    • Diversity within microglia, the resident brain immune cells, is reported. Whether microglial subsets constitute different subtypes with intrinsic properties and unique functions has not been fully elucidated. Here, we describe a microglial subtype characterized by the expression of the enzyme Arginase-1, i.e.Arg1+microglia, which is found predominantly in the cholinergic neuron-rich forebrain region during early postnatal development. Arg1+microgliacontain cellular inclusions and exhibit a distinct molecular signature including upregulation of genes such as Apoe, Clec7a, Igf1, Lgals3and Mgl2. Arg1-knockout in microglia results in a deficient cholinergic innervation along with impaired dendritic spine maturation in the hippocampus where cholinergic neurons project, impaired long-term potentiation and cognitive behavioural deficiencies in female mice. Our results expand on microglia diversity and provide insights into distinctive spatiotemporal functions exerted by microglial subtypes.

  • Light elements Na and Al in 58 bulge spheroid stars from APOGEE

    • We identified a sample of 58 candidate stars with metallicity [Fe/H]-0.8 that likely belong to the old bulge spheroid stellar population, and analyse their Na and Al abundances from Apache Point Observatory Galactic Evolution Experiment (APOGEE) spectra. In a previous work, we inspected APOGEE-Stellar Parameter and Chemical Abundance Pipeline abundances of C, N, O, Mg, Al, Ca, Si, and Ce in this sample. Regarding Na lines, one of them appears very strong in about 20 per cent of the sample stars, but it is not confirmed by other Na lines, and can be explained by sky lines, which affect the reduced spectra of stars in a certain radial velocity range. The Na abundances for 15 more reliable cases were taken into account. Al lines in the H band instead appear to be very reliable. Na and Al exhibit a spread in abundances, whereas no spread in N abundances is found, and we found no correlation between them, indicating that these stars could not be identified as second-generation stars that originated in globular clusters. We carry out the study of the behaviour of Na and Al in our sample of bulge stars and literature data by comparing them with chemodynamical evolution model suitable for the Galactic bulge. The Na abundances show a large spread, and the chemodynamical models follow the main data, whereas for aluminum instead, the models reproduce very satisfactorily the nearly secondary-element behaviour of aluminum in the metallicity range below [Fe/H]-1.0. For the lower-metallicity end ([Fe/H <-2.5), hypernovae are assumed to be the main contributor to yields.

    • Background Neurofilament light chain (NfL) is a promising biomarker of active axonal injury and neuronal degeneration. We aimed to characterise cross-sectional and longitudinal plasma NfL measurements and determine the age at which NfL concentrations begin to differentiate between carriers of the presenilin 1 (PSEN1) E280A (G1u280A1a) mutation and age-matched non-carriers from the Colombian autosomal dominant Alzheimer's disease kindred. Methods In this cross-sectional and longitudinal cohort study, members of the familial Alzheimer's disease Colombian kindred aged 8-75 years with no other neurological or health conditions were recruited from the Alzheimer's Prevention Initiative Registry at the University of Antioquia (Medellin, Colombia) between Aug 1, 1995, and Dec 15, 2018. We used a single molecule array immunoassay and log-transformed data to examine the relationship between plasma NfL concentrations and age, and establish the earliest age at which NfL concentrations begin to diverge between mutation carriers and non-carriers. Findings We enrolled a cohort of 1070 PSEN1 E280A mutation carriers and 1074 non-carriers with baseline assessments; of these participants, longitudinal measures (with a mean follow-up of 6 years) were available for 242 mutation carriers and 262 non-carriers. Plasma NfL measurements increased with age in both groups (p<0 . 0001), and began to differentiate carriers from non-carriers when aged 22 years (22 years before the estimated median age at mild cognitive impairment onset of 44 years), although the ability of plasma NfL to discriminate between carriers and non-carriers only reached high sensitivity close to the age of clinical onset. Interpretation Our findings further support the promise of plasma NfL as a biomarker of active neurodegeneration in the detection and tracking of Alzheimer's disease and the evaluation of disease-modifying therapies.   • Chrono-chemodynamical analysis of the globular cluster NGC 6355 : Looking for the fundamental bricks of the Bulge

    • The information on Galactic assembly time is imprinted on the chemodynamics of globular clusters. This makes them important probes that help us to understand the formation and evolution of the Milky Way. Discerning between in-situ and ex-situ origin of these objects is difficult when we study the Galactic bulge, which is the most complex and mixed component of the Milky Way. To investigate the early evolution of the Galactic bulge, we analysed the globular cluster NGC 6355. We derived chemical abundances and kinematic and dynamic properties by gathering information from high-resolution spectroscopy with FLAMES-UVES, photometry with the Hubble Space Telescope, and Galactic dynamic calculations applied to the globular cluster NGC 6355. We derive an age of 13:2 ± 1:1 Gyr and a metallicity of [Fe/H] =-1:39 ± 0:08 for NGC 6355, with α-enhancement of [α/Fe] = +0:37 ± 0:11. The abundance pattern of the globular cluster is compatible with bulge field RR Lyrae stars and in-situ well-studied globular clusters. The orbital parameters suggest that the cluster is currently confined within the bulge volume when we consider a heliocentric distance of 8:54 ± 0:19 kpc and an extinction coefficient of RV = 2:84 ± 0:02. NGC 6355 is highly likely to come from the main bulge progenitor. Nevertheless, it still has a low probability of being formed from an accreted event because its age is uncertain and because of the combined [Mg/Mn] [Al/Fe] abundance. Its relatively low metallicity with respect to old and moderately metal-poor inner Galaxy clusters may suggest a low-metallicity floor for globular clusters that formed in-situ in the early Galactic bulge.

  • High Prevalence of Strongyloidiasis in Spain: A Hospital-Based Study

    • Introduction: Strongyloidiasis is a prevailing helminth infection ubiquitous in tropical and subtropical areas, however, seroprevalence data are scarce in migrant populations, particularly for those coming for Asia. Methods: This study aims at evaluating the prevalence of S. stercoralis at the hospital level in migrant populations or long term travellers being attended in out-patient and in-patient units as part of a systematic screening implemented in six Spanish hospitals. A cross-sectional study was conducted and systematic screening for S. stercoralis infection using serological tests was offered to all eligible participants. Results: The overall seroprevalence of S. stercoralis was 9.04% (95%CI 7.76–10.31). The seroprevalence of people with a risk of infection acquired in Africa and Latin America was 9.35% (95%CI 7.01–11.69), 9.22% (7.5–10.93), respectively. The number of individuals coming from Asian countries was significantly smaller and the overall prevalence in these countries was 2.9% (95%CI −0.3–6.2). The seroprevalence in units attending potentially immunosuppressed patients was significantly lower (5.64%) compared with other units of the hospital (10.20%) or Tropical diseases units (13.33%) (p < 0.001). Conclusions: We report a hospital-based strongyloidiasis seroprevalence of almost 10% in a mobile population coming from endemic areas suggesting the need of implementing strongyloidiasis screening in hospitalized patients coming from endemic areas, particularly if they are at risk of immunosuppression.

  • Chip-based multimodal super-resolution microscopy for histological investigations of cryopreserved tissue sections

    • Histology involves the observation of structural features in tissues using a microscope. While diffraction-limited optical microscopes are commonly used in histological investigations, their resolving capabilities are insufficient to visualize details at subcellular level. Although a novel set of super-resolution optical microscopy techniques can fulfill the resolution demands in such cases, the system complexity, high operating cost, lack of multi-modality, and low-throughput imaging of these methods limit their wide adoption for histological analysis. In this study, we introduce the photonic chip as a feasible high-throughput microscopy platform for super-resolution imaging of histological samples. Using cryopreserved ultrathin tissue sections of human placenta, mouse kidney, pig heart, and zebrafish eye retina prepared by the Tokuyasu method, we demonstrate diverse imaging capabilities of the photonic chip including total internal reflection fluorescence microscopy, intensity fluctuation-based optical nanoscopy, single-molecule localization microscopy, and correlative light-electron microscopy. Our results validate the photonic chip as a feasible imaging platform for tissue sections and pave the way for the adoption of super-resolution high-throughput multimodal analysis of cryopreserved tissue samples both in research and clinical settings.
