    • We recently demonstrated that the LKB1 tumour suppressor kinase, in complex with the pseudokinase STRAD and the scaffolding protein MO25, phosphorylates and activates AMP-activated protein kinase (AMPK). A total of 12 human kinases (NUAK1, NUAK2, BRSK1, BRSK2, QIK, QSK, SIK, MARK1, MARK2, MARK3, MARK4 and MELK) are related to AMPK. Here we demonstrate that LKB1 can phosphorylate the T-loop of all the members of this subfamily, apart from MELK, increasing their activity >50-fold. LKB1 catalytic activity and the presence of MO25 and STRAD are required for activation. Mutation of the T-loop Thr phosphorylated by LKB1 to Ala prevented activation, while mutation to glutamate produced active forms of many of the AMPK-related kinases. Activities of endogenous NUAK2, QIK, QSK, SIK, MARK1, MARK2/3 and MARK4 were markedly reduced in LKB1-deficient cells. Neither LKB1 activity nor that of AMPK-related kinases was stimulated by phenformin or AICAR, which activate AMPK. Our results show that LKB1 functions as a master upstream protein kinase, regulating AMPK-related kinases as well as AMPK. Between them, these kinases may mediate the physiological effects of LKB1, including its tumour suppressor function.

    • A careful reinvestigation by high-field F-19 NMR (470 MHz) spectroscopy has been made of the Al3+/F- system in aqueous solution under carefully controlled conditions of pH, concentration, ionic strength (I), and temperature. The F-19 NMR spectra show five distinct signals at 278 K and I = 0.6 M (TMACl) which have been attributed to the complexes AlFi(3-i)+(aq) with i less than or equal to 5. There was no need to invoke AlFi(OH)(j)((3-i-j)+) mixed complexes in the model under our experimental conditions (pH less than or equal to 6.5), nor was any evidence obtained for the formation of AlF63-(aq) at very high ratios of F-/Al3+. The stepwise equilibrium constants obtained for the complexes by integration of the F-19 signals are in good agreement with literature data given the differences in medium and temperature. In I = 0.6 M TMACl at 278 K and in I = 3 M KCl at 298 K the log K-i values are 6.42, 5.31, 3.99, 2.50, and 0.84 (for species i = 1-5) and 6.35, 5.25, and 4.11 (for species i = 1-3), respectively. Disappearance of the F-19 NMR signals under certain conditions was shown to be due to precipitation. Certain 19F NMR signals exhibit temperature- and concentration-dependent exchange broadening. Detailed line shape analysis of the spectra and magnetization transfer measurements indicate that the kinetics are dominated by F- exchange rather than complex formation. The detected reactions and their rate constants are AlF22+ + *F- reversible arrow AIF*F2+ + F- (k(02) = (1.8 +/- 0.3) x 10(6) M-1 s(-1)), AlF30 + *F- reversible arrow (AlF2F0)-F-* + F- (k(03) = (3.9 +/- 0.9) x 10(6) M-1 s(-1)), and AlF30 + H*F reversible arrow AlF2*F-0 + HF (k(03)(H) = (6.6 +/- 0.5) x 10(4) M-1 s(-1)). The rates of these exchange reactions increase markedly with increasing F- substitution. Thus, the reactions of AlF2+(aq) were too inert to be detected even on the T-1 NMR time scale, while some of the reactions of AlF30(aq) were fast, causing large line broadening. The ligand exchange appears to follow an associative interchange mechanism. The cis-trans isomerization of AlF2+(aq), consistent with octahedral geometry for that complex, is slowed sufficiently to be observed at temperatures around 270 R. Difference between the Al3+/F- system and the much studied Al3+/OH- system are briefly commented on.

    • Abstract BACKGROUND: Most individuals with Helicobacter pylori infection in Western countries have no evidence of peptic ulcer disease (PUD). We therefore assessed the PiZ deficiency variant of the major plasma protease inhibitor alpha1-antitrypsin (alpha1AT) as a risk factor for PUD in H. pylori-infected individuals. METHODS: The cohort comprised 100 patients with endoscopically or surgically proven PUD (30 patients with duodenal ulcer (DU) and 70 patients with gastric ulcer (GU)) and 162 age- and sex-matched controls with PUD-negative endoscopic findings and no history of PUD. Plasma samples were screened for alpha1AT deficiency (PiZ) with an enzyme-linked immunosorbent assay (ELISA) and phenotyped by isoelectric focusing. H. pylori infection was evaluated with an IgG ELISA technique. RESULTS: Among the 262 patients 17 (6.5%) were positive for the PiZ alpha1AT deficiency, a frequency of the same magnitude as in the Swedish general population (4.7%). Of the PiZ carriers 76% (13 of 17) had H. pylori antibodies compared with 61% (151 of 245) of the non-PiZ carriers (NS). The prevalence of DU tended to be higher in H. pylori-positive PiZ carriers than in non-PiZ carriers (15.4%, 4 of 26 versus 0 of 4). Furthermore, among patients with DU a high PiZ allele frequency (13.3%, 4 of 30) was found compared with the general population (4.7%) (odds ratio (OR), 3.2; 95% confidence interval (CI), 1.09-8.94; P = 0.02). All DU patients carrying the PiZ allele were positive for H. pylori. In addition, four of five PiZ carriers with H. pylori infection and PUD had DU. CONCLUSIONS: The PiZ allele may be a contributing factor in the development of DU in H. pylori-positive individuals.

    • In order to search for novel giant-magnetoresistance systems, we fabricated and investigated narrow constrictions in the layers of the ferromagnetic semiconductor (Ga,Mn)As. We found that constrictions a few hundred nanometers wide, tailored by means of the electron-beam lithography and wet etching, were not conducting at liquid helium temperatures unless illuminated, probably due to the trapping action of surface states appearing on an extra surface area denuded by the etching. To avoid this, we used selective implantation of oxygen ions into the ferromagnetic layer to tailor the constrictions. We have shown that such an implantation inactivates Mn acceptors in the layer and destroys ferromagnetism. We propose an application of oxygen ion implantation as a method of fabricating microcircuits in future spin electronics based on Mn-containing III-V semiconductor compounds.

    • The formation of three [TI(en)(n)](3+) complexes (n = 1-3) in a pyridine solvent has been established by means of Tl-205 and H-1 NMR. Their stepwise stability constants based on concentrations, K-n = [Tl(en)(n)(3+)]/{[Tl(en)(n-1)(3+)](.)[en]}, at 298 K in 0.5 M NaClO4 ionic medium in pyridine, were calculated from Tl-205 NMR integrals: log K-1 = 7.6 +/- 0.7; log K-2 = 5.2 +/- 0.5 and log K-3 = 2.64 +/- 0.05. Linear correlation between both the Tl-205 NMR shifts and spin-spin coupling Tl-205-H-1 versus the stability constants has been found and discussed. A single crystal with the composition [Tl(en)(3)](ClO4)(3) was synthesized and its structure determined by X-ray diffraction. The Tl3+ ion is coordinated by three ethylenediamine ligands via six N-donor atoms in a distorted octahedral fashion.

  • Determination of transport in the Caco-2 cell assay of compounds varying in lipophilicity using LC-MS : enhanced transport of Leu-enkephalin analogues

    • PURPOSE: To synthesize a number of analogues of Leu-enkephalin with different lipophilicities and to develop an LC-MS method for determining the Caco-2 cell permeability values of these compounds. METHODS: A number of sugar and sugar plus lipoamino acid analogues of Leu-enkephalin were synthesized by solid-phase and solution methods. An LC-MS method was developed for analyzing the Caco-2 cell assay samples and validated against the traditional method using radiolabelled compounds. RESULTS: A sensitive and specific LC-MS assay was developed. Standard curves were linear in the range of 0.025-5 microM. Apparent permeability values determined by LC-MS and liquid scintillation counter were identical, for both a hydrophilic drug, cephalexin and a lipophilic Leu-enkaphalin analogue. Caco-2 permeability values for the analogues of Leu-enkephalin were determined and it was found that attachment of sugar or sugar and lipoamino acid to the Leu-enkephalin peptide resulted in an increase in the apparent permeability values compared to the native peptide, which was not transported across the Caco-2 cell monolayers. CONCLUSIONS: A rapid, generic LC-MS method for analyzing a range of compounds was developed. Attachment of a sugar or sugar and lipoamino acid to Leu-enkephalin improves the apparent permeability across Caco-2 cell monolayers.
