  • A global agenda for advancing freshwater biodiversity research

    • Global freshwater biodiversity is declining dramatically, and meeting the challenges of this crisis requires bold goals and the mobilisation of substantial resources. While the reasons are varied, investments in both research and conservation of freshwater biodiversity lag far behind those in the terrestrial and marine realms. Inspired by a global consultation, we identify 15 pressing priority needs, grouped into five research areas, in an effort to support informed stewardship of freshwater biodiversity. The proposed agenda aims to advance freshwater biodiversity research globally as a critical step in improving coordinated actions towards its sustainable management and conservation.

  • Coulomb dissociation of O-16 into He-4 and C-12

    • We measured the Coulomb dissociation of O-16 into He-4 and C-12 within the FAIR Phase-0 program at GSI Helmholtzzentrum fur Schwerionenforschung Darmstadt, Germany. From this we will extract the photon dissociation cross section O-16(alpha,gamma)C-12, which is the time reversed reaction to C-12(alpha,gamma)O-16. With this indirect method, we aim to improve on the accuracy of the experimental data at lower energies than measured so far. The expected low cross section for the Coulomb dissociation reaction and close magnetic rigidity of beam and fragments demand a high precision measurement. Hence, new detector systems were built and radical changes to the (RB)-B-3 setup were necessary to cope with the high-intensity O-16 beam. All tracking detectors were designed to let the unreacted O-16 ions pass, while detecting the C-12 and He-4.

  • Coulomb dissociation of 16O into 4He and 12C

    • We measured the Coulomb dissociation of 16O into 4He and 12C at the R3B setup in a first campaign within FAIR Phase 0 at GSI Helmholtzzentrum für Schwerionenforschung, Darmstadt. The goal was to improve the accuracy of the experimental data for the 12C(a,?)16O fusion reaction and to reach lower center-ofmass energies than measured so far. The experiment required beam intensities of 109 16O ions per second at an energy of 500 MeV/nucleon. The rare case of Coulomb breakup into 12C and 4He posed another challenge: The magnetic rigidities of the particles are so close because of the same mass-To-charge-number ratio A/Z = 2 for 16O, 12C and 4He. Hence, radical changes of the R3B setup were necessary. All detectors had slits to allow the passage of the unreacted 16O ions, while 4He and 12C would hit the detectors' active areas depending on the scattering angle and their relative energies. We developed and built detectors based on organic scintillators to track and identify the reaction products with sufficient precision.

    • As a special type of multiword expressions, binomials are a frequent phenomenon in many languages. We focus on binomial adverbs that are coordinations of two adverbial constituents. Their syntactic contribution to a sentence is adverbial as well and their semantic contribution is idiomatic. They have many uses, such as to intensify (first and foremost), express tendency (more and more), frequency (over and over [again]), vagueness (more or less), determination (sooner or later) etc.In this work, we describe our approaches to identify binomial adverbs in a large multiparallel corpus. Alongside the well-known measure of reversibility, we also calculate measures of statistical association and look for single-word translation equivalents in other languages. Combining these features facilitates the identification of binomial adverbs.

  • A Quality Assessment of the ARM-Net Registry Design and Data Collection

    • Background: Registries are important in rare disease research. The Anorectal Malformation Network (ARM-Net) registry is a well-established European patient registry collecting demographic, clinical, and functional outcome data. We assessed the quality of this registry through review of the structure, data elements, collected data, and user experience. Material and methods: Design and data elements were assessed for completeness, consistency, usefulness, accuracy, validity, and comparability. An intra- and inter-user variability study was conducted through monitoring and re-registration of patients. User experience was assessed via a questionnaire on registration, design of registry, and satisfaction. Results: We evaluated 119 data elements, of which 107 were utilized and comprised 42 string and 65 numeric elements. A minority (37.0%) of the 2278 included records had complete data, though this improved to 83.5% when follow-up elements were excluded. Intra-observer variability demonstrated 11.7% incongruence, while inter-observer variability was 14.7%. Users were predominantly pediatric surgeons and typically registered patients within 11–30 min. Users did not experience any significant difficulties with data entry and were generally satisfied with the registry, but preferred more longitudinal data and patient-reported outcomes. Conclusions: The ARM-Net registry presents one of the largest ARM cohorts. Although its collected data are valuable, they are susceptible to error and user variability. Continuous evaluations are required to maintain relevant and high-quality data and to achieve long-term sustainability. With the recommendations resulting from this study, we call for rare disease patient registries to take example and aim to continuously improve their data quality to enhance the small, but impactful, field of rare disease research.   • High throughput generation of a resource of the human secretome in mammalian cells

    • The proteins secreted by human tissues and blood cells, the secretome, are important both for the basic understanding of human biology and for identification of potential targets for future diagnosis and therapy. Here, a high-throughput mammalian cell factory is presented that was established to create a resource of recombinant full-length proteins covering the majority of those annotated as 'secreted' in humans. The full-length DNA sequences of each of the predicted secreted proteins were generated by gene synthesis, the constructs were transfected into Chinese hamster ovary (CHO) cells and the recombinant proteins were produced, purified and analyzed. Almost 1,300 proteins were successfully generated and proteins predicted to be secreted into the blood were produced with a success rate of 65%, while the success rates for the other categories of secreted proteins were somewhat lower giving an overall one-pass success rate of ca. 58%. The proteins were used to generate targeted proteomics assays and several of the proteins were shown to be active in a phenotypic assay involving pancreatic beta-cell dedifferentiation. Many of the proteins that failed during production in CHO cells could be rescued in human embryonic kidney (HEK 293) cells suggesting that a cell factory of human origin can be an attractive alternative for production in mammalian cells. In conclusion, a high-throughput protein production and purification system has been successfully established to create a unique resource of the human secretome.

  • Entering the dynamic risk space : Assessing planetary boundary interactions through process-based quantifications

    • The planetary boundaries framework is an effort to define a safe operating space for humanity. Its rationale is that sustainable development needs to be achieved in ways that safeguard the stability of the Earth system on which human prosperity relies. However, very few studies explicitly examine the interactions of the Earth system processes that underlie individual boundaries.My overarching research question is: how can continued anthropogenic climate change affect the geospatially resolvable land and water planetary boundaries, and what are the implications for human livelihood? For most of my analysis, I use the LPJmL dynamic global vegetation model because it contains suitable process representations that provide a dynamic and adaptive Earth system perspective for my investigation of key planetary boundary interactions of the climate, land, water and ecosystem nexus.Paper I emphasizes the importance of green water dynamics (that is terrestrial precipitation, evapotranspiration and plant-available soil moisture) for ecosystem resilience and human well-being. The underlying analysis suggests that the current status of the proposed planetary boundary for green water is already transgressed. Paper II reveals long-term spatiotemporal dynamics of planetary boundary interactions as breaching the climate change boundary critically affects the world’s major forest biomes. Notably, the most extreme climate change scenarios led to the emergence of a southern boreal dieback in the simulations. Tropical forests further show a shift from evergreen to deciduous rainforest, an important process which is not captured by the definition of the land-system change boundary. Maintaining climate change at the planetary boundary co-stabilizes the land-system change boundary. Paper III extends the biophysical understanding of planetary boundary interactions by discussing their impact on human livelihood and the attainment of the Sustainable Development Goals. Future climate change causes increases in dry anomalies of green water in ~30% of the global land area by the end of the century. As of today (here referring to 2015), nearly a quarter of the world population and ~28% of global harvest would be affected. The dynamic risk space terminology is established to fill the conceptual gap in the analysis of planetary boundary interactions. Paper IV highlights how planetary stability constitutes the non-negotiable fundament for human development and argues why the Sustainable Development Goals have to be aligned with the planetary boundaries framework and which perils might arise from their interactions. Paper V presents the land-system change reallocation tool algorithm which allows for a scenario-driven rearrangement of human land-use to meet varying transgression levels of the land-system change boundary. My results of Paper I-V advance the understanding of interactions in the planetary boundaries framework. Moreover, my analysis in a process-based and validated modeling environment gives spatiotemporal detail of the processes at play that exceeds the potential of previously used conceptual models. My work fills a crucial gap in the operationalization of the planetary boundary framework by providing insights into how and where different policy options produce positive or negative outcomes across boundaries. The holistic understanding I present is a prerequisite for any application of the planetary boundaries framework that focuses on future conditions.
