  • Multi-leptons with high transverse momentum at HERA

    • Events with at least two high transverse momentum leptons (electrons or muons) are studied using the H1 and ZEUS detectors at HERA with an integrated luminosity of 0.94 fb(-1). The observed numbers of events are in general agreement with the Standard Model predictions. Seven di- and tri-lepton events are observed in e(+)p collision data with a scalar sum of the lepton transverse momenta above 100 GeV while 1.94 +/- 0.17 events are expected. Such events are not observed in e(-)p collisions for which 1.19 +/- 0.12 are predicted. Total visible and differential di-electron and di-muon photoproduction cross sections are extracted in a restricted phase space dominated by photon-photon collisions.

  • One-neutron knockout of O-23

    • Breakup reactions were used to study the ground-state configuration of the neutron-rich isotope O-23. The O-22 fragments produced in one-nucleon removal from O-23 at 938 MeV/nucleon in a carbon target were detected in coincidence with de-exciting gamma rays, allowing to discern between 220 ground-state and excited-states contributions. From the comparison of exclusive experimental momentum distributions for the one-neutron removal channel to theoretical momentum distributions calculated in an Eikonal model for the knockout process, and spin and parity assignment of I-pi = 1/2(+) was deduced for the 230 ground state. This result solved the existent experimental discrepancy.

    • In the pig, a functional tubal sperm reservoir (SR) is established before ovulation to ensure availability of suitable numbers of viable spermatozoa for fertilization. The boars large ejaculate is split: most spermatozoa are delivered in a sperm-rich fraction (SRF) followed by a post-SRF fraction containing increasing amounts of the spermadhesin PSP-I/PSP-II-rich seminal vesicle secretion. This heterodimer acts as leukocyte chemoattractant both in vitro and in vivo, contributing to the phagocytosis of those spermatozoa not reaching the SR. Sequential ejaculate deposition of marked spermatozoa and SR screening showed that most spermatozoa in the SR arose from the fortuitous PSP-poor, first portion of the SRF fraction, escaping phagocytosis and replenishing the SR within 23 h. The SR-sperm numbers diminish gradually in relation to ovulation, spermatozoa. being continuously redistributed toward the upper isthmus. In vitro, only uncapacitated spermatozoa bind to epithelial explants, suggesting that the SR influences sperm capacitation. In vivo, most viable spermatozoa - usually harbored in the deep furrows in the pre- or peri-ovulatory SR during spontaneous standing estrus - are uncapacitated, but capacitation significantly increases after ovulation. Pre-/peri-ovulatory SR spermatozoa promptly capacitate in vitro when exposed to the effector bicarbonate, an influence that can be reversed by co-incubation with SR fluid or its component hyaluronan. Fluid collected from the ampullar segment (rich in bicarbonate) induces capacitation in vitro. In conclusion, the lack of massive sperm capacitation in the SR and the diverse individual response to capacitation shown by tubal spermatozoa would relate both to the insurance of full sperm viability before ovulation and the presence of spermatozoa at different stages of capacitation in the upper oviduct, thus maximizing the chances of normal fertilization.   • Identification of a new JNK inhibitor targeting the JNK-JIP interaction site

    • NK is a stress-activated protein kinase that modulates pathways implicated in a variety of disease states. JNK-interacting protein-1 (JIP1) is a scaffolding protein that enhances JNK signaling by creating a proximity effect between JNK and upstream kinases. A minimal peptide region derived from JIP1 is able to inhibit JNK activity both in vitro and in cell. We report here a series of small molecules JIP1 mimics that function as substrate competitive inhibitors of JNK. One such compound, BI-78D3, dose-dependently inhibits the phosphorylation of JNK substrates both in vitro and in cell. In animal studies, BI-78D3 not only blocks JNK dependent Con A-induced liver damage but also restores insulin sensitivity in mouse models of type 2 diabetes. Our findings open the way for the development of protein kinase inhibitors targeting substrate specific docking sites, rather than the highly conserved ATP binding sites. In view of its favorable inhibition profile, selectivity, and ability to function in the cellular milieu and in vivo, BI-78D3 represents not only a JNK inhibitor, but also a promising stepping stone toward the development of an innovative class of therapeutics.

  • GLUE Based Assessment on the Overall Predictions of a MIKE SHE Application

    • The generalised likelihood uncertainty estimation (GLUE) approach was applied to assess the performance of a distributed catchment model and to estimate prediction limits after conditioning based on observed catchment-wide streamflow. Prediction limits were derived not only for daily streamflow but also for piezometric levels and for extreme events. The latter analysis was carried out considering independent partial duration time series (PDS) obtained from the observed daily streamflow hydrograph. Important data uncertainties were identified. For streamflow the stage-discharge data analysis led to estimate an average data uncertainty of about 3 m(3) s (-aEuro parts per thousand 1). For piezometric levels, data errors were estimated to be in the order of 5 m in average and 10 m at most. The GLUE analysis showed that most of the inspected parameters are insensitive to model performance, except the horizontal and vertical components of the hydraulic conductivity of one of the geological layers that have the most influence on the streamflow model performance in the application catchment. The study revealed a considerable uncertainty attached to the simulation of both high flows and low flows (i.e., in average terms 5 m(3) s (-aEuro parts per thousand 1) before the Bayesian updating of the prediction limits). Similarly, wide prediction intervals were obtained for the piezometric levels in relevant wells, in the order of 3.3 and 1.5 m before and after the Bayesian updating of the prediction limits, respectively. Consequently, the results suggest that, in average terms, the model of the catchment predicts overall outputs within the limitations of the errors in the input variables.
