  • First-in-human, phase I/IIa clinical study of the peptidase potentiated alkylator melflufen administered every three weeks to patients with advanced solid tumor malignancies

    • Purpose Melflufen (melphalan flufenamide, previously designated J1) is an optimized and targeted derivative of melphalan, hydrolyzed by aminopeptidases overexpressed in tumor cells resulting in selective release and trapping of melphalan, and enhanced activity in preclinical models. Methods This was a prospective, single-armed, open-label, first-in-human, dose-finding phase I/IIa study in 45 adult patients with advanced and progressive solid tumors without standard treatment options. Most common tumor types were ovarian carcinoma (n = 20) and non-small-cell lung cancer (NSCLC, n = 11). Results In the dose-escalating phase I part of the study, seven patients were treated with increasing fixed doses of melflufen (25-130 mg) Q3W. In the subsequent phase IIa part, 38 patients received in total 115 cycles of therapy at doses of 30-75 mg. No dose-limiting toxicities (DLTs) were observed at 25 and 50 mg; at higher doses DLTs were reversible neutropenias and thrombocytopenias, particularly evident in heavily pretreated patients, and the recommended phase II dose (RPTD) was set to 50 mg. Response Evaluation Criteria In Solid Tumors (RECIST) evaluation after 3 cycles of therapy (27 patients) showed partial response in one (ovarian cancer), and stable disease in 18 patients. One NSCLC patient received nine cycles of melflufen and progressed after 7 months of therapy. Conclusions In conclusion, melflufen can safely be given to cancer patients, and the toxicity profile was as expected for alkylating agents; RPTD is 50 mg Q3W. Reversible and manageable bone marrow suppression was identified as a DLT. Clinical activity is suggested in ovarian cancer, but modest activity in treatment of refractory NSCLC.

  • The Elevation of Sensitivity Over Truth: Political Correctness and Related Phrases in the Time Magazine Corpus.

    • This article is a quantitative and qualitative diachronic study of how the expression politically correct (PC) and related phrases are used in the American magazine Time from 1923 through 2006. The data show a dramatic increase in the frequency with which PC-phrases are used in the early 1990s. From this time onwards, the phrases are often used as a means of passing evaluative subjective opinions off as objectively reported facts, especially in reviews of cultural events, where they figure prominently. In contrast to earlier studies, our data show that PC-phrases are not inherently negative; this applies primarily to discourse on environment and business, where to be PC often implies being environmentally or socially conscious in a positive sense. Nevertheless, negative or ironic uses of the terms predominate. Most often they express criticism of unspoken cultural norms rather than being attempts to close down debate or criticizing the replacement of offensive terms by more neutral expressions

  • Margaret Atwood's Environmentalism : Apocalypse and Satire in the MaddAddam Trilogy

    • This study considers the way in which Margaret Atwood’s post-apocalyptic MaddAddam Trilogy functions as an environmental project. The main focus is on how the three novels, Oryx and Crake (2003), The Year of the Flood (2009), and MaddAddam (2013), simultaneously draw on and destabilise the apocalypticism inherent in so much environmental discourse, primarily through the use of satire. The trilogy is securely anchored in the concerns of contemporary readers, and transposition of the action to the near future is integral to Atwood’s environmental project: attention is focussed on the present causes of anticipated environmental catastrophe, which readers implicitly are implored to avoid. Atwood’s environmentalism is performed in the interplay between her literary stature, the equivocal content of her work, and the irreverence with which she metaleptically blurs distinctions between fact and fiction, art and commodity, and activism and aesthetics. Whereas the satiric mode serves as a way of avoiding some of the limitations of apocalyptic thinking by maintaining and even creating complexity, it also renders the entire project ambiguous. Uncertainty about the exact environmental injunction presented in the trilogy creates doubts about the degree to which Atwood’s extradiegetic environmental activism should be taken seriously, or conversely. Storytelling is foregrounded in all three novels, and through its concurrent critique of and reliance on market forces and the political potential of art, the MaddAddam Trilogy demonstrates that there is no external position from which the imagination can perform environmentalist miracles. As such, Atwood’s environmental project furthers a profoundly ecological understanding of the world.

    • We tested whether polygenic risk scores for schizophrenia and bipolar disorder would predict creativity. Higher scores were associated with artistic society membership or creative profession in both Icelandic (P = 5.2 x 10(-6) and 3.8 x 10(-6) for schizophrenia and bipolar disorder scores, respectively) and replication cohorts (P = 0.0021 and 0.00086). This could not be accounted for by increased relatedness between creative individuals and those with psychoses, indicating that creativity and psychosis share genetic roots.

  • Predictive Factors of Concerns about Falling in People with Parkinson's Disease : A 3-Year Longitudinal Study

    • Introduction: Fear of falling (FOF) is more common in people with Parkinson's disease (PD) than in healthy controls. It can lead to several negative consequences such as restrictions in everyday life. Moreover, FOF is a risk factor for future falls. Aim: This study aimed to identify predictive factors of FOF (conceptualized as concerns about falling) after three years, with and without adjusting for concerns about falling at baseline, in people with PD. Methods: This study included 151 participants (35% women) with PD. At baseline, their mean (SD) age and PD duration were 68 (±9.0) and 9 (±6.1) years, respectively. The Falls Efficacy Scale-International (FES-I) was used as the dependent variable in multivariable linear regression analyses. Results: The mean (SD) FES-I score increased from 28.1 (11.9) to 33.1 (14.0) three years later (p<0.001). The strongest (according to the standardized regression coefficient, β) predictor of concerns about falling was walking difficulties (β = 0.378), followed by age (0.227), problems maintaining balance while dual tasking (0.172), and needing help in daily activities (0.171). When adjusting for baseline FES-I scores, the strongest predictive factor was problems maintaining balance while dual tasking (β = 0.161), which was followed by age (0.131) and female sex (0.105). Conclusions: This study pinpoints several predictive factors of concerns about falling that are modifiable and which could be addressed in rehabilitation: perceived walking difficulties, having problems maintaining balance while dual tasking, and dependence on others in daily activities. The importance of dual tasking is a novel finding, which future studies need to confirm or refute. One should be aware of the fact that an increased age predicts concerns about falling with and without adjusting for baseline FES-I scores, whereas female sex predicts concerns about falling only when adjusting for baseline FES-I scores.

    • Torsten Petterssons forskning utmärks av en mångfald och ett djup med få motstycken i nutida svenskspråkig litteraturvetenskap. Under de senaste 40 åren har han rört sig obehindrat mellan litteratur på en rad olika språk och från flertalet epoker. Till den vetenskapliga gärningen ska också läggas Torstens skönlitterära författarskap som omfattar dikter, noveller och kriminalromaner.Med denna festskrift, som utges i samband med att Torsten fyller 60 år, vill vi hylla honom och hans insats inom litteraturvetenskapen. Bidragen tar fasta på Torstens förmåga att överskrida gränser mellan discipliner, teman och metoder, och är skrivna av svenska och internationella forskare från en rad olika forskningsområden. Texterna behandlar ämnen som tolkningsteori och empirisk läsforskning, svensk, finlandssvensk, engelskspråkig och tysk litteratur, teologi och livsåskådning samt Torstens egen insats både som forskare och som skönlitterär författare.

  • “A tangled web of mindfuck" : Andrea Dworkin and the Truth of Pornography

    • This essay argues that Andrea Dworkin’s much derided argument against pornography may still prove productive if read in relation to her fiction. Taking note of the fact that Dworkin’s own novels have been accused of being pornographic according to her criteria, the essay explores the possibility that her radical feminist anti-porn stance, inevitably described as something of a moralistic backlash by sex-liberals, could be seen as a continuation of the pornographic imagination it so vehemently protests. Through critical readings of Dworkin’s novels Ice and Fire and Mercy, as well as her autobiographical works and theoretical writings on pornography, the essay demonstrates that the contradic- tions that beset her writings echo the contradictions inherent in our contemporary under- standing of pornography. Much like the pornography she would resist, Dworkin’s writings invite a solipsistic mode of reading, the experiential rewards of which are not so much of an interpretive as of a sensual nature. In so doing, Dworkin’s fiction paradoxically proves an invaluable point of entry into the pornographic imagination, making evident its essentially monologic nature. While this circumstance may be damaging for her status as an agitator, it points to her continued—if as yet largely unacknowledged—importance as a writer.
