  • Complementary methods assessing short and long-term prey of a marine top predator ‒ Application to the grey seal-fishery conflict in the Baltic Sea

    • The growing grey seal (Halichoerus grypus) population in the Baltic Sea has created conflicts with local fisheries, comparable to similar emerging problems worldwide. Adequate information on the foraging habits is a requirement for responsible management of the seal population. We investigated the applicability of available dietary assessment methods by comparing morphological analysis and DNA metabarcoding of gut contents (short-term diet; n = 129/125 seals, respectively), and tissue chemical markers i.e. fatty acid (FA) profiles of blubber and stable isotopes (SIs) of liver and muscle (mid- or long-term diet; n = 108 seals for the FA and SI markers). The methods provided complementary information. Short-term methods indicated prey species and revealed dietary differences between age groups and areas but for limited time period. In the central Baltic, herring was the main prey, while in the Gulf of Finland percid and cyprinid species together comprised the largest part of the diet. Perch was also an important prey in the western Baltic Proper. The DNA analysis provided firm identification of many prey species, which were neglected or identified only at species group level by morphological analysis. Liver SIs distinguished spatial foraging patterns and identified potentially migrated individuals, whereas blubber FAs distinguished individuals frequently utilizing certain types of prey. Tissue chemical markers of adult males suggested specialized feeding to certain areas and prey, which suggest that these individuals are especially prone to cause economic losses for fisheries. We recommend combined analyses of gut contents and tissue chemical markers as dietary monitoring methodology of aquatic top predators to support an optimal ecosystem-based management.

    • The concept of healthy workplace has been defined as an organization that maximizes the integration of worker goals for wellbeing and company objectives for profitability and productivity. Conditions in today’s working life make new approaches necessary in order to limit negative health effects of work and to enhance wellbeing and health at work. About 24 % of the working population in Sweden report to have had work-related disorders during the last twelve months. In order to achieve a sustainable working life it is likely that strategies and actions from different and new angles are needed.ObjectivesThe present study is a part of a larger study (the GodA –study; a Swedish acronym for good work environments and healthy workplaces) and aims to investigate how work environment factors, work ability, work motivation, work and life balance predict well-being at work.MethodsThe GodA study is a 2-year follow up study in Sweden with a survey feedback design in three companies with both blue- and white collar workers. One of the companies serves as “intervention-company”, the other two as controls. A baseline questionnaire was sent out 2013 and the results from the survey were reported back to the companies, which have been processing their results. In spring 2015 a follow up survey has been administered. Data have been analysed with univariate and multivariate linear regression analyses.ResultsA baseline multivariate linear regression model, which included background factors, perceived psychosocial work climate and work environmental factors (motivation, leadership, employee responsibilities, efficacy, work ability and management committed to employee health) and work life balance, showed that psychosocial work climate (B= .48, 95% CI=.27 – .69) leadership, (B= .27, 95% CI=.05– .49), work ability (B= -.12, 95% CI= .03 – .21), motivation (B= -33, 95% CI= .14 – .51) and work life balance (B= -.34, 95% CI=-.57– -.12), were signifi-cantly associated with well-being at work and explained 40% of the variance (Adjusted R2=.40, p<.001).ConclusionsResults showed that not only work environment factors are important predictors. To maintain a healthy work place a promotion of balance between work and private life is needed.

  • Being physically active as an adult with congenital heart disease

    • Background: Due to advances in medical and surgical care adults with congenital heart disease (CHD) is a growing and aging population, that now outnumbers the children with CHD. In general, adults with CHD have reduced aerobic exercise capacity and nearly half of the patients do not reach current recommendations on physical activity. It is known that a low level of physical activity is associated with an increased risk for acquired cardiovascular disease. Studies has shown that adults with CHD are at the same, or even higher risk as the general population, for developing acquired cardiovascular disease.Aim: The overall aim was to explore physical activity in adults with CHD with respect to associated factors, exercise self-efficacy and their own experiences.Methods: This thesis is based on four papers. Paper I included 471 adults with CHD from three tertiary care centres in Sweden. The participants completed questionnaires measuring patient reported outcomes (e.g. SF-12) including physical activity level. Paper II was based on data from 79 adults with CHD from two tertiary care centers in Sweden and 42 matched controls. All participants completed questionnaires on exercise self-efficacy and quality of life, wore an activity monitor during four consecutive days and performed muscle endurance tests. Paper I and II were of cross-sectional design and analyses were done using logistic regression. In paper III and IV data were collected through structured interviews for 14 participants. They were asked about their experiences of being physically active (paper III), what they considered as physical activities, and their experiences of enablers and barriers to physical activity (paper IV). Qualitative content analysis was used in papers III and IV.Results: Physical activity level (paper I) and exercise self-efficacy (paper II) were strongly associated with age where those over 40 years had a lower level of physical activity and lower exercise self-efficacy. Further, in paper I, it appeared that patient reported outcomes from SF-12 were strongly associated with physical activity level. In paper II, exercise self-efficacy was associated with performance in a muscle endurance tests. Paper III revealed an overall theme – It´s like balancing on a slackline that illustrates how adults with CHD described themselves in relation to physical activity. This overall theme consisted of four themes: (1) Being an adventurer – enjoying the challenges of physical activity; (2) Being a realist – adapting to physical ability; (3) Being a non-doer – lacking prerequisites for physical activity and (4) Being an outsider – feeling excluded depending on physical ability. In paper IV, the analysis revealed a description of what adults with CHD consider to be physical activity and considered as enablers and barriers for physical activity. Four categories appeared; physical aspects, psychological aspects, psychosocial aspects and environmental aspects. In the psychosocial aspect, social support and encouragement in childhood to be physically active and no restrictions from e.g. parents, teachers and health care increased physical activity in adulthood.Conclusions: Age, social support and accepting physical limitations seem to have an important impact regarding physical activity level and exercise self-efficacy. In contrast, the complexity of CHD and other medical factors appear to be of less importance for adults with CHD in relation to physical activity. In order to support adults with CHD to increase their physical activity and reach their full potential, it is important to explore and consider the various aspects that may affect physical activity in this population.

  • Magnetic nanobeads present during enzymatic amplification and labeling for a simplified DNA detection protocol based on AC susceptometry

    • Magnetic biosensors are promising candidates for low-cost point-of-care biodiagnostic devices. For optimal efficiency it is crucial to minimize the time and complexity of the assay protocol including target recognition, amplification, labeling and read-out. In this work, possibilities for protocol simplifications for a DNA biodetection principle relying on hybridization of magnetic nanobeads to rolling circle amplification (RCA) products are investigated. The target DNA is recognized through a padlock ligation assay resulting in DNA circles serving as templates for the RCA process. It is found that beads can be present during amplification without noticeably interfering with the enzyme used for RCA (phi29 polymerase). As a result, the bead-coil hybridization can be performed immediately after amplification in a one-step manner at elevated temperature within a few minutes prior to read-out in an AC susceptometer setup, i.e. a combined protocol approach. Moreover, by recording the phase angle xi = arctan(chi ''/chi'), where chi and chi '' are the in-phase and out-of-phase components of the AC susceptibility, respectively, at one single frequency the total assay time for the optimized combined protocol would be no more than 1.5 hours, often a relevant time frame for diagnosis of cancer and infectious disease. Also, applying the phase angle method normalization of AC susceptibility data is not needed. These findings are useful for the development of point-of-care biodiagnostic devices relying on bead-coil binding and magnetic AC susceptometry.

  • Predicting participation in the population-based Swedish cardiopulmonary bio-image study (SCAPIS) using register data

    • Aims: To illustrate the importance of access to register data on determinants and predictors of study participation to assess validity of population-based studies. In the present investigation, we use data on sociodemographic conditions and disease history among individuals invited to the Swedish cardiopulmonary bio-image study (SCAPIS) in order to establish a model that predicts study participation. Methods: The pilot study of SCAPIS was conducted within the city of Gothenburg, Sweden, in 2012, with 2243 invited individuals (50% participation rate). An anonymous data set for the total target population (n = 24,502) was made available by register authorities (Statistics Sweden and the National Board of Health and Welfare) and included indicators of invitation to and participation in SCAPIS along with register data on residential area, sociodemographic variables, and disease history. Propensity scores for participation were estimated using logistic regression. Results: Residential area, country of birth, civil status, education, occupational status, and disposable income were all associated with participation in multivariable models. Adding data on disease history only increased overall classification ability marginally. The associations with disease history were diverse with some disease groups negatively associated with participation whereas some others tended to increase participation. Conclusions: The present investigation stresses the importance of a careful consideration of selection effects in population-based studies. Access to detailed register data also for non-participants can in the statistical analysis be used to control for selection bias and enhance generalizability, thereby making the results more relevant for policy decisions.

    • Forskningsrådet vid Akademin för Hälsa och Arbetsliv har identifierat behovet av att synliggöra den forskning som bedrivs inom akademin ämnesområden. Som en del i detta arbete samlade forskningsrådet inför 2018 in projektbeskrivningar för pågående projekt. Det huvudsakliga syftet var att få överblick över forskningsverksamheten vid akademin. För att även kunna kommunicera den forskning som pågår, såväl internt som externt, har vi valt att publicera projektbeskrivningarna i denna rapport.Titeln på rapporten ”Forskning pågår” signalerar att forskning bedrivs i föränderliga processer. Forskningens ständiga utveckling gör att rapporten ger en ögonblicksbild av forskningens inriktning och innehåll. Projekt formas, omformas och nya projekt startar upp. I rapporten har projekten sorterats under temporära samlingsrubriker som vi idag ser som starka inriktningar för forskningen vid akademin. I arbetet med att samla forskningen i styrkeområden kommer även dessa inriktningar att kunna förändras.

    • Genom denna rapport vill vi synliggöra den forskning som bedrivs inom Akademin för hälsa och arbetsliv. Som underlag samlar forskningsrådet vid akademin årligen in projektbeskrivningar från forskarna kring pågående projekt. Rapporten är tänkt att ge en överblick över forskningens inriktning och innehåll. Rapporten bidrar även till att kunna kommunicera pågående forskning såväl internt som externt.I rapporten har projekten sorterats under temporära samlingsrubriker som vi idag ser som starka inriktningar för forskningen vid akademin. Forskning och kunskapande sker i processer där både innehåll och inriktning flyttas under resans gång. Projekt formas, omformas och nya projekt startar upp. Titeln på rapporten ”Forskning pågår” signalerar just denna föränderlighet.Det är med stor glädje som vi för andra året i rad kan presentera över 100 pågående projekt med inriktning mot hälsa och arbetsliv.
