    • Sedan undervisning fick en mer framträdande position i den svenska reviderade läroplanen för förskolan (Lpfö 18) har lärare i såväl förskola som förskollärarutbildning kämpat med att tolka konceptet. När 2011 års skollag trädde i kraft några år tidigare fick förskolans arbete med normer och värden en starkare ställning. Mot bakgrund av detta blir det intressant att studera båda dessa stärkta mandat – undervisning och värden. Med utgångspunkt i erfarenheter från ett didaktiskt och pragmatiskt informerat undervisningsupplägg är syftet med denna artikel att bidra med kunskap om vilka värderingar som kan framträda i undervisning i förskola. Det empiriska material som genererats genom undervisningsupplägget består av totalt 364 dokument, inklusive 64 videoinspelningar. Detta genomfördes på cirka 120 förskolor och/eller förskoleavdelningar i tio svenska kommuner. Analysen har en didaktisk ansats och kan metodo-logiskt beskrivas som abduktiv analys. Resultatet indikerar att pragmatiskt informerad undervisning om värden kretsar kring sociala värden, såsom demokrati och (barns) delaktighet. I förskolans praktik är undervisningen organiserad som möjligheter till reflektion, samt som ett ”flerdeltagande”. Vidare framträdde spår av flerstämmighet i relation till kunskapsinnehåll som samspelar på flera olika nivåer samtidigt.

  • Comb-based Characterization of Photonic Devices

    • Integrated photonics has been one of the fastest-growing fields in science. Measuring photonic devices in amplitude and phase (i.e. complex response) provides insight into their performance. Swept-wavelength interferometry is a prominent technique for the broadband characterization of the complex response. It leverages continuous advances in rapidly tunable laser sources, but is prone to systematic errors associated to the calibration of the frequency. This thesis focuses on the non-destructive characterization of ultralow-loss photonic devices using swept wavelength interferometric technique. We overcome issues associated to nonlinear tuning by calibrating the frequency of the laser with the aid of a frequency comb. We apply the concept to diverse components of relevance including microresonators and spiral waveguides. In addition, we provide an overview and comparative assessment of the state of the art in the field.

  • Collagen VI Contains Multiple Host Defense Peptides with Potent In Vivo Activity

    • Collagen VI is a ubiquitous extracellular matrix component that forms extensive microfibrillar networks in most connective tissues. In this study, we describe for the first time, to our knowledge, that the collagen VI von Willebrand factor type A like domains exhibit a broad-spectrum antimicrobial activity against Gram-positive and Gram-negative bacteria in human skin infections in vivo. In silico sequence and structural analysis of VWA domains revealed that they contain cationic and amphipathic peptide sequence motifs, which might explain the antimicrobial nature of collagen VI. In vitro and in vivo studies show that these peptides exhibited significant antibacterial activity against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa through membrane disruption. Our findings shed new light on the role of collagen VI derived peptides in innate host defense and provide templates for development of peptide-based antibacterial therapies.

  • Monitoring Arthritis - Biochemical, Metodological and Clinical Aspects

    • Arthritides are musculoskeletal diseases, which predominantly affect the joints e.g. osteoarthritis (OA) and rheumatoid arthritis (RA). Modern treatment strategies include early detection and careful monitoring of the disease course. Close monitoring is also the basis for individualized treatment. Monitoring should be done using methods reflecting different aspects of the disease to get a broad perspective of the disease course. It is of great importance that the methods used for monitoring arthritis are standardized. In paper I, we studied the natural course of knee OA in a population based cohort of middle-aged individuals with chronic knee pain at inclusion, over a 12 year period with a structural method, radiography, and a clinical method. The main finding was that individuals with chronic knee pain were at high risk for developing radiographic knee OA. In paper II, we studied the presence of low-grade systemic inflammation in individuals with chronic knee pain with and without radiographically verified knee OA by measuring high-sensitivity(hs)CRP. We found that low-grade inflammation is a feature of knee OA especially in individuals developing OA in both tibiofemoral (TF) and patellofemoral (PF) compartments. In paper III the diurnal variation of serum COMP was studied in individuals with clinically verified OA and in patients with RA. We found that serum COMP has a half-life of about 7 hours and it has a significant diurnal variation but serum levels were stable during day-time. The objective of study IV was to monitor serum levels of COMP during a randomised controlled trial of physical exercise vs. standardised rest in individuals with symptomatic and radiographic knee OA. Serum COMP levels increased after one hour of physical exercise and decreased after one hour of rest. No increase was seen after a six-week exercise program. In a supplementary study in paper IV we monitored serum COMP with repeated measurements after the exercise session. Serum COMP levels increased immediately after exercise but 30 minutes after the exercise session the levels did not differ significantly from baseline levels. From these studies I conclude that knee pain is often the first sign of knee OA. Elevated levels of hsCRP in serum, particularly in individuals developing combined PF and TFOA, suggest that low-grade inflammation is a feature of early knee OA. COMP has a short half-life in serum and a significant diurnal variation. Serum COMP levels increase after physical exercise but normalize after about 30 minutes of rest. Samples for measuring serum COMP should be obtained at day-time after resting for at least 30 minutes in a sitting position.
