization in dogs living with an MRSP-infected dog is reduced if the index dog becomes MRSP negative. All of the contact dogs will not carry MRSP continuously during the time the index dog is MRSP-positive. The information yielded from whole genome sequencing showed the methodology to be a promising additional tool in epidemiologic investigations of MRSP transmission.
