  • Assessment of Antibacterial Properties of Polyvinylamine (PVAm) with Different Charge Densities and Hydrophobic Modifications

    • Hydrophobically modified and unmodified polyvinylamines (PVAm), including a total of five polymers, were tested against both gram-negative (Escherichia coli) and gram-positive (Bacillus subtilis) bacteria for antimicrobial activity. The assessment of PVAm in solution against bacteria is described, and the influence of the charge density and of the hydrophobic modification of the polyelectrolyte is discussed. The antimicrobial activity was found to depend upon the concentration of PVAm and also on the type of bacteria used. The results also indicated that no direct relationship exists between antimicrobial activity and charge density of the different PVAms. It was, however, observed that an alkyl chain length of six or eight alkane units had a substantial effect on the bacteria investigated. The best combined antibacterial activity for the two bacteria tested was achieved for PVAm with a C-6 alkane substituent (PVAm C-6). To evaluate the antimicrobial activity on a solid substrate, PVAm C-6 was further studied after being deposited onto a glass slide and the results show a large reduction in bacterial infection.

  • Diltiazem infusion for renal protection in cardiac surgical patients with preexisting renal dysfunction

    • Objective: To evaluate if the calcium channel blocker diltiazem protects postoperatively renal function in cardiac surgical patients with preexisting mild-to-moderate renal dysfunction. Design: Prospective, randomized, placebo-controlled, double-blind, clinical study. Setting: Cardiothoracic anesthesia department at a university hospital. Participants: Adult patients undergoing elective cardiac surgery using cardiopulmonary bypass, with a preoperatively elevated serum creatinine level (n = 24). Interventions: Randomized infusions of diltiazem (bolus 0.25 mg/kg followed by a continuous infusion of 1.7 pg/kg/min) (DTZ, n = 12) or placebo (C, n = 12) were started 30 minutes before induction of anesthesia and continued for 24 hours. Measurements and Main Results: Median plasma concentrations of diltiazem (DTZ group) were 79 mug/L before cardiopulmonary bypass, 67 mug/L at the end of cardiopulmonary bypass, and 164 mug/L at 24 hours postoperatively. Serum creatinine levels; on postoperative days 1, 3, and 5; and 3 weeks postoperatively were similar between groups. lohexol clearance did not differ between the groups on day 5 but was higher in the DTZ group than in the placebo group 3 weeks after surgery (median, 51 v 40 mL/min/1.73 m(2); p < 0.05). Urinary N-acetyl-β-glucosamidase concentrations were similar between the groups during the study but were increased from baseline on days 2 and 4 and 3 weeks postoperatively. Conclusion: Diltiazem can be safely used in patients who have mild-to-moderate renal dysfunction and undergo cardiac surgery using cardiopulmonary bypass. Within the limits of this study, the data suggest that addition of prophylactic diltiazem may prevent further glomerular damage resulting from cardiopulmonary bypass and may improve glomerular function 3 weeks after cardiac surgery.

    • In this review we discuss the merits and drawbacks with the use of proteomic and peptidomic strategies for identification of proteins and peptides in their multidimensional interactions in complex biological processes. The progress in proteomics and peptidomics during the last years offer us new challenges to study changes in the protein and peptide synthesis. These strategies also offer new tools to follow post-translational modifications and other disturbed chemical processes that may be indicative of pathophysiological alteration(s). Furthermore these techniques can contribute to improvements in the diagnosis and therapy of neurodegenerative diseases, such as Alzheimer's disease, and psychiatric diseases, as depression and post traumatic stress disorders. We also consider different practical aspects of the applications of mass spectrometry in clinical neuroscience, illustrated by example from our laboratories. The new proteomic and peptidomic strategies will further enable the progress for clinical neuroscience research.

    • Growth of bacteria on various surfaces may cause major concern if encountered in the wrong environment.  The ability to impart antimicrobial properties onto surfaces is therefore desirable.  Modification of both glass surfaces and cellulose membranes using weak polyelectrolytes as antimicrobial agent is presented.  The aim was to incorporate a known carrier of antimicrobial activity into a multilayer structure of adsorbed polyelectolytes at different surfaces and evaluate the antibacterial activities.  The approach involved screening for antimicrobial activity in soln. of modified and unmodified polymers.  The polymers were evaluated against gram-neg. E. coli and gram-pos. B. subtilis. In order to elucidate the mode of action of the polymers, the charge was detd.  This is done since earlier studies have shown that both charge and degree of hydrophobicity are of importance in terms of exerted activity.  Results show that the chosen polymers exert activity in the immobilized state.  Different techniques have been used to evaluate the mechanisms.

    • During the 1990s rural Swedish women positioned themselves as women and formed networks aimed at addressing various local and most often gender-related problems that limited their quality of life. The aim of this article is to empirically describe and theoretically discuss the idea of agency in the context of women's social practices of networking and to examine how the participants either reproduce or transform the gendered structures that shape them. The empirical data for this article consist primarily of discussions with focus groups and interviews with network participants. Through interpretations, a pattern of gendered power-relations was illuminated, which both influenced and constrained the participants' activities. We have interpreted the participants' networking agency as acts of protest against everything that limits their living conditions. As women develop strategies through networking, their resistance seems to become increasingly significant for the ongoing transformations of the gender order.
