  • End-stage renal disease and low level exposure to lead, cadmium and mercury; a population-based, prospective nested case-referent study in Sweden.
  • 2013
  • Ingår i: Environmental health : a global access science source. - : BioMed Central (BMC). - 1476-069X. ; 12
  • Tidskriftsartikel (refereegranskat)abstract
    • ABSTRACT: Background: Cadmium (Cd), lead (Pb), and mercury (Hg) cause toxicological renal effects, but the clinical relevance at low-level exposures in general populations is unclear. The objective of this study is to assess the risk of developing end-stage renal disease in relation to Cd, Pb, and Hg exposure. Methods: A total of 118 cases who later in life developed end-stage renal disease, and 378 matched (sex, age, area, and time of blood sampling) referents were identified among participants in two population-based prospective cohorts (130,000 individuals). Cd, Pb, and Hg concentrations were determined in prospectively collected samples. Results: Erythrocyte lead was associated with an increased risk of developing end-stage renal disease (mean in cases 76 μg/L; odds ratio (OR) 1.54 for an interquartile range increase, 95% confidence interval (CI) 1.18-2.00), while erythrocyte mercury was negatively associated (2.4 μg/L; OR 0.75 for an interquartile range increase, CI 0.56-0.99). For erythrocyte cadmium, the OR of developing end-stage renal disease was 1.15 for an interquartile range increase (CI 0.99-1.34; mean Ery-Cd among cases: 1.3 μg/L). The associations for erythrocyte lead and erythrocyte mercury, but not for erythrocyte cadmium, remained after adjusting for the other two metals, smoking, BMI, diabetes, and hypertension. Gender-specific analyses showed that men carried almost all of the erythrocyte lead and erythrocyte cadmium associated risks. Conclusions: Erythrocyte lead is associated with end-stage renal disease but further studies are needed to evaluate causality. Gender-specific analyses suggest potential differences in susceptibility or in exposure biomarker reliability.

  • Assessing Command and Control Effectiveness : Dealing with a changing world
  • 2014
  • Samlingsverk (redaktörskap) (övrigt vetenskapligt/konstnärligt)abstract
    • Assessing Command and Control Effectiveness: Dealing with a Changing World offers a description of the current state of Command and Control (C2) research in imperfect settings, showing how a research process should assess, analyse and communicate results to the development cycle of methods, work, manning and C2-technology. Special attention is given to the development of C2 research methods to meet the current and coming needs. The authors also look forward towards a future where effective assessment of C2 abilities are even more crucial, for instance in agile organisations. The purpose of the C2 research is to improve the process and make it more effective while still saving time and money. Research methods have to be chosen carefully to be effective and simple, yet provide results of high quality. The methodological concerns are a major consideration when working under such circumstances. Furthermore, there is often a need for a swift iterative development cycle, and thus a demand to quickly deliver results from the research process. This book explains how field research experimentation can be quick, simple and effective, being able to draw valid conclusions even when sample sizes are small and resources are limited, collecting empirical data using measures and procedures that are minimally intrusive.

  • De Novo Determination of the Crystal Structure of a Large Drug Molecule by Crystal Structure Prediction-Based Powder NMR Crystallography
  • 2013
  • Ingår i: Journal of the American Chemical Society. - : American Chemical Society (ACS). - 0002-7863 .- 1520-5126. ; 135:46, s. 17501-17507
  • Tidskriftsartikel (refereegranskat)abstract
    • The crystal structure of form 4 of the drug 4-[4-(2-adamantylcarbamoyl)-5-tert-butyl-pyrazol-1-yl]benzoic acid is determined using a protocol for NMR powder crystallography at natural isotopic abundance combining solid-state H-1 NMR spectroscopy, crystal structure prediction, and density functional theory chemical shift calculations. This is the first example of NMR crystal structure determination for a molecular compound of previously unknown structure, and at 422 g/mol this is the largest compound to which this method has been applied so far.

  • Extracellular water and blood pressure in adults with growth hormone (GH) deficiency: a genotype-phenotype association study.
  • 2014
  • Ingår i: PloS one. - : Public Library of Science (PLoS). - 1932-6203. ; 9:8
  • Tidskriftsartikel (refereegranskat)abstract
    • Growth hormone deficiency (GHD) in adults is associated with decreased extracellular water volume (ECW). In response to GH replacement therapy (GHRT), ECW increases and blood pressure (BP) reduces or remains unchanged. Our primary aim was to study the association between polymorphisms in genes related to renal tubular function with ECW and BP before and 1 year after GHRT. The ECW measures using bioimpedance analysis (BIA) and bioimpedance spectroscopy (BIS) were validated against a reference method, the sodium bromide dilution method (Br(-)).

  • Genotypes associated with lipid metabolism contribute to differences in serum lipid profile of GH-deficient adults before and after GH replacement therapy.
  • 2012
  • Ingår i: European journal of endocrinology / European Federation of Endocrine Societies. - 1479-683X .- 0804-4643. ; 167:3, s. 353-62
  • Tidskriftsartikel (refereegranskat)abstract
    • bjective: GH deficiency (GHD) in adults is associated with an altered serum lipid profile that responds to GH replacement therapy (GHRT). This study evaluated the influence of polymorphisms in genes related to lipid metabolism on serum lipid profile before and after 1 year of GHRT in adults. Design and methods: In 318 GHD patients, total cholesterol (TC) serum concentrations, LDL-C, HDL-C, and triglycerides (TG) were assessed. Using a candidate gene approach, 20 single nucleotide polymorphisms (SNPs) were genotyped. GH dose was individually titrated to obtain normal serum IGF1 concentrations. Results: At baseline, the minor alleles of cholesteryl ester transfer protein (CETP) gene SNPs rs708272 and rs1800775 were associated with higher serum TC and apolipoprotein E (APOE) gene SNP rs7412 with lower TC concentrations; CETP SNPs rs708272, rs1800775, and rs3764261 and apolipoprotein B (APOB) gene SNP rs693 with higher serum HDL-C; APOE SNP rs7412, peroxisome proliferator-activated receptor gamma (PPARG) gene SNP rs10865710 with lower LDL-C, and CETP SNP rs1800775 with higher LDL-C; and APOE/C1/C4/C2 cluster SNP rs35136575 with lower serum TG. After treatment, APOB SNP rs676210 GG genotype was associated with larger reductions in TC and LDL-C and PPARG SNP rs10865710 CC genotype with greater TC reduction. All associations remained significant when adjusted for age, sex, and BMI. Conclusions: In GHD adults, multiple SNPs in genes related to lipid metabolism contributed to individual differences in baseline serum lipid profile. The GH treatment response in TC and LDL-C was influenced by polymorphisms in the APOB and PPARG genes.

  • Närhet och kommunikation på distans : en studie om hur studenter och lärare beskriver och upplever lärcenter/högskolecentrumbaserad utbildning
  • 2011
  • Rapport (refereegranskat)abstract
    • Since 2003 the University West in Sweden offers distance courses in Social work and Social pedagogy. The three year program is given in collaboration with a number of local learning centers throughout the country. The technology of videoconferencing enables the teacher to interact with students even though they are separated geographically. The study deals with this kind of education seen from the perspective of both students and teachers. The main objective is to illuminate significant aspects of this kind of phenomenon in terms of how students and teachers describe and experience problems and potential embedded in this model. The discussion rests on four overall questions:1) What are the motives for students to choose this kind of education?2) What kind of expectations did the students have before they started the program?3) How do students experience/describe strengths and weaknesses connected to this model?4) What experiences do the teacher's have working with this kind of distance education? The empirical data has been collected using mainly group interviews and observations. The analysis rests heavily on theories concerning symbolic interactionism and socio-cultural learning. The study underlines that both students and teachers embrace the potential in this form of education, giving more groups the possibility of combining their studies with a sense of regional belonging. However the interviews and observations also reveal weaknesses with this kind of educational model. There is a risk that students at the local learning centers consider themselves as being excluded by teachers and other students at campus; and that the gap between the university and the local learning centers becomes too wide. To prevent this from happening communication and confirmation from the teacher is crucial. But eventhough this scrutiny identifies weaknesses within the technology of videoconferencing, technology alone does not determine the outcome of the teaching. If a teacher manages to show great engagement and structure, this functions as a mechanism weakening the barrierof technology. At the same time it is clear that the model of videoconferencing, from a teacher perspective, can be an obstacle, which emphasizes the importance of developing the technology further and how this technology is being handled by the teacher. Finally, we consider the model with local learning centers and videoconferencing as a unique model for education, but that the model, when it comes to areas such as pedagogy, organization and technology, must be allowed to function and develop according to its own premises.

  • 2014
  • Ingår i: Arbetsmarknad & Arbetsliv. - Karlstad : Karlstads universitet. - 1400-9692 .- 2002-343X. ; 20:2, s. 25-40
  • Tidskriftsartikel (refereegranskat)abstract
